Intranasal delivery of a live attenuated vaccine confers efficient protection against Riemerella anatipestifer serotype 1 in ducklings.

Mucosal immunity serves as the first line of defense against pathogenic infections in animals. Respiratory mucosal vaccines elicit multi-layered immune responses that confer distinct advantages for disease prevention and control. Riemerella anatipestifer (RA) infection causes duck infectious serositis, leading to sustained economic losses in the duck industry. Commercially available inactivated RA vaccines are hampered by drawbacks such as stress induction and adjuvant residue. Targeting the RA-YM strain of serotype 1, the dominant epidemic RA serotype in China, this study employed markerless

Mapping the evolutionary and translational landscape of antibiotic resistance genes in Elizabethkingia anopheles.

Elizabethkingia anophelis is an emerging multidrug-resistant pathogen of significant clinical concern, yet little is known about the evolutionary and translational dynamics of its resistance genes. This study aimed to analyze codon and amino acid usage patterns in the genome of E. anophelis strain 502 and three resistance genes (blaB-11, blaCME-1, and blaGOB-6) to understand the forces shaping their evolution and expression. The genome exhibited moderate codon usage bias, while resistance genes displayed distinct patterns, reflecting gene-specific selection pressures. Nucleotide composition fa

Threshold level of Riemerella anatipestifer crossing blood-brain barrier and expression profiles of immune-related proteins in blood and brain tissue from infected ducks.

Riemerella anatipestifer (R. anatipestifer) is a Gram-negative bacterium that crosses the blood-brain barrier (BBB) and causes meningitis with neurological symptoms in infected ducks. A threshold level of bacteremia must be reached before the BBB can be breached. In this study, the bacterial burden and expression profiles of immune-related proteins in blood and brain tissue samples from R. anatipestifer-infected ducks were investigated. The data showed that R. anatipestifer could cross the BBB with low-level bacteremia of 7.5 × 102 CFU/ml in infected blood. Analysis of immune-related proteins

Quinolone-resistant phenotype of Flavobacterium columnare isolates harbouring point mutations both in gyrA and parC but not in gyrB or parE.

OBJECTIVES: The aim of this study was to determine mutations associated with a quinolone-resistant (QR) phenotype of Flavobacterium columnare isolates. METHODS: The susceptibility of 53 F. columnare isolates to 11 antimicrobials, including 2 quinolones, was investigated by the disk diffusion method. Oxolinic acid (OXO) was subsequently chosen for minimum inhibitory concentration (MIC) assay. Sequence analysis of four genes within the quinolone resistance-determining regions (QRDRs) of OXO-resistant F. columnare compared with susceptible isolates was subsequently performed. RESULTS: The disk

Pharmacokinetic and pharmacodynamic properties of gamithromycin in turkey poults with respect to Ornithobacterium rhinotracheale.

The macrolide gamithromycin (GAM) has the ability to accumulate in tissues of the respiratory tract. Consequently, GAM might be a suitable antibiotic to treat bacterial respiratory infections in poultry, such as Ornithobacterium rhinotracheale. As O. rhinotracheale infections are common in turkey flocks, the aim of this study was to determine the pharmacokinetic (PK) parameters of GAM in plasma, lung tissue, and pulmonary epithelial lining fluid (PELF) of turkeys and to correlate them with pharmacodynamic (PD) characteristics (PK/PD). The animal experiment was performed with 64 turkeys, which

Transmission of an uncultivated Bergeyella strain from the oral cavity to amniotic fluid in a case of preterm birth.

Intrauterine infection is a recognized cause of preterm birth. The infectious organisms are believed to originate primarily from the vaginal tract and secondarily from other parts of the body. It is plausible that microbes in the oral cavity can be transmitted to the pregnant uterus. However, direct evidence supporting such a transmission is lacking. In this study, amniotic fluids of 34 pregnant women were examined by PCR using 16S and 23S rRNA universally conserved primers. Bacterial DNA was amplified from the only patient with clinical intrauterine infection and histologic necrotizing acute

Clinical Profile, Comorbidities, and Antimicrobial Resistance Patterns of Myroides Species Isolated From Nosocomial Urinary Tract Infections at a Tertiary Care Center in India: A Retrospective Observational Study.

Background Myroides species are gram-negative, aerobic bacilli within the family Flavobacteriaceae that have emerged as clinically significant, multidrug-resistant nosocomial pathogens. Although historically regarded as organisms of low pathogenicity, they are increasingly associated with opportunistic infections, particularly urinary tract infections (UTIs), among hospitalized, immunocompromised, and catheterized patients. Reports of Myroides infections have increased in recent years across different geographical regions, highlighting its emerging global clinical significance. The incidence o…

Comparative genomic analysis of Flavobacterium species causing columnaris disease of freshwater fish in Thailand: insights into virulence and resistance mechanisms.

BACKGROUND: Columnaris disease, a prevalent disease among farmed and wild freshwater fish, is caused by the Flavobacterium columnare group, which includes four distinct species: F. columnare, F. oreochromis, F. covae, and F. davisii. Among these, F. oreochromis, F. covae, and F. davisii are particularly prevalent in farmed freshwater fish in Thailand. In this study, a comparative genomic analysis of 22 isolates was conducted to elucidate virulence factors, antibiotic resistance genes (ARGs), genomic islands (GIs), phages, insertion elements (ISs), and clustered regularly interspaced short pali

Ornithobacterium rhinotracheale in Moroccan Poultry: Antimicrobial Susceptibility Profiles, Characterization of Recent Isolates, and Retrospective Study (2019-23) of Its Occurrence in Different Poultry Production Systems.

Ornithobacterium rhinotracheale (ORT) is one of the most important respiratory pathogens of poultry, notably in turkeys, while Riemerella anatipestifer (RA) is the etiologic agent of infectious serositis, the most economically significant disease of domestic ducks and, to a lesser extent, geese and turkeys. Currently, little is known about ORT and RA infections in Moroccan poultry. Thus, the main aim of this study was to describe ORT incidence in Moroccan poultry flocks during the period from 2019 to 2023 and gain better insights into genetic characteristics in combination with assessment of a

Expression profiles of NOD1 and NOD2 and pathological changes in gills during Flavobacterium columnare infection in yellow catfish, Tachysurus fulvidraco.

NOD-like receptors are significant contributors to the immune response of fish against different types of pathogen invasion. NOD1 and NOD2 genes of yellow catfish (Tachysurus fulvidraco) were identified and characterized in this study. Yellow catfish NOD1 and NOD2 have open reading frames (ORFs) of 2841 and 2949 bp, encoding 946 and 982 amino acids, respectively. Both NOD1 and NOD2 are intracellular proteins lacking transmembrane regions and signal peptides. Sequence homology analysis revealed that the protein sequences of NOD1 and NOD2 of yellow catfish are highly similar to those of channel

Immunoglobulin (Ig) heavy chain gene locus and immune responses upon parasitic, bacterial and fungal infection in loach, Misgurnus anguillicaudatus.

Teleost fish are the most primitive bony vertebrates that contain immunoglobulin (Ig). Although teleost Ig is known to be important during tetrapod evolution and comparative immunology, little is known about the genomic organization of the immunoglobulin heavy-chain (IgH) locus. Here, three Ig isotype classes, IgM, IgD and IgT, were firstly identified in dojo loach (Misgurnus anguillicaudatus), and the IgH locus covering τ, μ and δ genes was also illustrated. Variable (V) gene segments lie upstream of two tandem diversity (D), joining (J) and constant (C) clusters and the genomic organization

Whole-genome sequencing of a large collection of Myroides odoratimimus and Myroides odoratus isolates and antimicrobial susceptibility studies.

The genus Myroides comprises several species of Gram-negative, non-motile, and non-fermenting bacteria, which have been regarded as non-pathogenic for decades. Multiple recent reports, however, underscore the pathogenic potential that Myroides sp. possesses for humans. These bacteria seem to be resistant to a wide range of antibiotics (including ß-lactams and aminoglycosides). Therefore, treatment options are limited. Knowledge of antimicrobial resistance, however, is based on only one meaningful comprehensive study and on data published from case reports. This lack of data motivated us to tes

Chryseobacterium indologenes: an emerging infection in the USA.

Nursing home-associated infections and antibiotic resistant pathogens constitute common and serious problems in the geriatric population.Chryseobacterium indologenes, a non-motile Gram-negative rod, though widely distributed in nature, is an uncommon human pathogen. Typically thought of as an organism of low virulence, it may cause serious infections, particularly among the immunocompromised. The majority of reported cases are nosocomial, often associated with immunosuppression or indwelling catheters. It has been reported as the causative agent in bacteraemia, peritonitis, pneumonia, empyema,

Riemerella anatipestifer OMP85, a BamA family outer membrane protein, enhances virulence through recruiting host complement regulator vitronectin to mediate complement evasion.

Riemerella anatipestifer infection causes septicemia and polyserositis in poultry, leading to serious economic losses to the global poultry industry. The ability to break through the host complement defense barrier is an important characteristic of R. anatipestifer. However, the underlying mechanisms are still not well understood at present. This study identifies outer membrane protein 85, a conserved member in the OMP family, as a key mediator that recruits of host complement regulatory factor vitronectinto inhibit formation of the membrane attack complex allowing R. anatipestifer to evade ho
