Induction of immune tolerance in mice with a novel mucosal nanoemulsion adjuvant and self-antigen.

AIM: The aim of this study was to investigate the impact of a novel nanoemulsion (NE) adjuvant, a soybean oil emulsion, on autoimmune response. To this end, we used murine thyroglobulin (mTg)-induced experimental autoimmune thyroiditis in mice as a study model. MATERIALS & METHODS: Mice received NE or NE + mTg by nasal delivery. At 1 week after the second nasal delivery of NE with or without mTg, all mice were immunized with mTg and lipopolysaccharides to induce experimental autoimmune thyroiditis. RESULTS: Compared with controls, mTg-NE-treated mice had much more antigens accumulated in the

Immune tolerance induction in patients with IgA anaphylactoid reactions following long-term intravenous IgG treatment.

To date, there is very little information regarding the pathomechanism of IgA anaphylactoid reactions and the management of affected patients. Five adult patients with common variable immunodeficiency (CVID) and a history of anaphylactic reactions due to the administration of immunoglobulin preparations were studied. The activity of anti-IgA was determined by the gel agglutination technique using IgA-coated beads. Antibodies to IgA were detected in the serum of all five patients. Initially, IgA 'depleted' intravenous (i.v.) IgG preparations were infused carefully into the patients until the ac

Kynurenine pathway metabolism in human blood-brain-barrier cells: implications for immune tolerance and neurotoxicity.

The catabolic pathway of l-tryptophan (l-trp), known as the kynurenine pathway (KP), has been implicated in the pathogenesis of a wide range of brain diseases through its ability to lead to immune tolerance and neurotoxicity. As endothelial cells (ECs) and pericytes of the blood-brain-barrier (BBB) are among the first brain-associated cells that a blood-borne pathogen would encounter, we sought to determine their expression of the KP. Using RT-PCR and HPLC/GC-MS, we show that BBB ECs and pericytes constitutively express components of the KP. BBB ECs constitutively synthesized kynurenic acid, a

Enhanced response to enzyme replacement therapy in Pompe disease after the induction of immune tolerance.

Pompe disease, which results from mutations in the gene encoding the glycogen-degrading lysosomal enzyme acid alpha -glucosidase (GAA) (also called "acid maltase"), causes death in early childhood related to glycogen accumulation in striated muscle and an accompanying infantile-onset cardiomyopathy. The efficacy of enzyme replacement therapy (ERT) with recombinant human GAA was demonstrated during clinical trials that prolonged subjects' overall survival, prolonged ventilator-free survival, and also improved cardiomyopathy, which led to broad-label approval by the U.S. Food and Drug Administra

Immune tolerance induction in the treatment of paediatric haemophilia A patients with factor VIII inhibitors.

The development of an inhibitor to transfused factor VIII (FVIII) is a serious treatment-related problem in haemophiliac children. The management of patients with high titre FVIII inhibitors is difficult, and immune tolerance induction (ITI) is the only method available for the eradication of these inhibitors. The results of the ITI regimen used at the Children's Hospital of Michigan Haemophilia Treatment Center are described and discussed. ITI was attempted in 14 children with severe haemophilia A (13 high responders, one low responder), with daily doses of FVIII alone. FVIII dosage was chose

Safety and tolerability of KAN-101, a liver-targeted immune tolerance therapy, in patients with coeliac disease (ACeD): a phase 1 trial.

BACKGROUND: Coeliac disease management is limited to strict adherence to a gluten-free diet with no approved therapies. This first-in-human phase 1 study evaluated the safety and tolerability of KAN-101, a liver-targeting glycosylation signature conjugated to a deaminated gliadin peptide designed to induce immune tolerance to gliadin. METHODS: Adults (aged 18-70 years) with biopsy-confirmed, HLA-DQ2.5 genotype coeliac disease were enrolled from clinical research units and hospitals in the USA. Part A of the trial was an open-label, single ascending dose study of intravenous KAN-101 using sent

Spontaneous organ-specific eye autoimmune disease resulting from two engineered gene mutations compromising immune tolerance.

In order to address the question of how genetic compromises in multiple immune tolerance mechanisms might confer susceptibility to autoimmune disease development, we have bred together a partial loss of function in Aire, which is important for central tolerance of T cells, with a deficiency of the intracellular protein tyrosine kinase Lyn, which is important for function of inhibitory receptors of dendritic cells, B lymphocytes, and myeloid cells. These AireGW/+Lyn-/- mice spontaneously develop severe eye autoimmunity, characterized by retinal destruction and damage to the surrounding uvea, re

Regulatory T cells and peripheral immune tolerance: key mechanisms and therapeutic strategies for cardiovascular diseases.

Cardiovascular diseases (CVDs) remain the leading global cause of disability and mortality, with chronic immune inflammation as their central pathological mechanism. Regulatory T cells (Tregs) and the peripheral immune tolerance they mediate hold a pivotal position and therapeutic potential in CVDs. This review provides a comprehensive overview of Tregs biology, their dysfunction in major CVDs, current therapeutic strategies targeting Tregs, and future directions for achieving immune homeostasis. By moving beyond the traditional focus on numerical deficiency, we highlight the critical role of

Beyond dysbiosis: microbial metabolites as key remodelers of nasal mucosal immune tolerance in chronic rhinosinusitis.

Chronic rhinosinusitis (CRS) is a heterogeneous inflammatory disorder of the nasal and paranasal sinus mucosa affecting approximately 11% of adults worldwide. Although compositional dysbiosis of the sinonasal microbiome has historically dominated etiological discourse, this framework inadequately accounts for the mechanistic complexity of mucosal immune dysregulation in CRS. Emerging evidence positions microbial metabolites, rather than microbial identity per se, as the proximate immunological mediators of commensal microbiota-host crosstalk. This review presents a comprehensive analysis of th

PD-L1 expression in trophoblasts contributes to maternal-fetal immune tolerance in the canine placenta.

To elucidate the mechanisms of maternal-fetal immune tolerance in the canine endotheliochorial placenta, we investigated the localization and functional significance of programmed death-ligand 1 (PD-L1). Immunohistochemical analysis revealed region-specific PD-L1 expression, with negative staining in syncytiotrophoblasts and cytotrophoblasts within the labyrinth zone, and positive staining in monolayered columnar trophoblasts within the junctional zone. As the junctional zone is the primary interface for maternal tissue invasion, this site-specific expression suggests that PD-L1 functions as a

Fluconazole tolerance is associated with altered extracellular vesicle-mediated epithelial immune responses in Candida albicans.

Candida albicans is a major opportunistic fungal pathogen and a leading cause of oropharyngeal candidiasis, particularly in immunocompromised individuals. Extracellular vesicles (EVs) have emerged as important mediators of fungal-host communication; however, whether antifungal tolerance is associated with EV-mediated epithelial immune responses remains poorly understood. Here, we investigated the immunomodulatory effects of EVs released by fluconazole-tolerant and fluconazole-susceptible C. albicans clinical isolates on human oral epithelial cells (FaDu). EVs from tolerant and susceptible isol

Methionine regulates maternal-fetal immune tolerance and endometrial receptivity by enhancing embryonic IL-5 secretion.

Endometrial receptivity and maternal-fetal immune tolerance are two crucial processes for a successful pregnancy. However, the molecular mechanisms of nutrition involved are largely unexplored. Here, we showed that maternal methionine supply significantly improved pregnancy outcomes, which was closely related to interleukin-5 (IL-5) concentration. Mechanistically, methionine induced embryonic IL-5 secretion, which enhanced the conversion of CD4+ T cells to IL-5+ Th2 cells in the uterus, thereby improving maternal-fetal immune tolerance. Meanwhile, methionine-mediated IL-5 secretion activated t

Short-Term Immunosuppression in Rats Induces Prolonged Immune Tolerance Towards a Human Monoclonal Antibody, Erenumab.

Administration of human therapeutic proteins such as monoclonal antibodies (mAb) to animals during preclinical drug development often leads to the development of anti-drug antibodies (ADA). ADA may reduce the systemic exposure of the mAb by enhancing its immune-complex mediated clearance. Thus, ADA may hinder the preclinical pharmacokinetic and toxicology assessments of mAbs. To mitigate this effect, we explored the ability of short-term administration of immunosuppressants to induce prolonged immune tolerance towards a human mAb, erenumab, in rats. In two studies, we investigated dosing regim

Incomplete Immune Tolerance in Pediatric ABO-Incompatible Liver Transplantation: Insights From Donor-Specific Antibody Titers.

BACKGROUND Advancements in transplant immunology and pharmacotherapy have led to improved outcomes in pediatric ABO-incompatible liver transplantation. However, the long-term dynamics of donor-specific versus non-donor-specific blood group antibodies (BGAs) in this population remain unclear. This study aimed to determine whether incomplete immune tolerance to donor-specific BGAs persists after ABO-incompatible liver transplantation. MATERIAL AND METHODS This retrospective study included 29 pediatric patients who underwent ABO-incompatible living donor liver transplantation with extended follow

Single coadministration of erythrocyte-uricase conjugate and immunosuppressant induces durable immune tolerance for gout therapy.

Uricase (UOX) is effective for treating refractory gout but is limited by antidrug antibody (ADA) formation, which leads to loss of efficacy and infusion reactions. Here, we demonstrate that a single coadministration of erythrocyte-conjugated UOX (UOX-Ery) with an immunosuppressant such as cyclophosphamide (CTX) induces robust and durable antigen-specific immune tolerance. This one-time intervention effectively prevented ADA formation in mice and rats, even after repeated challenges with UOX-Ery or free UOX protein, an outcome not achieved by CTX or UOX plus CTX. This tolerogenic effect relied

Transdermal Microneedles Alleviated Rheumatoid Arthritis by Inducing Immune Tolerance via Skin-Resident Antigen Presenting Cells.

Restoring immune tolerance is the ultimate goal for rheumatoid arthritis (RA) treatment. The most reported oral or intravenous injection routes for the immunization of autoantigens cause gastrointestinal side effects, low patient compliance, and unsatisfied immune tolerance induction. Herein, the use of a transdermal microneedle patch is for the first time investigated to codeliver CII peptide autoantigen and rapamycin for reversing immune disorders of RA. The immunized microneedles efficiently recruit antigen-presenting cells particularly Langerhans cells, and induce tolerogenic dendritic cel

Helios-Negative Regulatory T Cells as a Key Factor of Immune Tolerance in Nonallergic Beekeepers.

BACKGROUND AND OBJECTIVES: Although exposure to stings has been identified as the leading risk factor for anaphylaxis due to Hymenoptera venom allergy, professional beekeepers receive hundreds of stings yearly without developing systemic reactions. This study aims to analyze the mechanisms underlying bee venom tolerance in beekeepers. METHODS: A cross-sectional study was conducted. Participants were recruited and classified into 3 groups: allergic patients (APs), who experienced systemic reactions after bee stings, with a positive intradermal test and specific IgE (sIgE) to Apis mellifera ven

[Progress of researches on roles of dendritic cells in immune tolerance caused by Echinococcus infections].

Dendritic cells (DCs), a type of antigen-presenting cells (APC), are recognized as an important regulator of immune response and immune tolerance, and play a critical role in the host innate immunity and adaptive immunity. Previous studies have shown that the long-term parasization of Echinococcus in the host is strongly associated with the host immune tolerance induced by DCs. This review summarizes the research progress of the role of DCs in host immune tolerance caused Echinococcus infection, aiming to provide the theoretical basis and insights into the management and immunotherapy of Echin

Nanoemulsions Target to Ectopic Lymphoids in Inflamed Joints to Restore Immune Tolerance in Rheumatoid Arthritis.

Inducing immune tolerance through repeated administration of self-antigens is a promising strategy for treating rheumatoid arthritis (RA), and current research indicates that coadministration of immunomodulators can further orchestrate the tolerogenic response. However, most of the clinical trials based on tolerance induction have negligible therapeutic effects. Peripheral lymphoid organs play critical roles in immunotherapy. Here, we design an engineered nanoemulsion for targeted codelivery of self-antigens and an immunomodulator to ectopic lymphoid structures (ELSs) in inflamed joints of RA.
