When Phased without Water: Biophysics of Cellular Desiccation, from Biomolecules to Condensates.

The molecular machinery that enables life has evolved in water, yet many of the organisms around us are able to survive even extreme desiccation. Especially remarkable are single-cell and sedentary organisms that rely on specialized biomolecular machinery to survive in environments that are routinely subjected to a near-complete lack of water. In this review, we zoom in on the molecular level of what is happening in the cellular environment under water stress. We cover the various mechanisms by which biochemical components of the cell can dysfunction in dehydrated cells and detail the differen

Phenotypic Plasticity of Invasive Edge Glioma Stem-like Cells in Response to Ionizing Radiation.

Unresectable glioblastoma (GBM) cells in the invading tumor edge can act as seeds for recurrence. The molecular and phenotypic properties of these cells remain elusive. Here, we report that the invading edge and tumor core have two distinct types of glioma stem-like cells (GSCs) that resemble proneural (PN) and mesenchymal (MES) subtypes, respectively. Upon exposure to ionizing radiation (IR), GSCs, initially enriched for a CD133+ PN signature, transition to a CD109+ MES subtype in a C/EBP-β-dependent manner. Our gene expression analysis of paired cohorts of patients with primary and recurrent

Root system response in Argania spinosa plants under drought stress and recovery.

The argane tree is a remarkable essence by its botanical interest and its socioeconomic value. It is endemic species in the southwest of Morocco, where prolonged drought stress may occur. Although its tolerance has been commonly attributed to various mechanisms at the whole plant, the root system has a main role in the whole process of adaptation. We studied in argane tree plants the change in hydraulic conductivity, electrolyte leakage in root as well as root growth under drought stress and recovery. Our findings showed that the root hydraulic conductivity (Lpr) value significantly decreased

Endocrine response to realimentation in young northern elephant seals (Mirounga angustirostris): Indications for development of fasting adaptation.

Most organisms undergo changes in their environment, both predictably and unpredictably, which require them to alter priorities in nutrient allocation with regards to food availability. Species that more predictably encounter extended periods of limited food resources or intake while mitigating the negative effects of starvation are considered to be fasting adapted. Northern elephant seals (NES) are one such species and routinely undergo extended periods of fasting for breeding, molting, as well as a post-weaning fast at 6-8weeks of age. However, during unusual times of nutritional deprivation

The hearts of competitive athletes: an up-to-date overview of exercise-induced cardiac adaptations.

Intense and regular physical exercise is responsible for various cardiac changes (electrical, structural and functional) that represent physiological adaptation to exercise training. This remodeling, commonly referred to as 'athlete's heart', can overlap with several pathological entities, in which sudden cardiac death may be the first clinical presentation. Although pre-competitive screening can identify athletes with life-threatening cardiovascular abnormalities, there are no widely used standardized pre-participation programs and those currently implemented are controversial. Data from pers

A variation on chloroplast development: the bizonoplast and photosynthetic efficiency in the deep-shade plant Selaginella erythropus.

UNLABELLED: • PREMISE OF THE STUDY: Chloroplast development and structure are highly conserved in vascular plants, but the bizonoplast of Selaginella is a notable exception. In the shade plant S. erythropus, each dorsal epidermal cell contains one bizonoplast, while other cells have normal chloroplasts. Our quest was to (1) determine the origin of bizonoplasts, (2) explore developmental plasticity, and (3) correlate developmental changes with photosynthetic activity to provide insights unavailable in other green plants with more constrained development.• METHODS: Bizonoplast development was

Environmental discontinuity hypothesis: Buffer dysfunctions as a source of human disease.

Adaptive physiologic buffers enable organisms to respond to environmental variation with appropriate plasticity. Modern humans have substantially remodeled their environment such that many interactions with the environment have become relatively discontinuous functions compared to the past. Examples include sunlight, temperature, and altitude. We propose that environmental discontinuity represents a Darwinian maladaptation and may promote disease by inducing buffer dysfunctions. Skin pigmentation is an adaptive, dynamic buffer that normalizes sunlight exposure to balance the potential harm of

Pulsatile urea excretion in the gulf toadfish: mechanisms and controls.

Opsanus beta expresses a full complement of ornithine-urea cycle (OUC) enzymes and is facultatively ureotelic, reducing ammonia-N excretion and maintaining urea-N excretion under conditions of crowding/confinement. The switch to ureotelism is keyed by a modest rise in cortisol associated with a substantial increase in cytosolic glutamine synthetase for trapping of ammonia-N and an upregulation of the capacity of the mitochondrial OUC to use glutamine-N. The entire day's urea-N production is excreted in 1 or 2 short-lasting pulses, which occur exclusively through the gills. The pulse event is n

[Adaptation of Leptospira interrograns (sensu stricto) to fresh water].

The contact with polluted waters is one of the main risk factors to catch leptospirosis. A study is presented about the adaptation of Leptospira interrograns to nutrient-lacking water media. For this end, leptospires were incubated in distilled water and tampon saline solution for an undetermined period of time. Leptospires kept viable in water for 98 days whereas the incubated ones in tampon saline solution survived 3 weeks only. Protein cellular and external membrane components were analyzed with electrophoresis in acrylamide gel (SDS-PAGE). When OM protein profiles of leptospires kept in wa

Ecophysiological and phylogenetic studies of Nevskia ramosa in pure culture.

During the last 100 years, the neuston bacterium Nevskia ramosa has been described several times. This bacterium forms conspicuous rosette-like microcolonies at the air-water interface. In this study, pure cultures of Nevskia ramosa were obtained for the first time, from a bog lake (strain Soe1, DSMZ 11499T) and a freshwater ditch (strain OL1, DSMZ 11500). The isolates showed special adaptations to life in the epineuston. They formed hydrophobic surface films with a dull appearance. N. ramosa is sensitive to UV radiation but revealed a very effective photorepair mechanism. Exposure to light at

Biological Insights into Intestinal Adaptation from Preclinical Models of Short Bowel Syndrome.

Short bowel syndrome is a rare and clinically heterogeneous condition resulting from extensive intestinal resections or functional impairment, leading to malabsorption, fluid and electrolyte losses, and potential progression to intestinal failure requiring long-term parenteral nutrition. The long-term outcome of SBS is largely determined by intestinal adaptation, a progressive physiological response involving epithelial remodeling, intestinal stem cell expansion, lineage-specific proliferation, vascular remodeling, and the activity of trophic mediators-most notably glucagon-like peptide-2 (GLP

The hierarchical stratum response system of organism to microgravity during spaceflight.

With the advancement of manned spaceflight technology, significant progress has been made in the fields of aerospace medicine and space biology to address the needs of astronauts in terms of health, safety, and mission performance. The physiological and pathological changes during microgravity exposure, including perception, response, and adaptation mechanisms, are gradually being unraveled. This study aimed to systematically summarize the common features of various molecules and structures within the cell membrane, cytoplasm, and nucleus responsible for sensing microgravity. It further examin

Adaptations to hypoxia in the vertebrate retina.

As part of the CNS, the retina maintains high metabolic activity yet is more tolerant to the adverse effects of low oxygen (hypoxia) than the brain. Tissue hypoxia is a component of ischaemia and has devastating effects on the CNS, including neuronal death. Differences in blood supply, oxygen consumption, and the expression of excitatory receptors and oxygen-binding proteins contribute to regional differences in the sensitivity of retinal neurons to hypoxia. Although highly variable and species-dependent, the mammalian retina has the capacity to withstand the effects of oxygen deprivation thro

The predatory behavior of ants: an impressive panoply of morphological adaptations.

Ants are typically omnivorous insects with ovoid heads equipped with short mandibles, but there is great diversity in both the adaptations of their morphological and behavioral traits, as well as their dietary habits. Here, we review the variety of form and function in ant predation. Predation and scavenging were likely the plesiomorphic modes, with Cretaceous ants evolving mandibles well-suited to prey capture. In contemporary ground-dwelling species, mandible morphology can vary with adaptations to capturing particular prey as they may possess trap-jaw, snapping, or falciform mandibles chara

Novel rebreathing adaptation extends dive time in a semi-aquatic lizard.

Bubble use evolved in many small invertebrates to enable underwater respiration, but, until recently, there has been no evidence that vertebrate animals use bubbles in a similar manner. Only one group of vertebrates, semi-aquatic Anolis lizards, may be an exception: these lizards dive underwater when threatened and, while underwater, rebreathe a bubble of air over their nostrils. Although it seems that rebreathing should be adaptive, possibly functioning to extend the time that lizards remain in underwater refugia, this has not been empirically tested. Here, I demonstrate that rebreathing serv
