Calcium-Collagen Coupling is Vital for Biomineralization Schedule.

Biomineralization is a chemical reaction that occurs in organisms in which collagen initiates and guides the growth and crystallization of matched apatite minerals. However, there is little known about the demand pattern for calcium salts and collagen needed by biomineralization. In this study, natural bone biomineralization is analyzed, and a novel interplay between calcium concentration and collagen production is observed. Any quantitative change in one of the entities causes a corresponding change in the other. Translocation-associated membrane protein 2 (TRAM2) is identified as an intermed…

Critical role for αvβ6 integrin in enamel biomineralization.

Tooth enamel has the highest degree of biomineralization of all vertebrate hard tissues. During the secretory stage of enamel formation, ameloblasts deposit an extracellular matrix that is in direct contact with the ameloblast plasma membrane. Although it is known that integrins mediate cell-matrix adhesion and regulate cell signaling in most cell types, the receptors that regulate ameloblast adhesion and matrix production are not well characterized. We hypothesized that αvβ6 integrin is expressed in ameloblasts where it regulates biomineralization of enamel. Human and mouse ameloblasts were f…

Biomineralization State of Viruses and Their Biological Potential.

In nature, viruses can realize self-mineralization under metal-ion-abundant conditions. Interestingly, the mineralized state is a transition state of the virus when the host is not available. Mammalian viruses that share the similar chemical properties also stand a chance of transformation into a mineralized state. In this review, we focus on the possibility of mammalian viruses to undergo mineralization under a physiological environment and the development of biomineralized-based virus engineering. We will introduce the effect of biomineralization on the physiochemical or biological propertie…

The ABCC6 Transporter: A New Player in Biomineralization.

Pseudoxanthoma elasticum (PXE) is an inherited metabolic disease with autosomal recessive inheritance caused by mutations in the ABCC6 gene. Since the first description of the disease in 1896, alleging a disease involving the elastic fibers, the concept evolved with the further discoveries of the pivotal role of ectopic mineralization that is preponderant in the elastin-rich tissues of the skin, eyes and blood vessel walls. After discovery of the causative gene of the disease in 2000, the function of the ABCC6 protein remains elusive. More than 300 mutations have been now reported and the conc

Precipitation in liposomes as a model for intracellular biomineralization.

Liposomes present a versatile platform to model intracellular, biologically controlled mineralization. Perhaps, most importantly, precipitation in the confinement of liposomes excludes heterogeneous nucleators that facilitate formation of the thermodynamically most stable crystalline phase in bulk. This provides access to metastable amorphous precursors even in the absence of other additives that interact strongly with the mineral and is fundamental to the capability of cells to prevent spurious nucleation and to select a specific polymorph. Herein, we summarize methods to prepare liposomes fr

[Interface science in formation of biominerals and biomolecules].

The prototype for the minerals in bone and teeth is usually considered to be the basic calcium phosphate hydroxyapatite (HA), containing impurities, such as carbonate and structural defects. It has been proposed that the biological apatite is formed via precursor phases, such as octacalcium phosphate (OCP). The initial precursors maturate with aging and may alter its surface property depending on the proceeding of the hydrolysis into HA regarding adsorption affinity for various biomolecules existing around the forming biominerals. The maturation of the biominerals could be modulating the proce

Nutritional Modulation of the Immuno-Biomineral Interface: From Vascular Calcification to a Hypothesis-Generating Framework for Tumor-Associated Biomineralization.

Pathological biomineralization is increasingly recognized as a regulated biological process involving interactions among mineral metabolism, inflammation, cellular stress, extracellular vesicles (EVs), extracellular matrix (ECM) components, and mineral precursors. These processes are particularly well characterized in vascular calcification, where immune, metabolic, vesicular, and matrix-associated pathways contribute to ectopic mineral deposition. Tumor-associated mineralization is also documented in several malignancies, but its cellular origins, mineral composition, and biological significa…

Terebratulide brachiopod shell biomineralization by mantle epithelial cells.

To understand mineral transport pathways for shell secretion and to assess differences in cellular activity during mineralization, we imaged with TEM and FE-SEM ultrastructural characteristics of outer mantle epithelium (OME) cells. Imaging was carried out on Magellania venosa shells embedded/etched, chemically fixed/decalcified and high-pressure frozen/freeze-substituted samples from the commissure, central shell portions and from puncta. Imaging results are complemented with morphometric evaluations of volume fractions of membrane-bound organelles. At the commissure the OME consists of sever

Secreted kinase phosphorylates extracellular proteins that regulate biomineralization.

Protein phosphorylation is a fundamental mechanism regulating nearly every aspect of cellular life. Several secreted proteins are phosphorylated, but the kinases responsible are unknown. We identified a family of atypical protein kinases that localize within the Golgi apparatus and are secreted. Fam20C appears to be the Golgi casein kinase that phosphorylates secretory pathway proteins within S-x-E motifs. Fam20C phosphorylates the caseins and several secreted proteins implicated in biomineralization, including the small integrin-binding ligand, N-linked glycoproteins (SIBLINGs). Consequently,…

Regulation of biomineralization by proteoglycans: From mechanisms to application.

Proteoglycans consist of core proteins and one or more covalently-linked glycosaminoglycan chains. They are structurally complex and heterogeneous. Proteoglycans bind to cell surface receptors, cytokines, growth factors and have strong affinity for collagen fibrils. Together with their complex spatial structures and different charge densities, proteoglycans are directly or indirectly involved in biomineralization. The present review focused on the potential mechanisms of proteoglycans-mediated biomineralization. Topics covered include the ability of proteoglycans to influence the proliferation…

A lipocalin mediates unidirectional heme biomineralization in malaria parasites.

During blood-stage development, malaria parasites are challenged with the detoxification of enormous amounts of heme released during the proteolytic catabolism of erythrocytic hemoglobin. They tackle this problem by sequestering heme into bioinert crystals known as hemozoin. The mechanisms underlying this biomineralization process remain enigmatic. Here, we demonstrate that both rodent and human malaria parasite species secrete and internalize a lipocalin-like protein, PV5, to control heme crystallization. Transcriptional deregulation of PV5 in the rodent parasite Plasmodium berghei results in…

Vibrational spectral fingerprinting for chemical recognition of biominerals.

Pathologies associated with calcified tissue, such as osteoporosis, demand in vivo and/or in situ spectroscopic analysis to assess the role of chemical substitutions in the inorganic component. High energy X-ray or NMR spectroscopies are impractical or damaging in biomedical conditions. Low energy spectroscopies, such as IR and Raman techniques, are often the best alternative. In apatite biominerals, the vibrational signatures of the phosphate group are generally used as fingerprint of the materials although they provide only limited information. Here, we have used first principles calculation

Biomineralization of Enamel and Dentin Mediated by Matrix Proteins.

Biomineralization of enamel, dentin, and bone involves the deposition of apatite mineral crystals within an organic matrix. Bone and teeth are classic examples of biomaterials with unique biomechanical properties that are crucial to their function. The collagen-based apatite mineralization and the important function of noncollagenous proteins are similar in dentin and bone; however, enamel is formed in a unique amelogenin-containing protein matrix. While the structure and organic composition of enamel are different from those of dentin and bone, the principal molecular mechanisms of protein-pr…

In vivo biocompatibility and biomineralization of calcium silicate cements.

A new mineral trioxide aggregate (MTA) material has been developed with a modified composition that requires investigations to support its clinical use. This study evaluated the biocompatibility and biomineralization of this new MTA material and compared it with that of two other MTA cements over time. Tubes containing materials (or empty tubes as controls) were inserted into the subcutaneous tissues of 40 rats. On days 7, 15, 30, 60, and 90, the tubes were removed with the surrounding tissues, which were either stained with haematoxylin and eosin or von Kossa for further analyses or unstained…

The effect of alendronate on biomineralization at the bone/implant interface.

A recent approach to improve the osseointegration of implants is to utilize local drug administration. The presence of an osteoporosis drug may influence both bone quantity and quality at the bone/implant interface. Despite this, the performance of bone-anchoring implants is traditionally evaluated only by quantitative measurements. In the present study, the osteoporosis drug alendronate (ALN) was administrated from mesoporous titania thin films that were coated onto titanium implants. The effect that the drug had on biomineralization was explored both in vitro using simulated body fluid (SBF)…

The magnetosome model: insights into the mechanisms of bacterial biomineralization.

Though the most ready example of biomineralization is the calcium phosphate of vertebrate bones and teeth, many bacteria are capable of creating biominerals inside their cells. Because of the diversity of these organisms and the minerals they produce, their study may reveal aspects of the fundamental mechanisms of biomineralization in more complex organisms. The best-studied case of intracellular biomineralization in bacteria is the magnetosome, an organelle produced by a diverse group of aquatic bacteria that contains single-domain crystals of the iron oxide magnetite (Fe3O4) or the iron sulf…

Rock-making bacteria: manganese oxidation and biomineralization in pseudomonads.

The oxidation of manganese from Mn(II) to Mn(III) or Mn(IV) is a common trait in the Pseudomonas genus. Many pseudomonads oxidize Mn enzymatically using one or more multicopper oxidases (MnxG and McoA) and in some cases a heme peroxidase (MopA). The oxidation of Mn leads to the extracellular precipitation of reactive Mn oxides in association with extracellular polymeric substances (EPS). Here, we review the current state of knowledge on Mn oxidation and biomineralization in pseudomonads. While most of the genetic, biochemical, and physiological insights we present derive from studies on Pseudo…
