Determination of extracellular and intracellular pH of Bacillus subtilis suspension under CO2 treatment.

In this study, we consider the effect of carbon dioxide (CO(2)) on the intracellular and extracellular pH of a saline solution of a test-microorganisms Bacillus subtilis. The cytoplasmatic pH was determined by means of a flow cytometry with the fluorescent probe 5(and 6-)-carboxyfluorescein ester (cFSE). The physiological suspension of cells with the addition of the probe was first exposed to high pressure CO(2) for 5 min at different temperatures. The flow cytometry analysis indicated an intracellular depletion inside the cell caused by the action of CO(2), down to 3, the depletion being depe

Natural statin derivatives as potential therapy to reduce intestinal fluid loss in cholera.

As a leading cause of death in children under 5 years old, secretory diarrheas including cholera are characterized by excessive intestinal fluid secretion driven by enterotoxin-induced cAMP-dependent intestinal chloride transport. This study aimed to identify fungal bioactive metabolites possessing anti-secretory effects against cAMP-dependent chloride secretion in intestinal epithelial cells. Using electrophysiological analyses in human intestinal epithelial (T84) cells, five fungus-derived statin derivatives including α,β-dehydrolovastatin (DHLV), α,β-dehydrodihydromonacolin K, lovastatin, m

Psoriatic arthropathy and crystal-induced synovitis.

We demonstrated the simultaneous occurrence of psoriatic arthropathy and the 2 major types of crystalline arthritis--pseudogout and gout. The crystals found within aspirated synovial fluid and exudative leukocytes were unequivocally identified using the compensating polarizing microscope as calcium pyrophosphate dihydrate in one case and sodium monourate in the other. The surface topography of the intracellular sodium monourate crystals were also displayed.

Macropinocytosis of type XVII collagen induced by bullous pemphigoid IgG is regulated via protein kinase C.

Macropinocytosis is an endocytic pathway that is involved in the nonselective fluid uptake of extracellular fluid. Bullous pemphigoid (BP) is an autoimmune subepidermal blistering disease associated with autoantibodies to type XVII collagen (COL17), which is a component of hemidesmosome. When keratinocytes are treated with BP-IgG, COL17 internalizes into cells by way of the macropinocytosis. We investigated the mechanism of COL17 macropinocytosis using DJM-1 cells, a cutaneous squamous cell carcinoma cell line. First, non-hemidesmosomal COL17 was preferentially depleted by stimulation with the

Cerebral edema and neurointensive care.

Cerebral edema may be vasogenic or cytoxic, and its main significance is raised intracranial pressure. Vasogenic edema results from damage to the blood vessel wall. The hallmark of cytotoxic edema is intracellular accumulation of fluid which occurs because of the exposure of the cells of the brain to toxic substances. Few diagnostic tests will document edema; the best is presently the CAT scan.

Participation of the mineralocorticoid receptor in cardiac and vascular remodeling.

The mineralocorticoid receptor mediates the effects of aldosterone. In addition to its influence on fluid and electrolyte, aldosterone plays a role in cardiac hypertrophy, tissue fibrosis, and modulation of vascular reactivity. However, the intracellular signaling mechanisms induced by aldosterone are not fully understood. Aldosterone causes slow transcriptional and rapid non-transcriptional effects. The latter include changes in intracellular calcium, c-Jun N-terminal kinase (JNK) and extracellularly regulated kinase (ERK) phosphorylation. In addition, autocrine loops involving other molecule

Anaerobes and short-chain fatty acids in crevicular fluid from adults with chronic periodontitis.

Pathogeny of adult chronic periodontitis is still unclear. Bacteriological and chemical analysis of crevicular fluid have shown, in active sites of the disease, a simultaneous presence of anaerobes and their major by-product: short-chain fatty acids. The last can decrease "in vitro" the neutrophil intracellular pH, whenever these cells are incubated in an acid medium. Clinical investigations are scarce which hold out data useful to attempt verifying this possible physiopathological mechanism. This work shows the presence of anaerobes in the active periodontal pockets, together with the presenc

Fluid transport and cystogenesis in autosomal dominant polycystic kidney disease.

Autosomal dominant polycystic kidney disease (ADPKD) is the most frequent inherited nephropathy. The development and enlargement of cysts in ADPKD requires tubular cell proliferation, abnormalities in the extracellular matrix and transepithelial fluid secretion. Multiple studies have suggested that fluid secretion across ADPKD cyst-lining cells is driven by the transepithelial secretion of chloride, mediated by the apical CFTR channel and specific basolateral transporters. The whole secretory process is stimulated by increased levels of cAMP in the cells, probably reflecting modifications in t

A physiological model for the distribution of injected agents, with special reference to pethidine.

The model is based on Mapleson's Model P for inhaled anaesthetics, but has compartments for lungs, peripheral shunt, kidneys, portal bed, liver, other viscera, muscle, other lean, fat, brain, i.m. injection site and for various blood "pools". Intracellular and extracellular fluids are represented separately in each compartment and in blood. In each fluid, four forms of the agent are distinguished: unionized dissolved in water ("standard" form), ionized dissolved in water, unionized dissolved in lipid, and bound to protein. Equations define the equilibrium between these four forms in any one fl

CNS Viral Infections-What to Consider for Improving Drug Treatment: A Plea for Using Mathematical Modeling Approaches.

Neurotropic viruses may cause meningitis, myelitis, encephalitis, or meningoencephalitis. These inflammatory conditions of the central nervous system (CNS) may have serious and devastating consequences if not treated adequately. In this review, we first summarize how neurotropic viruses can enter the CNS by (1) crossing the blood-brain barrier or blood-cerebrospinal fluid barrier; (2) invading the nose via the olfactory route; or (3) invading the peripheral nervous system. Neurotropic viruses may then enter the intracellular space of brain cells via endocytosis and/or membrane fusion. Antivira

DA-6034-induced mucin secretion via Ca2+-dependent pathways through P2Y receptor stimulation.

PURPOSE: We evaluated whether DA-6034 is involved in mucin secretion via P2Y receptor activation and/or intracellular Ca2+ concentration ([Ca2+]i) change. Also, we investigated the effect of P2Y receptor inhibitors or Ca2+ chelators on the DA-6034-induced mucin secretion and [Ca2+]i increases. METHODS: Effects of DA-6034 on mucin expression in primary, cultured, conjunctival epithelial cells was studied using RT-PCR, Western blot analysis, and periodic acid-schiff (PAS) staining. To evaluate thin film layer thickness generated by mucin and fluid secretion, cells were incubated in DA-6034 with

Polycystin-2 expression and function in adult mouse lacrimal acinar cells.

PURPOSE: Lacrimal glands regulate the production and secretion of tear fluid. Dysfunction of lacrimal gland acinar cells can ultimately result in ocular surface disorders, such as dry eye disease. Ca(2+) homeostasis is tightly regulated in the cellular environment, and secretion from the acinar cells of the lacrimal gland is regulated by both cholinergic and adrenergic stimuli, which both result in changes in the cytosolic Ca(2+) concentration. We have previously described the detailed intracellular distribution of inositol-1,4,5-trisphosphate receptors (IP(3)Rs), and ryanodine receptors (RyRs

Concentrations of phenoxymethylpenicillin and cefadroxil in tonsillar tissue and tonsillar surface fluid.

Thirty patients who underwent elective tonsillectomy were given phenoxymethylpenicillin (0.8 g) or cefadroxil (1 g) at different times before operation. The concentrations of the antibiotics were analysed in serum, tonsillar tissue, fluid from the surface of the tonsils, and mixed saliva. The concentrations in tonsillar tissue for both drugs were much lower than the corresponding serum concentrations. This apparently low tissue accessibility could be ascribed to the limited intracellular penetration of beta-lactam antibiotics. For both antibiotics the concentrations in the tonsillar surface fl

Progressive changes in cortical water and electrolyte content at three stages of rat infantile hydrocephalus and the effect of shunt treatment.

Infantile hydrocephalus causes injury to the developing brain and despite surgical treatment, neurological deficits persist. The H-Tx rat develops inherited hydrocephalus in late gestation. Rapid postnatal ventricular enlargement, results in severe hydrocephalus by 21 days after birth. This is accompanied by changes in cortical morphology and metabolite content that indicate possible changes in intracellular composition. This study has tested the hypothesis that tissue water and electrolyte content is altered in hydrocephalus. The objective was to gain further insight into the mechanisms leadi

Sodium requirement for insulin release: putative role in regulation of intracellular pH.

The effects on insulin release of Na+ removal, alteration of extracellular pH, and inhibition of acid extrusion processes were examined using freshly isolated and 46-h cultured islets of Langerhans. Na+ removal inhibited the secretory response to 16.7 mM glucose specifically in fresh islets but stimulated the low-glucose response in cultured islets. These divergent effects of Na+ depletion were perfectly mimicked by amiloride, an inhibitor of Na+-H+ exchange, and could be overcome in a raised HCO-3, pH 8.0 medium. Simply raising extracellular pH at constant HCO-3 had no effect. Na+ removal als

Intracellular electrolytes in cardiac failure.

In congestive heart failure (CHF) there are several compensatory mechanisms operating which may influence electrolyte metabolism. The activation of the renin-angiotensin-aldosterone system causes retention of sodium (Na) and losses of potassium (K) and magnesium (Mg). The secondary hyperaldosteronism may give rise to high intracellular Na and low intracellular K through a direct permeability effect on the cell membrane. The Mg deficiency may lead to a further increase of intracellular Na and decrease of intracellular K since Mg is a necessary ion for the function of the Na-K pump. In 297 patie

Ungulate cardiac purkinje fibres: the influence of intracellular pH on the electrical cell-to-cell coupling.

1. Passive electrical properties of sheep cardiac Purkinje fibres were assessed by performing linear cable analysis. In a separate set of experiments pH(i) was monitored using recessed-tip pH-sensitive micro-electrodes.2. In Tris-buffered Tyrode solution (nominally CO(2)-free), the pH(i) was 7.27, in bicarbonate-buffered solution equilibrated with 6% CO(2), the mean pH(i) was 7.02.3. Application of 15 mM-NH(4)Cl produced a rapid intracellular alkalinization (0.19 pH units), followed by a slower acidification. Removal of NH(4)Cl gave rise to a slow and transient intracellular acidification (0.5

Mitochondrial malfunction mediates impaired cholinergic Ca2+ signalling and submandibular salivary gland dysfunction in diabetes.

Xerostomia (dry-mouth syndrome) is a painful and debilitating condition that frequently occurs in individuals with diabetes and is associated with impaired saliva production and salivary gland hypofunction. Saliva fluid production relies on Ca2+-coupled secretion driven by neurotransmitter stimulation of submandibular acinar cells. Although impairments in intracellular Ca2+ signalling have been reported in various xerostomia models, the specific Ca2+-dependent mechanisms underlying saliva fluid hypofunction in diabetes remain unclear. In this study, we show that diabetic animals exhibit severe

Serotonin triggers cAMP and PKA-mediated intracellular calcium waves in Malpighian tubules of Rhodnius prolixus.

Rhodnius prolixus is a hematophagous insect vector of Chagas disease capable of ingesting up to 10 times its unfed body weight in blood in a single meal. The excess water and ions ingested with the meal are expelled through a rapid postprandial diuresis driven by the Malpighian tubules. Diuresis is triggered by at least two diuretic hormones, a CRF-related peptide and serotonin, which were traditionally believed to trigger cAMP as an intracellular second messenger. Recently, calcium has been suggested to act as a second messenger in serotonin-stimulated Malpighian tubules. Thus, we tested the
