Maternal levels of xenobiotics that affect fetal development and childhood health.

Children are not only being born into a chemically toxic world, they are exposed to these toxins throughout their gestational development. They are exposed to hundreds of toxic compounds via cord blood and release many of them in the meconium. These toxins include numerous neuro-, immuno-, and endocrine-toxic compounds present during the critical stages of hormonal, immunological, and neurological development. Outcome studies have shown that animal and human offspring who are so exposed can not only be born with birth defects, but suffer from lifelong health and behavior problems. This article

Human fetal ovary development involves the spatiotemporal expression of p450c17 protein.

OBJECTIVE: The purpose of this research was to characterize the spatiotemporal expression of P450c17 in the human fetal ovary. DESIGN: P450c17 protein was visualized in sections of control and anencephalic ovaries using immunohistochemistry. SUBJECTS: Subjects included control (nonanencephalic) and anencephalic human fetal ovaries during the second and third trimesters. RESULTS: In second-trimester control ovaries, P450c17 was highly expressed in primary interstitial cells (PIC) located between the ovigerous cords near the cortical-medullary border where meiosis and primordial follicle form

Brain development during fetal life: influences of the intra-uterine environment.

The intrauterine environment can significantly affect fetal brain development. Here we review our recent findings using animal models that mimic adverse intrauterine conditions which could exist during human pregnancy. We have focused on effects of both acute and chronic hypoxic and inflammatory insults. Relatively brief periods of hypoxemic compromise can have significant effects on the fetal brain causing neuronal loss and cerebral white matter damage. Subtle brain injury can occur, for example to a particular class of neuron, and this can have a significant effect on the function of a speci

Congenital metatarsus adductus in early human fetal development: a histologic study.

Two feet with congenital metatarsus adductus from fetuses at early development (16 and 19 weeks of gestation) were studied by making serial histologic sections in the horizontal plane of the foot. It was observed that the shape of the medial cuneiform was altered and the first cuneometatarsal joint tilted toward the medial and dorsal directions. The first metatarsal appeared normal, whereas the other metatarsals were deformed in slight adduction at the metaphyseal level. Subluxation at the other cuneometatarsal joints and naviculocuneiform joint was not observed, and the navicular showed no si

Extrinsic fetal akinesia and skeletal development: a study in oligohydramnios sequence.

Long-bone morphometry and cephalometry were performed in 13 newborns with oligohydramnios sequence (OS) in order to establish whether or not skeletal changes existed in extrinsic fetal akinesia similar to those observed in the fetal akinesia deformative sequence (FADS) (i.e., hypoplastic long bones and micrognathia). Oligohydramnios sequence was caused by bilateral renal agenesis in five cases and obstructive uropathy in eight cases. Twenty-one stillborns and newborns who had died from conditions other than renal anomalies or congenital malformations were used as controls. Normal longitudinal

Progesterone secretion and fetal development during prolonged starvation in the pig.

Roles of ovarian progesterone secretion and maternal nutrition in fetal development were investigated in a species that normally experiences considerable embryonic and fetal mortality. Pregnancies were maintained in 81% of Yorkshire pigs during prolonged starvation (e.g., 40 days; 0 kcal/day, water only) in either the middle third (days 30-70) or last third (days 70-110) of gestation compared with 100% in full-fed controls (7,028 kcal/day). In spite of severe maternal deprivation, fetal survival rates averaged 65% in starved dams and 63% in controls; mean number of living fetuses was 9.9 in st…

Inapparent maternal ZIKV infection impacts fetal brain development and postnatal behavior.

Zika virus (ZIKV) has emerged as a significant public health concern due to its association with severe neurological outcomes in infants, including microcephaly and congenital Zika syndrome (CZS). However, while the majority of ZIKV infections during pregnancy do not result in CZS, the potential long-term neurological effects of mild or inapparent maternal infections remain poorly understood. In this study, we adapted a model of maternal ZIKV infection in human STAT2 knock-in (hSTAT2) mice to investigate the effects of ZIKV infection during mid-gestation, aiming to mirror typical asymptomatic

Temporal impact of hypothyroidism on porcine fetal growth and skeletal development.

In the fetal pig, circulating thyroid hormone and growth rate increase throughout gestation; however, the functional association between these coincident factors has not been established. We used a nonpathogenic model of induced fetal hypothyroidism, wherein pregnant gilts were treated with methimazole (MMI) for 21 days, starting on gestation days (GD) 34, 45, 55, or 65. The resulting fetal populations were deeply phenotyped, including body and organ weights, cranial and whole body morphometrics, radiography, and dual energy X-ray absorptiometry (DXA) of the fore- and hindlimbs. MMI increased

Follicular development of fetal gonads under the skin of adult mice.

Adult ovarian tissues or biopsies isolated from patients prior to chemotherapy or irradiation can reconstitute ovarian functions when transplanted either in the abdomen or subcutaneously. Subcutaneously transplantation avoids invasive surgery and potential risks associated with internal procedures. We investigated whether functional ovaries could develop subcutaneously from early E12.5 fetal gonads without entering meiosis in mouse model. Unexpectedly, the subcutaneously transplanted fetal gonads failed to undergo folliculogenesis in the recipient mice. The transplanted gonads experienced meio

Metabolomic landscape of fetal organ development during late gestation in mice.

Metabolism is the critical basis for mammalian physiological functions. The systematic metabolic characteristics of major organ development during late gestation to adapt to postnatal environmental changes are still absent. Here, we detected metabolic patterns of the ICR mouse fetal organs, including the heart, stomach, liver, brain, and placenta, from embryonic days (E)15.5 to 19.5 using liquid chromatography-mass spectrometry combined with RNA sequencing and proteomics data. Our metabolic and multi-omics data showed that organs exhibited their unique metabolic characteristics during late ges

Nutrition during pregnancy: Influence on the gut microbiome and fetal development.

Pregnancy is a finely tuned process, with the health and well-being of the developing fetus determined by the metabolic status and dietary intake of the mother. The maternal gut microbiome is remodeled during pregnancy, and this, coupled with the maternal nutrient intake during gestation shapes the production of metabolites that can cross the placenta and affect fetal development. As posited by the Developmental Origins of Health and Disease Hypothesis, such environmental influences can have major effects on the developing organ systems. When occurring at particularly sensitive gestational tim…

Effects of Sevoflurane Exposure on Fetal Brain Development Using Cerebral Organoids.

Sevoflurane is a safe and well-known inhaled anesthetic. Given that sevoflurane can be delivered to developing fetuses through the mother, it is critical to determine whether this agent affects fetal neurodevelopment. Recent research has sought to determine whether sevoflurane affects fetal brain development when the mother is exposed during the second to third trimester of pregnancy, considered to be the crucial period for the development of nervous system. However, even though the first trimester is a critical period for fetal organogenesis and the most susceptible time to teratogen exposure

Associations of maternal dietary patterns during pregnancy and fetal intrauterine development.

Dietary pattern is excellent in reflecting an individual's eating conditions. Longitudinal data on fetal growth can reflect the process of intrauterine growth. We aimed to evaluate the associations between maternal dietary patterns and intrauterine parameters in middle and late pregnancy. The present study was conducted within Jiangsu Birth Cohort (JBC) study. Dietary information was assessed with a food frequency questionnaire (FFQ) in the second and third trimester of gestation. B-ultrasound scans were performed to obtain fetal intrauterine parameters, including head circumference (HC), femu
