Different coatings on magnetic nanoparticles dictate their degradation kinetics in vivo for 15 months after intravenous administration in mice.

BACKGROUND: The surface coating of iron oxide magnetic nanoparticle (MNPs) drives their intracellular trafficking and degradation in endolysosomes, as well as dictating other cellular outcomes. As such, we assessed whether MNP coatings might influence their biodistribution, their accumulation in certain organs and their turnover therein, processes that must be understood in vivo to optimize the design of nanoformulations for specific therapeutic/diagnostic needs. RESULTS: In this study, three different MNP coatings were analyzed, each conferring the identical 12 nm iron oxide cores with diffe

New methods for amlodipine and valsartan native spectrofluorimetric determination, with factors optimization study.

Four native fluorescence methods were suggested for simultaneous determination of amlodipine (AML) and valsartan (VAL). These methods were based on excitation of both drugs at λ(ex) 300 nm, in one step, to give maximum emission at λ(em) 378 and 496 nm for AML and VAL, respectively. The first method, single λ(ex) method, was used without any additions. The sensitivity of this method was further increased by the addition of hydroxy propylmethyl cellulose (HPMC) surfactant, β-cyclodextrin, or ferric oxide magnetite nanoparticles, in the other three methods. Different types of surfactants, and dif…

Verification and validation in canine cadaver of an MPI-integrable RF head coil for magnetic particle hyperthermia.

PURPOSE: Magnetic particle hyperthermia (MPH), the mild heating of solid tumors with magnetic nanoparticles, is an emerging treatment modality used as an adjuvant to therapies such as ionizing radiation. MPH can be paired with magnetic particle imaging (MPI) hardware to improve thermal treatment planning, spatially confined heating, and thermal dose delivery. In this work, we describe the design, verification, and validation of a radiofrequency (RF) head coil designed to integrate into a clinical MPI scanner. METHODS: The RF coil was designed to accommodate an adult human head, provide a homo

TriMag Microrobots: 3D-Printed Microrobots for Magnetic Actuation, Imaging, and Hyperthermia.

Microrobots hold immense potential in biomedical applications, including drug delivery, disease diagnostics, and minimally invasive surgeries. However, two key challenges hinder their clinical translation: achieving scalable and precision fabrication, and enabling non-invasive imaging and tracking within deep biological tissues. Magnetic particle imaging (MPI), a cutting-edge imaging modality, addresses these challenges by detecting the magnetization of nanoparticles and visualizing superparamagnetic nanoparticles (SPIONs) with sub-millimeter resolution, free from interference by biological ti

Unveiling Dipolar Interaction-Driven Magnetic Field Inhomogeneities in T2 MRI Contrast Agents.

Modulating local magnetic field inhomogeneities, combined with ultra-high-field MRI (UHF-MRI) is a promising strategy to enhance the performance of next-generation T2 contrast agents (CAs). Although dipolar interactions likely contribute to contrast enhancement, their role in magnetic inhomogeneities and proton dephasing remains poorly understood, limiting further optimization. In this study, the fundamental role of dipolar interactions on the modulation of the transverse relaxivity (r2) of iron oxide-based CAs is demonstrated by shaping local magnetic field inhomogeneities. A nanoscale-distan

Intermuscular Septum Delivery of High-Mechanical Injectable Magnetic Hydrogels to Improve Muscle Atrophy.

Injectable hydrogels often lack sufficient mechanical robustness for long-term function under physiological loading, limiting their application in musculoskeletal repair. Inspired by natural soft matter, we developed a hierarchical reinforcement strategy that combines sliding-ring structures with rigid magnetic nanocomposite microdomains. Through self-assembly driven by chelation and hydrogen bonding, hydrophobic drugs and magnetic nanoparticles formed microphase units within the hydrogel network, resulting in an injectable magneto-responsive platform (0.2F@UA-PC) with exceptional mechanical p

Preclinical and Clinical-Scale Magnetic Particle Imaging of Natural Killer Cells: in vitro and ex vivo Demonstration of Cellular Sensitivity, Resolution, and Quantification.

PURPOSE: Clinical adoption of NK cell immunotherapy is underway for medulloblastoma and osteosarcoma, however there is currently little feedback on cell fate after administration. We propose magnetic particle imaging (MPI) may have applications for the quantitative detection of NK cells. PROCEDURES: Human-derived NK-92 cells were labeled by co-incubation with iron oxide nanoparticles (VivoTrax™) for 24 h then excess nanoparticles were washed with centrifugation. Cytolytic activity of labeled versus unlabeled NK-92 cells was assessed after 4 h of co-incubation with medulloblastoma cells (DAOY)

Modified Jiles-Atherton Model for Dynamic Magnetization in X-Space Magnetic Particle Imaging.

OBJECTIVE: Magnetic particle imaging (MPI) is a promising medical modality that can image superparamagnetic iron-oxide nanoparticle (SPIO) concentration distributions safely and with high sensitivity. In the x-space reconstruction algorithm, the Langevin function is inaccurate in modeling the dynamic magnetization of SPIOs. This problem prevents the x-space algorithm from achieving a high spatial resolution reconstruction. METHODS: We propose a more accurate model to describe the dynamic magnetization of SPIOs, named the modified Jiles-Atherton (MJA) model, and apply it to the x-space algorit

Safety evaluation of PEGylated MNPs and p-PEGylated MNPs in SD rats.

Polyethylene glycol-coated magnetic nanoparticles (PEGylated MNPs) have demonstrated prominent advantages in cancer diagnosis and hyperthermia therapy. However, there is currently lack of standard mode and sufficient toxicity data for determining the delayed risk of PEGylated MNPs. Nevertheless, the toxicity potentials, especially those associated with the oxidative stress, were ubiquitously reported. In this study, PEGylated MNPs and p-PEGylated MNPs were administrated to SD (Sprague Dawley) rats by single intravenously injection, and various toxicity indicators were monitored till 56 days po

Functionalization of Fe3O4/rGO magnetic nanoparticles with resveratrol and in vitro DNA interaction.

Based on the previous research, we found that the magnetic nanocomposite Fe3O4/rGO (reduced graphene oxide) has a good drug loading effect. Therefore, in this paper, we studied the positive role of Fe3O4/rGO as a drug carrier in the interaction between resveratrol (RES) and calf-thymus DNA (ct-DNA). The fluorescence experiment is used to evaluate by the Stern-Volmer equation, the quenching constant of RES - ct-DNA system with and without Fe3O4/rGO decreases with the increasing temperature. It was found the quenching mode of RES - ct-DNA and Fe3O4/rGO - RES - ct-DNA systems were all static quen

β-Glucosidase Immobilized on Magnetic Nanoparticles: Controlling Biomolecule Footprint and Particle Functional Group Density to Navigate the Activity-Stability Tradeoff.

In the present work, the immobilized footprint of β-glucosidase (BGL) on silica-coated iron oxide was explored to produce reusable catalysts with flexible active sites for high activity and heightened storage stability. Synthesized iron oxide particles were coated with silica and functionalized with various densities of (3-aminopropyl)triethoxysilane (APTES) to obtain particles with amine densities ranging from 0 to 3 × 10-5 mol/g particle. The amine-modified particles were activated with glutaraldehyde, and subsequently, BGL was immobilized using either a 0.1 or 1 mg/mL enzyme solution to pro

Nanomagnetic lateral flow assay for high-precision quantification of diagnostically relevant concentrations of serum TSH.

Thyroid stimulating hormone (TSH) is the first-line marker for initial evaluation of the thyroid gland function. We present a lateral flow immunoassay based on superparamagnetic nanolabels for rapid (<25 min) quantitative determination of TSH at a point of care. The demonstrated limit of detection (LOD) of 0.017 μIU/mL in human serum is on the level of third-generation TSH laboratory tests. The wide linear dynamic range of more than 3 orders covers the whole range of clinically relevant TSH concentrations for confident quantitative diagnostics of the gland function from hyper- to hypothyroidis
