Interferon-epsilon, an estrogen-induced type I interferon, is uniquely exploited by Neisseria gonorrhoeae via effects on sialic acid metabolism.

The female genital mucosa expresses the hormone-dependent type I interferon (IFN), IFN-epsilon (IFN-ε), which protects against chlamydia and herpes infection. Surprisingly, we found that IFN-ε knockout (Ifnε-/-) mice and type I IFN receptor knockout (Ifnar1-/-) mice exhibited enhanced clearance of Neisseria gonorrhoeae (Ng). This result was phenocopied using blocking anti-IFNAR monoclonal antibody (mAb). Ng colonization of the Ifnε-/- urogenital tract was restored by exogenous recombinant IFN-ε or IFN-β. Clearance of Ng in anti-IFNAR-treated mice required the expression of the cathelicidin mCR

Coordination of retrotransposons and type I interferon with distinct interferon pathways in dermatomyositis, systemic lupus erythematosus and autoimmune blistering disease.

Type I interferon (IFN) plays a crucial role in innate and adaptive immunity, and aberrant IFN responses are involved in systemic autoimmune diseases, such as systemic lupus erythematosus (SLE) and dermatomyositis (DM). Type I IFNs can be induced by transcribed retrotransposons. The regulation of retrotransposons and type I IFN and the downstream IFN pathways in SLE, DM, and autoimmune blistering disease (AIBD) were investigated. The gene expression levels of retrotransposons, including LINE-1, type I-III IFNs, and IFN-stimulated genes (ISGs) in peripheral blood cells from patients with DM (n

Type I interferon reaction to viral infection in interferon-competent, immortalized cell lines from the African fruit bat Eidolon helvum.

Bats harbor several highly pathogenic zoonotic viruses including Rabies, Marburg, and henipaviruses, without overt clinical symptoms in the animals. It has been suspected that bats might have evolved particularly effective mechanisms to suppress viral replication. Here, we investigated interferon (IFN) response, -induction, -secretion and -signaling in epithelial-like cells of the relevant and abundant African fruit bat species, Eidolon helvum (E. helvum). Immortalized cell lines were generated; their potential to induce and react on IFN was confirmed, and biological assays were adapted to app

Early production of type I interferon during West Nile virus infection: role for lymphoid tissues in IRF3-independent interferon production.

Infection of cells with flaviviruses in vitro is reduced by pretreatment with small amounts of type I interferon (IFN-alpha/beta). Similarly, pretreatment of animals with IFN and experiments using mice defective in IFN signaling have indicated a role for IFN in controlling flavivirus disease in vivo. These data, along with findings that flavivirus-infected cells block IFN signaling, suggest that flavivirus infection can trigger an IFN response. To investigate IFN gene induction by the very first cells infected during in vivo infection with the flavivirus West Nile virus (WNV), we infected mice

Expression of interferon gamma by a recombinant rabies virus strongly attenuates the pathogenicity of the virus via induction of type I interferon.

UNLABELLED: Previous animal model experiments have shown a correlation between interferon gamma (IFN-γ) expression and both survival from infection with attenuated rabies virus (RABV) and reduction of neurological sequelae. Therefore, we hypothesized that rapid production of murine IFN-γ by the rabies virus itself would induce a more robust antiviral response than would occur naturally in mice. To test this hypothesis, we used reverse engineering to clone the mouse IFN-γ gene into a pathogenic rabies virus backbone, SPBN, to produce the recombinant rabies virus designated SPBNγ. Morbidity and

Role of type I interferons and innate immunity in systemic sclerosis: unbalanced activities on distinct cell types?

PURPOSE OF REVIEW: The role of type I IFNs (IFN-I) in the promotion of autoimmunity has been well established. However, its role in the skin fibrosis of systemic sclerosis (SSc) is less clear. IFN-I can participate to tissue repair, and, here, we will consider the extent to which IFN-I's role in SSc skin fibrosis may reflect in part IFN-I functions during wound healing. RECENT FINDINGS: Studies are beginning to delineate whether IFN-I has a protective or pathogenic role and how IFN-I affects tissue biology. Recent support for a pathogenic role came from a study depleting plasmacytoid dendriti

The interplay of type I and type II interferons in murine autoimmune cholangitis as a basis for sex-biased autoimmunity.

UNLABELLED: We have reported on a murine model of autoimmune cholangitis, generated by altering the AU-rich element (ARE) by deletion of the interferon gamma (IFN-γ) 3' untranslated region (coined ARE-Del-/- ), that has striking similarities to human primary biliary cholangitis (PBC) with female predominance. Previously, we suggested that the sex bias of autoimmune cholangitis was secondary to intense and sustained type I and II IFN signaling. Based on this thesis, and to define the mechanisms that lead to portal inflammation, we specifically addressed the hypothesis that type I IFNs are the d

GPLD1-mediated regulation of the type I interferon-induced signaling pathway and antiviral activity.

Glycosylphosphatidylinositol-specific phospholipase D1 (GPLD1) is traditionally known as a secreted enzyme that sheds glycosylphosphatidylinositol (GPI)-anchored proteins. Emerging evidence suggests its involvement in immune modulation. In this study, we report a non-canonical intracellular function of GPLD1 in potentiating IFN-I-mediated antiviral signaling. Mechanistically, GPLD1 physically interacts with signal transducer and activator of transcription 2 (STAT2) and competitively blocks its interaction with protein-tyrosine phosphatase 1B (PTP1B), a bona fide phosphatase that directly depho

Type I-interferon β induces a strong anti-tumour response in bladder cancer cells.

PURPOSE: For non-muscle invasive bladder cancer (NMIBC), instillation with Bacillus Calmette-Guérin (BCG) is a standard therapy. With a still unclear mechanism, instillation activates the innate immune system, resulting in an immunological effect on the tumour. The aim of this work was to investigate which bladder cancer (BLCA) cells can be activated by interferon (IFN) exposure. METHODS: The BLCA cell lines RT4 and SW780 were stimulated with IFN-α2, -β and -λ1 over 4-72 h. Quantitative PCR (qPCR) was used to determine the expression of IFN receptor subunits (RS) and selected interferon-stimu

Investigating the Role of Type I Interferon Signaling on Muscle Disease Using Mouse Models.

OBJECTIVE: Dysregulated type I interferon (IFN) signaling contributes to autoimmune myositis pathogenesis. We investigated the therapeutic effects of JAK inhibitors in two mouse models. We also examined how type I IFNs affect muscle vasculature. METHODS: Myositis was induced in major histocompatibility complex class I double transgenic ([TRE-H-2Kb (H mice)/MCK-tTA (T mice)] (HT)) female mice at day 21. Mice were randomized into four groups (n = 10-11/group): baricitinib (10 mg/kg), tofacitinib (20 mg/kg), vehicle, and healthy controls. Outcomes included survival, weight, muscle strength, hist

Baicalein activates the type I interferon signaling pathway to inhibit infectious bronchitis virus infection.

Avian infectious bronchitis virus (IBV) is a γ-coronavirus that primarily affects young chicks and laying hens. Its extensive serotype diversity and high mutation rate limit the effectiveness of current vaccines, leading to substantial economic losses in the poultry industry. In this study, we demonstrated that baicalein exerts potent antiviral activity against IBV, with a half-maximal inhibitory concentration (IC50) of 2.34 μM and a selectivity index (SI) of 17.39 in Vero cells. Baicalein inhibited multiple stages of the viral life cycle and reversed IBV-induced immunosuppression by activatin

Human ISG15 deficiency unveils impaired healing of ulcerations via type I interferon-mediated fibrosis.

ISG15 deficiency is a type I interferonopathy characterized by elevated circulating type I interferon (IFN-I), intracranial calcifications, fibrotic skin lesions, and occasionally inflammatory lung disease. However, the mechanisms driving immune-mediated fibrosis remain poorly understood. In this study, we combined molecular biology approaches with spatial transcriptomics in ex vivo patient samples and in vitro models to characterize a novel loss-of-function ISG15 variant and to elucidate disease-associated molecular pathways. Analysis of patient skin biopsies revealed increased IFN-I-dependen

Mitochondrial RNA-Type I Interferon Axis in Sjögren's Disease: Molecular Mechanisms and Translational Implications.

Sjögren's disease (SjD) is a systemic autoimmune disease characterized by exocrine dysfunction, lymphocytic infiltration of the exocrine glands, and prominent activation of the interferon (IFN) pathway. Although IFN signatures are recognized as a central feature of SjD, endogenous triggers that sustain chronic IFN-driven inflammation remain incompletely understood. Mitochondrial RNAs (mtRNAs), particularly double-stranded species, have recently emerged as immunostimulatory molecules capable of linking mitochondrial stress to innate immune activation. In this review, we discuss the biological b

Single-cell analysis identifies a type I interferon-associated neutrophil signature in bacterial meningitis.

Bacterial meningitis (BM) is a central nervous system infection characterized by high mortality and disability rates. Neutrophils, as the first-line innate immune cells in host defense against bacterial invasion, play a crucial role in the pathogenesis of this disease. However, the transcriptomic characteristics of neutrophils in BM remain poorly defined, limiting our understanding of the underlying mechanisms. In this study, we analyzed single-cell RNA sequencing (scRNA-seq) data from the public database GSE221681 to map the transcriptomic landscape of meningeal immune cells in a mouse model

FKBP5 promotes osteogenic differentiation of mesenchymal stem cells through type-I interferon pathway Inhibition.

The decreased osteogenesis of bone marrow mesenchymal stem cells (BMSCs) is an important factor causing bone loss. Nevertheless, its deep molecular mechanism has still not been fully clarified. To elucidate the regulatory mechanisms underlying BMSC osteogenesis, we conducted a bioinformatics screen using public datasets from the Gene Expression Omnibus (GEO) database to identify genes displaying significant expression dynamics during the osteogenic differentiation of BMSCs. We observed a significant upregulation of FK506 Binding Protein 5 (FKBP5) expression during the osteogenic differentiatio

Dried blood spot proteome identifies subclinical interferon signature in neonates with type I interferonopathy.

BACKGROUND: Type I interferonopathy is characterized by aberrant upregulation of type I interferon signaling. The mRNA interferon signature is a useful marker for activation of the interferon pathway and for diagnosis of type I interferonopathy; however, early diagnosis is challenging. OBJECTIVE: This study sought to identify the proteomic interferon signature in dried blood spot (DBS) samples. The aim was to evaluate the usefulness of the interferon signature for neonatal screening and to gain insight into presymptomatic state of neonates with inborn errors of immunity (IEIs). METHODS: DBS

Type I interferon limits interleukin-6 signalling in SLE through shedding interleukin-6 receptors.

OBJECTIVE: To fully understand why C-reactive protein (CRP) is usually only mildly elevated in active systemic lupus erythematosus (SLE), although interleukin-6 (IL-6) is increased, but is high in SLE patients with bacterial infections. METHODS: Sera and peripheral blood mononuclear cells (PBMCs) of SLE patients and healthy individuals were investigated. IL-6 and soluble IL-6 receptor (sIL-6R) were measured by ELISA. Membrane IL-6 receptor-α (CD126), gp130 (CD130) and signal transducer and activator of transcription 3 (STAT3) phosphorylation after IL-6 stimulation were analysed by flow cytome

Transient inhibition of type I interferon enhances CD8+ T cell stemness and vaccine protection.

Developing vaccines that promote CD8+ T cell memory is a challenge for infectious disease and cancer immunotherapy. TCF-1+ stem cell-like memory CD8+ T (TSCM) cells are important determinants of long-lived memory. Yet, the developmental requirements for TSCM cell formation are unclear. Here, we identify the temporal window for type I interferon receptor (IFNAR) blockade to drive TSCM cell generation following viral infection and mRNA-lipid nanoparticle vaccination. We reveal a reversible developmental trajectory where transcriptionally distinct TSCM cells emerged from a transitional precursor

Lipid peroxidation and type I interferon coupling fuels pathogenic macrophage activation causing tuberculosis susceptibility.

A quarter of the human population is infected with Mycobacterium tuberculosis, but less than 10% of those infected develop pulmonary TB. We developed a genetically defined sst1-susceptible mouse model that uniquely reproduces a defining feature of human TB: the development of necrotic lung granulomas and determined that the sst1-susceptible phenotype was driven by the aberrant macrophage activation. This study demonstrates that the aberrant response of the sst1-susceptible macrophages to prolonged stimulation with TNF is primarily driven by conflicting Myc and antioxidant response pathways lea

Type I interferon signaling pathway enhances immune-checkpoint inhibition in KRAS mutant lung tumors.

Lung cancer is the leading cause of cancer mortality worldwide. KRAS oncogenes are responsible for at least a quarter of lung adenocarcinomas, the main subtype of lung cancer. After four decades of intense research, selective inhibitors of KRAS oncoproteins are finally reaching the clinic. Yet, their effect on overall survival is limited due to the rapid appearance of drug resistance, a likely consequence of the high intratumoral heterogeneity characteristic of these tumors. In this study, we have attempted to identify those functional alterations that result from KRAS oncoprotein expression d
