Hydroxylamines inhibit tyrosine oxidation and nitration: The role of their respective nitroxide radicals.

In vivo, nitroxide antioxidants distribute within minutes throughout all tissues, but are reduced to their respective hydroxylamines due to the cellular reducing environment, which apparently limits their application. To distinguish their antioxidative activity from that of their respective nitroxides, the kinetics and mechanism of their inhibitory effect on the enzymatic oxidation and nitration of tyrosine have been studied. The inhibitory effect of the hydroxylamines on the oxidation and nitration of tyrosine induced by HRP/H2O2 and HRP/H2O2/nitrite was investigated by following the kinetics

Gamma Radiolysis of Hydroxylamine in Aqueous Nitric Acid Solutions.

The radiolytic stability of hydroxylamine (NH3OH+) in concentrated nitric acid (HNO3) is directly relevant to plutonium finishing in used nuclear fuel reprocessing, where NH3OH+ serves as both a plutonium reductant and nitrous acid (HNO2) scavenger under high-radiation conditions. In this study, aqueous 1 M HNO3 solutions containing NH3OH+ were gamma irradiated to accumulated doses of up to 100 kGy. The radiolytic loss of NH3OH+ was found to be independent of its initial concentration, pointing to its reaction with HNO2 and hydroxyl (•OH) and nitrate (NO3•) radicals as the primary degradation

Recent Advances in Electrocatalytic Reduction of Nitrogen Oxides to Hydroxylamine.

Hydroxylamine (NH2OH) is a crucial intermediate in the global chemical industry, but traditional production methods heavily rely on fossil fuels and demanding reagents, leading to severe environmental pollution and energy consumption. Electrochemical reduction of reactive nitrogen oxides (NOx) offers a sustainable alternative, but its selectivity faces significant mechanistic challenges because the thermodynamically unstable NH2OH is easily over-reduced to ammonia (NH3). This review systematically investigated the current development for the electrochemical synthesis of NH2OH. First, the react

Calorimetric study on the decomposition of hydroxylamine in the presence of transition metals.

Hydroxylamine (HA), hydroxylamine chloride (HAC1), and hydroxylamine nitrate (HAN) were each mixed with aqueous solutions of Cr3+, Cr6+, Mn7+, Co2+, Co3+, and Cu2+, and their heat flow profiles were monitored by a small-scaled reaction calorimeter, SuperCRC. These mixing tests demonstrated that HA was less reactive than HACl and HAN with Mn7+ and Cr6+. Their UV-vis spectra confirmed that the substrates reacted when Mn7+ and Cr6+ were reduced. HA was more reactive with Cu2+ than HAC1 and HAN and exhibited the highest reactivity among the three substrates with regard to metals in the intermediat

Developmental toxicity of hydroxylamine: an example of a maternally mediated effect.

Hydroxylamine (HA) is an important reducing agent that is used in several industries. HA is a moderate irritant and a powerful inducer of methemoglobinemia. HA has been shown to react readily with DNA in vitro. Several chemical derivatives of HA are potent developmental toxicants, whereas HA has been reported to cause no developmentally toxic effects. Since HA and its developmentally toxic derivatives share the presence of a terminal hydroxylamine functional group (-NHOH), and since that functional group has been proposed to be the biologically active portion of the molecule, it was deemed app

Hypotensive effects of hydroxylamine in intact anesthetized dogs and cats.

When given IV in bolus doses to intact anesthetized dogs or cats, hydroxylamine hydrochloride produced transient but precipitous falls in the mean arterial blood pressure in a dose-related manner, as well as a significant methemoglobinemia. The half-time for recovery of the mean arterial pressure was also dose-related. These effects were very similar to those elicited by comparable doses of sodium nitrite, except that the half-recovery time for return to normal blood pressure was somewhat longer with nitrite. Although hydroxylamine has long been known to relax vascular smooth muscle in vitro,

Formation of Hydroxylamine in Low-Temperature Interstellar Model Ices.

We irradiated binary ice mixtures of ammonia (NH3) and oxygen (O2) ices at astrophysically relevant temperatures of 5.5 K with energetic electrons to mimic the energy transfer process that occurs in the track of galactic cosmic rays. By monitoring the newly formed molecules online and in situ utilizing Fourier transform infrared spectroscopy complemented by temperature-programmed desorption studies with single-photon photoionization reflectron time-of-flight mass spectrometry, the synthesis of hydroxylamine (NH2OH), water (H2O), hydrogen peroxide (H2O2), nitrosyl hydride (HNO), and a series of

Novel convenient synthesis of biologically active esters of hydroxylamine.

Alkylation of ethyl N-hydroxyacetimidate with readily available methanesulfonates of functionally substituted alcohols and subsequent deprotection of aminooxy group is a novel and convenient method to prepare functionally substituted esters of hydroxylamine with high overall yield. This approach is a good alternative to well-known reaction of N-hydroxyphthalimide with alcohols under the Mitsunobu conditions. The properties of ethoxyethylidene protection of aminooxy group on the contrary to that of N-alkoxyphthalimide group allow to perform a wide spectra of the transformations in the radical o

Supramolecular stabilization of hydroxylamine TEMPOH by complexation with an amphiphilic calixarene.

In an ethanol/water mixture, the nitroxyl radical TEMPO abstracts a hydrogen atom from a phenolic OH group of the amphiphilic para-hexanoylcalix[4]arene, and the hydroxylamine TEMPOH formed yields a stable inclusion complex with another molecule of the calixarene.

Metabolism of dapsone to its hydroxylamine by CYP2E1 in vitro and in vivo.

Dapsone toxicity is putatively initiated by formation of a hydroxylamine metabolite by cytochromes P450. In human liver microsomes, the kinetics of P450-catalyzed N-oxidation of dapsone were biphasic, with the Michaelis-Menten constants of 0.14 +/- 0.05 and 0.004 +/- 0.003 mmol/L and the respective maximum velocities of 1.3 +/- 0.1 and 0.13 +/- 0.04 nmol/mg protein/min (mean +/- SEM). Troleandomycin (40 mumol/L) inhibited hydroxylamine formation at 100 mumol/L dapsone by 50%; diethyldithiocarbamate (150 mumol/L) and tolbutamide (400 mumol/L) inhibited at 5 mumol/L dapsone by 50% and 20%, respe

Metabolism of procainamide to a hydroxylamine by human neutrophils and mononuclear leukocytes.

The chronic use of procainamide is associated with a high incidence of drug-induced lupus and also agranulocytosis. We have previously demonstrated that procainamide is metabolized in the liver to reactive hydroxylamine (PAHA) and nitroso (nitroso-PA) metabolites which covalently bind to protein and are toxic to lymphocytes. We proposed that these metabolites were responsible for the toxicities of procainamide. However, PAHA and nitroso-PA do not appear to escape the liver in significant concentrations. In this paper we describe the metabolism of procainamide to a reactive hydroxylamine by neu

Metabolism of dapsone to a hydroxylamine by human neutrophils and mononuclear cells.

Dapsone is an effective anti-inflammatory agent in conditions in which inflammation is mediated by neutrophils. Dapsone also has been associated with agranulocytosis. We found that neutrophils, which had been activated by a phorbol ester or opsonized zymosan, oxidized dapsone to its nitroderivative. It appears as if this is due to oxidation of dapsone by myeloperoxidase to the hydroxylamine, followed by nonenzymatic oxidation of the hydroxylamine to the nitroderivative. The hydroxylamine can be isolated if ascorbic acid is added to the incubations. Monocytes also contain myeloperoxidase and ac

Role of dapsone hydroxylamine in dapsone-induced hemolytic anemia.

The hemolytic anemia which frequently accompanies treatment of individuals with dapsone and other arylamine drugs is believed to be caused not by the parent drugs per se, but rather by metabolites which are formed during the clearance of the drugs in vivo. To determine whether the N-hydroxyarylamine metabolites of dapsone could be responsible for dapsone-induced hemolysis, dapsone, dapsone hydroxylamine (DDS-NOH) and monoacetyldapsone hydroxylamine were administered to rats which had previously received 51Cr-labeled red blood cells. All three compounds caused an increase in the rate of disappe

Hydroxylamine effects on cryptogenic neoplasm development in C3H mice.

The effect of administration of hydroxylamine (HA) to male and female mice was studied because of reports suggesting an anticarcinogenic effect and an enhancement of lifespan. In this study, two C3H sublines were used: the C3H/HeN which carries a germinal provirus of the mouse mammary tumor virus and the C3H/HeJ(+) which also carries the milk-transmitted exogenous virus. Lifetime administration of 10 mM HA in the drinking water resulted in a decrease in mammary neoplasm incidence in female C3H/HeN mice, but not in female C3H/HeJ(+) mice. Ovarian neoplasms and cysts were common in all groups, i

Catalytic Asymmetric Umpolung (n+1) Cyclization of Bifunctional Hydroxylamines: Entry to Chiral Spirooxindole-Fused Azacycles.

Herein we report a general catalytic asymmetric umpolung (n+1) (n = 4, 5) cyclization using bifunctional hydroxylamines as bis-electrophiles, affording five- and six-membered azaspirooxindoles in good yields with excellent enantioselectivities (up to 98% ee) under mild phase-transfer catalytic conditions. Mechanistic studies reveal cooperative halogen-bonding, together with ion-pairing interactions, between the catalyst and substrate as key to reactivity and stereocontrol. Such discovery will inspire the design of new bifunctional phase-transfer catalysts with halogen-bonding donors in the fut

Differential protection by nitroxides and hydroxylamines to radiation-induced and metal ion-catalyzed oxidative damage.

Modulation of radiation- and metal ion-catalyzed oxidative-induced damage using plasmid DNA, genomic DNA, and cell survival, by three nitroxides and their corresponding hydroxylamines, were examined. The antioxidant property of each compound was independently determined by reacting supercoiled DNA with copper II/1,10-phenanthroline complex fueled by the products of hypoxanthine/xanthine oxidase (HX/XO) and noting the protective effect as assessed by agarose gel electrophoresis. The nitroxides and their corresponding hydroxylamines protected approximately to the same degree (33-47% relaxed form

Both hydroxylamine and nitroxide protect cardiomyocytes from oxidative stress.

The unique anti-oxidative activity of nitroxide radicals protecting against reactive oxygen-derived species (ROS) has been recently demonstrated in several model systems. The present study focuses on the activity of nitroxide and of its reduced form in cultured rat ventricular cardiomyocytes exposed to O2.- and H2O2 generated by hypoxanthine (HX) and xanthine oxidase (XO). To evaluate cell injury, spontaneous beating, leakage of lactate dehydrogenase (LDH), and depletion of cellular ATP were determined. The protective effect of 4-OH-2,2,6,6-tetramethyl-piperidine-N-oxyl (TPL) was compared with

Cyanide prevents the inhibition of platelet aggregation by nitroprusside, hydroxylamine and azide.

Sodium cyanide (CN-) in concentrations of 10 uM or more prevented the inhibition of epinephrine (2.5 uM) and of ADP (4.0 uM) induced primary and secondary aggregation brought about by 10 uM sodium nitroprusside (SNP). Cyanide alone in the same concentration had no effect on platelet aggregation induced by epinephrine or ADP. Even when the addition of CN- was delayed for as long as 9 min after epinephrine and SNP, it immediately reversed the SNP block and initiated a bimodal wave of aggregation. The effect of CN- on SNP inhibition of platelet aggregation appears to be competitive and reversible
