We have carried out an analysis of the serological and molecular diversity of a panel of monoclonal anti-DNA autoantibodies and serum autoan … subfamilies. A total of four different VH and at least six V kappa subfamilies are expressed by the hybridomas. An original set of 17 clones of hybridomas-producers of monoclonal antibodies (MCAB) against Fc-fragment of human IgG has been obtained. The possibility of using horseradish peroxidase conjugates of the obtained MCAB as a part of ELISA tests systems of the company "DiaProphMed" for diagnostics of syphilis, rubella, toxoplasmosis and cytomegalovirus infections has been comparatively studied. It has been shown, that the application of peroxidase conjugates of two monoclonal antibodies (156C10 and 153H11) as the part of five diagnostic ELISA test systems provides high specificity and sensitivity.

A rapid and efficient method for preparing monoclonal antibody (MAb) serotypes using Neisseria meningitidis outer membrane were used in BALB/c mouse footpads for the immunization. The popliteal lymph nodes were isolated 19 days later for MAb-producing hybridomas, from which the MAbs against the 37 kDa protein were screened. AIM: To express midkine (MK) protein, prepare its monoclonal antibody (mAb), and detect its expression in tumor cells.
METHODS: The human MK gene fragment was cloned from kidney cancer tissues by RT-PCR and inserted into the prokaryotic expression vector pMS-31b. The MS2-MK fusion protein was expressed in E.coli after temperature induction. The purified fusion protein was used to immunize BALB/c mice to prepare mAbs against MK by routine hybridoma technique. The Ig subclasses of the mAbs were identified by ELISA. The expression of MK in tumor cells was detected with the mAbs.

Monoclonal antibody (mAb) MR6 recognises a 200 kDa glycoprotein, gp200-MR6, which is expressed at high levels on the surface of human thymic cortical epithelium.
