Morphological and electrical properties of oligodendrocytes in the white matter of the corpus callosum and cerebellum.

In the central nervous system, electrical signals passing along nerve cells are speeded by cells called oligodendrocytes, which wrap the nerve cells with a fatty layer called myelin. This layer is important for rapid information processing, and is often lost in disease, causing mental or physical impairment in multiple sclerosis, stroke, cerebral palsy and spinal cord injury. The myelin speeds the information flow in two ways, by decreasing the capacitance of the nerve cell and by increasing its membrane resistance, but little is known about the latter aspect of myelin function. By recording e

A Gradient in Synaptic Strength and Plasticity among Motoneurons Provides a Peripheral Mechanism for Locomotor Control.

The recruitment of motoneurons during force generation follows a general pattern that has been confirmed across diverse species [1-3]. Motoneurons are recruited systematically according to synaptic inputs and intrinsic cellular properties and corresponding to movements of different intensities. However, much less is known about the output properties of individual motoneurons and how they affect the translation of motoneuron recruitment to the strength of muscle contractions. In larval zebrafish, spinal motoneurons are recruited in a topographic gradient according to their input resistance (Rin

Ionic conductance determinants of synaptic memory nets and their implications for Alzheimer's disease.

Electrical and chemical signals representing macroscopic "perturbations" in brain networks engage large numbers of transient "microscopic" ionic channel fluctuations in producing long-lasting changes of conductance (and thus potential). Repeated electrical and chemical signals that occur during associative training of living organisms (from mollusc to mammal) can cause ionic conductance changes lasting from days to many weeks. If a stimulus pattern reoccurs with sufficient frequency, voltage-dependent K(+) conductances-responsible for both synaptic and intrinsic membrane currents-become progre

Indirect synaptic inputs from filiform hair sensory neurons contribute to the receptive fields of giant interneurons in the first-instar cockroach.

The first-instar cockroach, Periplaneta americana, detects air movements using four filiform hair sensilla, which make synaptic connections to seven pairs of giant interneurons (GIs) in the terminal abdominal ganglion. The directional sensitivities of some of the GIs, predicted from their patterns of monosynaptic inputs, may not be the same as in the second instar or adult. Intracellular recordings were made to determine the contribution of polysynaptic inputs to the receptive fields of first-instar GIs. The ventral GI1, and the dorsal GI5, GI6, and GI7 were all found to have indirect synaptic

Changes in presynaptic release of acetylcholine during development of tolerance to the anticholinesterase, DFP.

The rat myenteric plexus was used as a peripheral model for studying muscarinic modulation of acetylcholine (ACh) release from presynaptic muscarinic neurons during development of tolerance to the anticholinesterase agent, diisopropylfluorophosphate (DFP). DFP in arachis oil was administered subcutaneously to intact animals according to both acute and chronic regimens, with arachis oil injections serving as controls. Post-mortem analyses showed that the mean AChE activity level in whole brain was reduced under all DFP conditions to 18.0 +/- 1.4% when compared with the control level. After 10 d

Synaptic excitatory and inhibitory interactions at distal dendritic sites on mitral cells in the isolated turtle olfactory bulb.

The olfactory nerves terminate exclusively on the distal dendritic tufts of mitral cells in the olfactory bulb, which makes this a favorable model for analysis of synaptic responses in distal dendrites. Intracellular recordings of responses to olfactory nerve volleys have been obtained in the isolated turtle olfactory bulb. Single mitral cells usually responded with excitatory postsynaptic potentials (EPSPs) to volleys in two different bundles, indicating convergence from separate receptor neuron populations. Paired volleys revealed long-lasting inhibition of a test EPSP by a conditioning voll

Long-lasting facilitation of a synaptic potential following tetanization in the in vitro hippocampal slice.

Field potentials evoked by stimulation of afferent fibers in stratum radiatum were recorded in the CA1 region of the hippocampal slice maintained in vitro. Stimulation rates of 3-50/sec produced a large increase in amplitude of the population spike in CA1. This increase was maintained for several hours after the tetanization. The facilitation phenomenon appeared to be specific to the synapse of stratum radiatum afferents onto CA1 pyramidal cells since: (1) stimulation outside the radiatum layer did not produce the effect, (2) antidromic field potentials recorded in CA3 were unchanged, (3) EPSP

The effect of a preceding stimulus on temporal facilitation at corticomotoneuronal synapses.

1. Intracellular recordings were made of minimal corticomotoneuronal e.p.s.p.s in lumbar motoneurones of anaesthetized monkeys. For intervals of 2 msec and greater between paired cortical shocks, the average time course of facilitation of the second e.p.s.p. with respect to the first could be fitted closely by a negative exponential with a time constant of 10 msec.2. In the same motoneurones, ;triplets' of corticomotoneuronal e.p.s.p.s were generated by delivering three identical stimuli to the motor cortex. Considering the triplet as a conditioning e.p.s.p. followed by a test pair, the facili

Inhibitory mechanisms in lateral geniculate nucleus of rat.

1. An examination was made of the mechanisms responsible for the inhibition of synaptic transmission through the lateral geniculate nucleus (LGN) of the rat following a single shock to the optic nerve.2. In the rat anaesthetized with paraldehyde we found no evidence that optic nerve stimulation produced any presynaptic inhibition in LGN. In agreement with other workers it was found that repetitive stimulation of visual cortex produced effects attributable to presynaptic inhibition. However, this was of small magnitude in the conditions of our experiments.3. Stimulation of the optic nerve elici

Nitrergic nerves derived from the pterygopalatine ganglion innervate arteries irrigating the cerebrum but not the cerebellum and brain stem in monkeys.

The functional roles of the nitrergic nerves innervating the monkey cerebral artery were evaluated in a tension-response study examining isolated arteries in vitro and cerebral angiography in vivo. Nicotine produced relaxation of arteries by stimulation of nerve terminals innervating isolated monkey arteries irrigating the cerebrum, cerebellum and brain stem. Relaxation of arteries induced by nicotine was abolished by treatment with N(G)-nitro-L-arginine, a nitric oxide synthase inhibitor, and was restored by addition of L-arginine. Cerebral angiography showed that electrical stimulation of th

Temporally interfering electric fields brain stimulation in primary motor cortex of mice promotes motor skill through enhancing neuroplasticity.

Temporal interference (TI) electric field brain stimulation is a novel neuromodulation technique that enables the non-invasive modulation of deep brain regions, but few advances about TI stimulation effectiveness and mechanisms have been reported. Conventional transcranial alternating current stimulation (tACS) can enhance motor skills, whether TI stimulation has an effect on motor skills in mice has not been elucidated. In the present study, TI stimulation was proved to stimulating noninvasively primary motor cortex (M1) of mice, and that TI stimulation with an envelope wave frequency of 20 H

Therapeutic potential of positive allosteric modulators of AMPA receptors in brain disorders.

α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPA receptors) are ionotropic glutamate receptors that mediate fast excitatory neurotransmission in the central nervous system. By translating chemical signals into rapid electrical depolarization, AMPA receptors, together with NMDA and kainate receptors, coordinate synaptic plasticity at glutamatergic synapses, thereby shaping learning, memory, and higher-order cognitive functions. Neurological disorders characterized by glutamatergic dysfunction represent a major clinical challenge, largely because pharmacological targeting of

Quantifying electrostatic control of docking and binding energetics in functional Cx36 gap junctions.

Connexin36 (Cx36) is broadly expressed in neurons and serves as the principal protein that forms interneuronal gap junctions (GJs), also known as electrical synapses. Recent high-resolution structures of human Cx36 GJ have revealed crucial electrostatic interactions (ESIs) of charged residues between two docked Cx36 hemichannels at the second extracellular (E2) loops. Despite their structural importance, the mechanistic roles of these ESIs remain poorly understood. To investigate their significance, we systematically designed and tested a series of missense variants targeting key E2 interface …

Adaptive remodeling of rat adrenomedullary stimulus-secretion coupling in a chronic hypertensive environment.

Chronic elevated blood pressure impinges on the functioning of multiple organs and therefore harms body homeostasis. Elucidating the protective mechanisms whereby the organism copes with sustained or repetitive blood pressure rises is therefore a topical challenge. Here we address this issue in the adrenal medulla, the master neuroendocrine tissue involved in the secretion of catecholamines, influential hormones in blood pressure regulation. Combining electrophysiological techniques with catecholamine secretion assays on acute adrenal slices from spontaneously hypertensive rats, we show that c

The impact of calcium current reversal on neurotransmitter release in the electrically stimulated retina.

OBJECTIVE: In spite of intense theoretical and experimental investigations on electrical nerve stimulation, the influence of reversed ion currents on network activity during extracellular stimulation has not been investigated so far. APPROACH: Here, the impact of calcium current reversal on neurotransmitter release during subretinal stimulation was analyzed with a computational multi-compartment model of a retinal bipolar cell (BC) that was coupled with a four-pool model for the exocytosis from its ribbon synapses. Emphasis was laid on calcium channel dynamics and how these channels influence

Bifacial Interface Engineering of Perovskite Memristors for High-Performance Optoelectronic Synapses and Neuromorphic Vision Systems.

Conventional complementary metal-oxide-semiconductor (CMOS)-based intelligent visual systems suffer from inherent bottlenecks of high energy consumption and latency, while perovskite memristors are constrained by critical issues including defect-induced instability, unimodal modulation, and random ion migration, hindering their practical deployment. Herein, a universal bifacial engineering strategy is proposed to address these challenges, featuring CF3-PEAI modification at both the prenucleation and postgrowth interfaces of the perovskite layer. The resulting memristor devices exhibit excellen
