[Infected cervical epidural Ewing's sarcoma (case report)].

INTRODUCTION: Ewing's sarcoma is a relatively rare osseous malignant tumour. The age of onset is generally in the youth. Epidural localisation of the malignant osseous tumour Ewing's sarcoma is exceptional. Only 29 cases were reported in surgical literature including only one infected case. OBSERVATION: We report a case of a 24 years old patient who displayed a complete flaccid tétraplégia with T1 sensitive level. Spinal cord MRI documented a cervical posterior epidural process with peripheric gadolinium enhancement. Patient was operated in emergency. A C6 and C7 laminectomy allowed complete

Hypertrichosis lanuginosa acquisita preceding extraskeletal Ewing's sarcoma.

A 62-year-old woman with acquired hypertrichosis lanuginosa as a paraneoplastic presenting sign of an extraskeletal Ewing's sarcoma is described. Despite initial comprehensive screening to rule out an associated malignancy, a definitive diagnosis of sarcoma was established only 1 year after the onset of the cutaneous symptoms. To the best of our knowledge, this is the first report of hypertrichosis lanuginosa acquisita associated with a soft tissue sarcoma. Our observation expands the spectrum of malignancies associated with this uncommon paraneoplastic disorder.

CNS involvement in Ewing's sarcoma. A report of 12 cases.

Ewing's sarcoma is one of the malignant bone tumours, which occurs in various bones of the body. When the spinal column or skull bones are involved, it may cause spinal cord compression or brain invasion. In this study, 12 cases of Ewing's sarcoma with central nervous system (CNS) involvement are presented. Seven of them were male and five female. In seven of the cases tumoural mass was located in the spinal column, whereas in the others it was found in the cranium. The major complaints were motor weakness in the lower extremities in seven cases, and a mass on the cranial vault in four cases.

Primary Ewing's sarcoma involving the bones of the head and neck.

Ewing's sarcoma is a rare tumor of the bone. In the Intergroup Ewing's Sarcoma Study (IESS) approximately 4% of the primary bone tumors arose in the bones of the head and neck. The mean age of patients was 10.9 years; boys slightly dominated the group. Signs and symptoms were local in distribution, with a mass or swelling most frequent. As in other sites, we categorized the dominant histologic pattern as diffuse or filigree, the latter carrying a more unfavorable prognosis. The radiographic appearance may be atypical as compared to Ewing's sarcoma at other sites. Prognosis of head and neck Ewi

Curability of Ewing's sarcoma and considerations for future therapeutic trials.

Twenty previously untreated children with primary Ewing's sarcoma and 8 children with primary tumor and metastatic disease were treated with surgery or radiation therapy (6,000-7,000 rads) for their primary tumor and T-2 chemotherapy. Of the 20 children with primary Ewing's sarcoma treated with T-2 "adjuvant" chemotherapy, 15 had no evidence of recurrent disease for from 31+-82+ months (median 46+ months) from the start of treatment. The actuarial 5-year disease-free survival rate for this group of patients was 75%. Eight patients presenting with metastatic disease had complete responses to T-

Druggable upregulated proteins in EWS-FLI-driven Ewing sarcoma as emerging new therapeutic targets.

Ewing sarcoma (ES) is a highly aggressive soft tissue tumor that primarily affects the long bones of children and young adults. It is distinguished by a characteristic chromosomal translocation between the Ewing sarcoma breakpoint region 1 (EWS) gene and the erythroblast transformation-specific (ETS) family of genes, most commonly resulting in the EWS-friend leukemia integration 1 (EWS-FLI1) fusion gene. This translocation is observed in approximately 80%-85% of ES cases. This fusion gene encodes a non-physiological chimeric fusion protein that plays a central role in tumorigenesis by interact

Recombinant production, biochemical characterization, and in vitro evaluation of an EWS-FLI1-derived peptide for Ewing sarcoma.

Ewing sarcoma is an aggressive malignancy characterized by the EWS-FLI1 fusion oncoprotein, which remains challenging to target because of its intrinsically disordered nature and lack of intrinsic enzymatic activity. EWSPEP, a peptide derived from the EWS-FLI1 fusion junction region, has previously been investigated in chemically synthesized form; however, its recombinant production has not been reported. In this study, recombinant EWSPEP (rEWSPEP) was expressed in Escherichia coli BL21 Star (DE3) using the pET-45b expression system and purified by Ni2+ affinity chromatography, yielding approx

Synovial sarcoma, histologically mimicking primitive neuroectodermal tumor/Ewing's sarcoma at distant sites.

We report a case of synovial sarcoma (SS) showing unusual histology at distant sites. A 47-year-old man was aware of a tumor on the sole of his left foot. After preoperative chemotherapy with a diagnosis of SS, wide excision was performed. During postoperative chemotherapy, multiple tumorous lesions developed in the bone (including the whole spine) and both lungs. The patient died 1 year later. Histologically, the excised tumor of the foot showed a biphasic cellular pattern typical of SS, whereas at autopsy the bone and lung lesions were composed only of undifferentiated small round cells with

Primary Ewing's sarcoma/primitive neuroectodermal tumor (ES/PNET) of the penis: a report of an extraordinarily unusual site and a literature review of extraskeletal Ewing's sarcoma.

Ewing's sarcomas/primitive neuroectodermal tumors (ES/PNETs) arise from a multipotent progenitor cell and are considered to be of neuroectodermal derivation. Most tumors commonly arise in the skeletal system, which are the classic ES/PNET and occasionally occur in the soft tissue of extraskeletal sites, which are named extraskeletal Ewing's sarcomas (EES/PNET). This study reports a case of a 28-year-old man with primary EES/PNET of the penis.

Pathogenic Germline PALB2 and RAD50 Variants in Patients With Relapsed Ewing Sarcoma.

Approximately 10% of patients with Ewing sarcoma (EwS) have pathogenic germline variants. Here, we report two cases: first, a novel germline pathogenic variant in partner and localizer of BRCA2 (PALB2) in a patient with a late EwS relapse. Its impact on homologous recombination is demonstrated, and breast cancer risk is discussed. Second, we report a pathogenic germline variant in RAD50 in a patient with a rare prostate primary EwS and later concurrent relapsed EwS and therapy-related acute myeloid leukemia (tAML) with two rare KMT2A translocations. Understanding rare pathogenic germline varia

Evaluation of HDAC8 as a Druggable Target in STAG2-Mutant Ewing Sarcoma.

Ewing sarcoma is an aggressive pediatric cancer for which no targeted therapies have been approved. This disease is driven by EWSR1::ETS family fusions that, like most fusion transcription factors, have proven difficult to target directly. Here, we focus on identifying actionable dependencies within the high-risk STAG2-mutant (STAG2-mut) disease subtype of Ewing sarcoma. In STAG2-mut Ewing sarcoma, STAG1 knockout is synthetically lethal but is not readily druggable. Using the Cancer Dependency Map (DepMap), we identify histone deacetylase 8 (HDAC8) as an enriched vulnerability in this molecula

Development of single chain antibodies for targeting Ewing sarcoma surface marker CD99.

Recombinant antibodies have become an important class of therapeutic agents in cancer treatment. CD99, a transmembrane glycoprotein, is upregulated in several malignancies, including Ewing sarcoma (ES). The current therapy for ES comes with significant toxicity and a substantial risk of life-threatening late complications. Hence, there is a growing need for new and more effective treatments. Thus, we developed CD99-targeting single-chain antibodies by isolating the variable regions from a hybridoma-derived anti-CD99 antibody and engineering two scFvs with distinct linkers. Both scFvs were expr

Proximal Humerus Ewing Sarcoma: Technical Challenges and Strategies for Bone Defect Reconstruction.

INTRODUCTION: The proximal humerus is the most common site for primary malignant tumors of the upper limb, with chondrosarcoma, Ewing sarcoma, and osteosarcoma being the most frequent malignancies. Conservative treatment presents significant challenges regarding oncological resection due to the complex neurovascular anatomy and subsequent bone defect reconstruction. CASE REPORT: We report the case of a 29-year-old North African male managed for Ewing sarcoma of the proximal humerus. Locoregional staging showed tumor extension to the triceps, but no involvement of the deltoid, rotator cuff, or…

Spontaneous Haemothorax as Initial Presentation of Pleural Ewing Sarcoma: A Case Report.

This case report discusses a middle-aged patient with metastatic pleuro-pulmonary Ewing sarcoma, with the rare initial presentation of spontaneous haemothorax. The patient had a prior history of Ewing sarcoma of the pelvis in young age, which had been treated with chemoradiotherapy with remission. There was a 2-week history of right sided chest pain and shortness of breath, and a chest x-ray showed complete opacification of the right hemithorax, with subsequent CT arising suspicion of a heterogeneous soft tissue pleural lesion with fluid in the pleural cavity. While awaiting further treatment

[18F]FDG PET/CT in a Rare Case of Pancreatic Ewing Sarcoma.

Extraosseous Ewing sarcoma (EES) is a rare malignant tumor originating from soft tissue, comprising approximately 20% of all Ewing sarcoma cases. EES commonly involves the trunk or extremities, with pancreatic involvement being extremely uncommon. Because of its rarity, EES of the pancreas may be misdiagnosed initially and requires a multimodality approach. We present the case of a 25-y-old woman who was initially treated for acute pancreatitis. However, her symptoms persisted, and a [18F]FDG PET/CT was conducted for evaluation, the results of which suggested malignancy, which was later confir

Ewing sarcoma extraosseous brain metastasis presenting with an intracranial hemorrhage: illustrative case.

BACKGROUND: Ewing sarcoma is a rare malignant tumor that primarily affects children and young adults, commonly metastasizing to the lungs, bone, and bone marrow. CNS involvement is extremely rare, especially with the primary presentation of intracranial hemorrhage. Current protocols do not include routine neuroimaging, potentially delaying the detection of brain metastasis. Additionally, Ewing sarcoma patients are at risk of developing chemotherapy-induced thrombocytopenia that can significantly increase the possibility of intracranial hemorrhage. OBSERVATIONS: A 19-year-old male with metasta

Lung and bone metastases patterns in Ewing sarcoma: Chemotherapy improves overall survival.

Ewing sarcoma (ES) is a small round cell malignancy, mainly in the bone tissue, followed by the soft tissue. Lung metastases (LM) and bone metastases (BM) are the most common types of metastases. From 2010 to 2018, the Surveillance, Epidemiology, and End Results database diagnosed 242 cases of ES with LM, 186 cases of ES with BM, and 74 cases of ES with LM and BM. Univariate and multivariate logistic regression analyses were used to determine the risk factors for LM and/or BM, and Kaplan-Meier curves and Cox regression analysis were used to determine the prognostic factors for LM and/or BM. Combinatorial macrophage induced innate immunotherapy against Ewing sarcoma: Turning "Two Keys" simultaneously.

BACKGROUND: Macrophages play important roles in phagocytosing tumor cells. However, tumors escape macrophage phagocytosis in part through the expression of anti-phagocytic signals, most commonly CD47. In Ewing sarcoma (ES), we found that tumor cells utilize dual mechanisms to evade macrophage clearance by simultaneously over-expressing CD47 and down-regulating cell surface calreticulin (csCRT), the pro-phagocytic signal. Here, we investigate the combination of a CD47 blockade (magrolimab, MAG) to inhibit the anti-phagocytic signal and a chemotherapy regimen (doxorubicin, DOX) to enhance the pr
