Modern research criteria and the genetics of schizophrenia.

The authors assessed the relevance of narrowly defined diagnostic criteria to genetic research in schizophrenia in the nuclear families of 84 chronic schizophrenic probands compared with families of 90 normal control probands. The morbidity risk for narrowly defined schizophrenia in first-degree relatives of patients with the narrow diagnosis was significantly higher than the control rate (3.8% versus 0.3%). The rate of chronic schizophrenia in the relatives of all schizophrenic patients was also significantly higher than the control rate (7.1% versus 0.6%), as was the rate of "spectrum" disor…

The advances of genetics research on Hirschsprung's disease.

Hirschsprung's disease (HSCR) is a rare and complex congenital disorder characterized by the absence of the enteric neurons in lower digestive tract with an incidence of 1/5 000. Affected infant usually suffer from severe constipation with megacolon and distended abdomen, and face long-term complications even after surgery. In the last 2 decades, great efforts and progresses have been made in understanding the genetics and molecular biological mechanisms that underlie HSCR. However, only a small fraction of the genetic risk can be explained by the identified mutations in the previously establi

Research progress in the genetics of hyperuricaemia and gout.

Gout is one of the most common inflammatory arthritis caused by hyperuricaemia, which is affected by both genetic factors and environmental factors. Early researches show that a few of rare monogenic mutations, such as PRPS1 and HPRT1 mutations, lead to abnormal purine anabolism and then cause hyperuricaemia and gout. In recent years, genome-wide association studies (GWAS) have identified dozens of susceptibility loci and/or candidate genes associated with hyperuricemia and gout. Loss-of-function mutations in SLC2A9, SLC22A11, and SLC22A12 cause hereditary hypouricaemia, while their overexpres

[Research progress of genetic study on Filippi syndrome].

Filippi syndrome is a rare autosomal recessive genetic disorder characterized primarily by syndactyly, microcephaly, distinctive facial features, and intellectual disability. It may also involve multiple systemic manifestations including growth retardation, language disorders, visual impairment, cryptorchidism, abnormal dental and skeletal development, and so forth. The pathogenic gene associated with Filippi syndrome has been identified as CKAP2L, which encodes a mitotic spindle protein localized in the spindle, centrosome, and neural progenitor cells. Deficiency of CKAP2L may disrupt the div

Genetic eye research in Tasmania: a historical overview.

Although considerable recent work on hereditary eye diseases in Tasmanian families has been published, much of this depended on a century of meticulous pedigree collection by earlier clinical researchers. This article reviews some of the historical papers and the importance they have played in gene discovery and understanding of ophthalmic genetics. Tasmanian families have contributed to the identification of genes for X-linked megalocornea, Leber's hereditary optic neuropathy, retinitis pigmentosa, congenital cataract, ptosis, keratoconus, glaucoma and myopia. The true value of the Tasmanian

Current status of genetic research in Rett syndrome.

Rett syndrome is a progressive neurological development syndrome. Evidences for a genetic transmission, but no conclusive data which entirely support the X chromosome involvement, exist. The chromosomal region 11p14-pte has been investigated with polymorphic markers and genes in the region have been sequenced. No sequence divergencies were detected.

[Advances in molecular genetic research on Myelodysplastic syndrome].

Myelodysplastic syndrome (MDS) is a chronic hematologic disorder characterized by ineffective hematopoiesis, dysplasia of one or more cell lines with or without definite genetic changes. Its diagnosis requires a comprehensive analysis combining morphology, immunology, cytogenetics, and molecular biology findings. In recent years, the development of second-generation sequencing (NGS) has provided great assistance in exploring the molecular pathogenesis of hematological malignancies and guidance for clinical practice. Mutations of a series of gene involved in RNA splicing, DNA methylation, trans

[Progress in genetic research on primary pulmonary hypertension].

Primary pulmonary hypertension (PPH) consisting of hereditary pulmonary arterial hypertension and idiopathic pulmonary arterial hypertension is an obstructive pulmonary hypertension caused by primary pulmonary artery hyperplasia. Both environmental and genetic factors are involved in the pathogenesis of PPH. Genes associated with TGF-β signaling pathway including BMPR2, ALK1, ENG, SMAD8 and other genes including NFU1, CAV1, KCNK3 and TopBPl have been associated with PPH. In this review, the function and mechanism of the above genes in the pathogenesis PPH are summarized.

[Clinical implications of molecular genetic research in otorhinolaryngology].

Molecular-genetic research in Otolaryngology has seen a rapid advancement during the last ten years, especially in the fields of otology and head and neck tumors. The results of this basic research have now started to be implemented in the clinic. In otology the understanding of auditive function has dramatically improved. The syndromic and non-syndromic forms of hereditary hearing impairment can be subdivided into their underlying genetic defects, as more and more genes are identified. Diagnostic of syndromic hearing loss has been improved and can be done earlier. But the molecular-genetic an…

Research Progress of Coronavirus Reverse Genetics Technology.

In recent years, coronavirus, a kind of virus with a wide host range and high variability, has a large and complex genome. Research on the reverse genetics of coronaviruses has always been a hot spot. Coronavirus cannot only infect mammals, but also infect humans, showing its wide host adaptability. The mutation ability of the coronavirus is extremely strong. Recently, the rapid mutation rate of SARS-CoV-2 has made the development of vaccines and therapeutic methods face great challenges. At present, reverse genetics technology is a molecular biology tool. This process primarily involves cloni

Molecular genetics and research progress of uterine leiomyosarcoma.

Uterine leiomyosarcoma (uLMS) is a type of malignant soft-tissue tumor, which is developed from myometrium in the female reproductive system. This disease is difficult to be distinguished from benign uterine leiomyoma in the early stages, but it progresses aggressively and relentlessly. Hence, uLMS has a dismal prognosis and high rates of both misdiagnosis and missed diagnosis. Unfortunately, current studies of uLMS pathogenesis and disease biology are inadequate. uLMS disease models are also very limited, hindering the development of effective therapeutics. In this review, we focus on the pat

Advances in Research on Genetic Relationships of Waterfowl Parvoviruses.

Derzsy's disease and Muscovy duck parvovirus disease have become common diseases in waterfowl culture in the world and their potential to cause harm has risen. The causative agents are goose parvovirus (GPV) and Muscovy duck parvovirus (MDPV), which can provoke similar clinical symptoms and high mortality and morbidity rates. In recent years, duck short beak and dwarfism syndrome has been prevalent in the Cherry Valley duck population in eastern China. It is characterised by the physical signs for which it is named. Although the mortality rate is low, it causes stunting and weight loss, which

Rigor and reproducibility in genetic research on eating disorders.

OBJECTIVE: We explored both within-method and between-method rigor and reproducibility in the field of eating disorders genetics. METHOD: We present critical evaluation and commentary on component methods of genetic research (family studies, twin studies, molecular genetic studies) and discuss both successful and unsuccessful efforts in the field. RESULTS: Eating disorders genetics has had a number of robust results that converge across component methodologies. Familial aggregation of eating disorders, twin-based heritability estimates of eating disorders, and genome-wide association studies…
