Componentizing autonomous underwater vehicles by physical-running algorithms.

Autonomous underwater vehicles (AUV) constitute a specific type of cyber-physical system that utilize electronic, mechanical, and software components. A component-based approach can address the development complexities of these systems through composable and reusable components and their integration, simplifying the development process and contributing to a more systematic, disciplined, and measurable engineering approach. In this article, we propose an architecture to design and describe the optimal performance of components for an AUV engineering process. The architecture involves a computin

Genetic Predisposition to Autoimmune Diabetes Mellitus.

Type 1 diabetes mellitus (T1DM) is a multifactorial disease wherein a genetic predisposition, upon exposure to environmental factors, triggers seroconversion and subsequent beta-cell destruction. The global incidence of T1DM has risen in recent decades, underscoring the significant role environmental factors play in actuating inherent genetic risk. However, the absence of an identifiable unique triggering factor complicates the identification of risk groups. Advances in bioinformatics and epigenetic research are opening new opportunities for early diagnosis, prevention, and novel therapies for

Diagnosis and Management of Hereditary Phaeochromocytoma and Paraganglioma.

About 30% of phaeochromocytomas or paragangliomas are genetic. Whilst some individuals will have clinical features or a family history of inherited cancer syndrome such as neurofibromatosis type 1 (NF1) or multiple endocrine neoplasia 2 (MEN2), the majority will present as an isolated case. To date, 14 genes have been described in which pathogenic mutations have been demonstrated to cause paraganglioma or phaeochromocytoma . Many cases with a pathogenic mutation may be at risk of developing further tumours. Therefore, identification of genetic cases is important in the long-term management of

Unsupervised machine learning algorithms identify expected haemorrhage relationships but define unexplained coagulation profiles mapping to thrombotic phenotypes in hereditary haemorrhagic telangiectasia.

Hereditary haemorrhagic telangiectasia (HHT) can result in challenging anaemia and thrombosis phenotypes. Clinical presentations of HHT vary for relatives with identical casual mutations, suggesting other factors may modify severity. To examine objectively, we developed unsupervised machine learning algorithms to test whether haematological data at presentation could be categorised into sub-groupings and fitted to known biological factors. With ethical approval, we examined 10 complete blood count (CBC) variables, four iron index variables, four coagulation variables and eight iron/coagulation

Gender Disparity between Absolute versus Relative Size of Condylar Chondral Defects: An MRI Analysis.

Surgical repair of articular cartilage defects in the knee currently utilizes surgical algorithms based on absolute defect size. These algorithms, which have not been validated, are currently utilized not only by surgeons but also by insurance carriers for justification of reimbursement policy. However, current algorithms do not account for morphological differences between individual patients and defect size relative to condylar dimension. We hypothesized that a significant difference in relative defect size compared with condylar dimension may exist between individuals. A 3T magnetic resonan

GAN-GA: A Deep-Evolutionary Approach for Synthesizing Functionally Similar Heat Shock Proteins.

De novo protein design has emerged as a transformative approach for generating functional proteins without relying on naturally occurring templates, offering vast potential in biomedical and industrial applications. The integration of generative deep learning and evolutionary computation has opened new avenues in synthetic protein engineering. In this study, a hybrid framework, termed GAN-GA (Generative Adversarial Networks + Genetic Algorithm), has been proposed to address the limitations of traditional sequence based generation methods. Specifically, Wasserstein GANs have been employed to pr

Genetic and clinical factors associated with the metabolism of valproic acid in post-neurosurgery patients.

Valproic acid (VPA) is commonly used to treat epilepsy and bipolar disorder, requiring therapeutic concentrations of 50-100 mg/L to balance efficacy and safety. Despite known clinical and genetic factors influencing VPA metabolism, integrated predictive models are limited. This study analyzed VPA concentrations in 497 samples from 275 Chinese participants, examining 23 clinical variables and 24 genetic SNPs. Participants were grouped by SNP profiles, with one cluster showing significantly elevated VPA levels. Key factors influencing VPA concentrations included genetic variants (e.g., rs1223371

Genotypes versus phenotypes: The potential paradigm shift in the diagnosis and management of pediatric neoplasms.

The gold standard of cancer diagnosis has long been based on histological characteristics. With the rapid advancement of genetic medicine, such standard algorithm of diagnostic approach is facing a challenge. The genetic findings have been changed from being a "supporting character" into the role of a "main character". More and more disease diagnosis and classification has to be defined by genetic basis. In this article, we focus on the challenges in the field of pediatric oncology. We cited 2 scenarios where genetic information plays a pivotal role in identifying the underlying pathology. The

Genetics of chronic post-surgical pain: a crucial step toward personal pain medicine.

PURPOSE: Most patients who undergo surgery or experience a traumatic injury suffer from acute pain that subsides once tissues heal. Nevertheless, the pain remains in 15-30% of patients, sometimes for life, and this chronic post-surgical pain (CPSP) can result in suffering, depression, anxiety, sleep disturbance, physical incapacitation, and an economic burden. The incorporation of genetic knowledge is expected to lead to the development of more effective means to prevent and manage CPSP using tools of personalized pain medicine. The purpose of this review article is to provide an update on the

Frequency Band Analysis of Multiple Stationary Time Series.

The frequency-domain properties of biomedical signals offer valuable insights into health and functioning of underlying physiological systems. The power spectrum, which characterizes these properties, is often summarized by partitioning frequencies into standard bands and averaging power within bands. These summary measures are regularly used for analysis in practice, but are not guaranteed to optimally retain differences in power spectra across signals from different participants. We propose a data-adaptive method for identifying frequency band summary measures that preserve spectral variabil

Computational Design of Drugs for Epilepsy using a Novel Guided Evolutionary Algorithm for Enhanced Blood Brain Barrier Permeability.

INTRODUCTION: Epilepsy is a common disorder of the Central Nervous System (CNS). The rational design of small-molecule drugs for disorders of the CNS is a difficult process because the majority of small molecules are unable to cross the Blood-Brain-Barrier. An efficient method for the design of inhibitors that have high permeability through the Blood-Brain-Barrier has the potential for application in drug design for CNS disorders such as Addiction, Alzheimer's disease, Bipolar disorder, Depression, Epilepsy, Gliomas, and Tuberculous meningitis. METHODS: Supervised learning was used to model t

Structural Design and Optimisation of a Semi-Direct-Drive Master Manipulator.

BACKGROUND: In recent years, laparoscopic surgical robots have rapidly developed. However, research on master manipulators remains relatively limited. Existing designs are often characterised by structural complexity and inadequate force feedback capabilities. METHODS: A semi-direct-drive serial 8-degree of freedom force-feedback master manipulator was developed. A global fluctuation performance index based on mean absolute deviation and a minimum gravity torque index were proposed, and structural optimisation was performed using a multi-objective genetic algorithm. A spring-cable gravity com

Cost-minimization analysis of sequential genetic testing versus targeted next-generation sequencing gene panels in patients with pheochromocytoma and paraganglioma.

INTRODUCTION: Pheochromocytomas and paragangliomas (PPGLs) are highly heritable tumours, with up to 40% of cases carrying germline variants. Current guidelines recommend genetic testing for all patients with PPGLs. Next-generation sequencing (NGS) enables accurate, fast, and inexpensive genetic testing. This study aimed to compare the costs related to PPGL genetic testing between the sequential testing using the decisional algorithm proposed in the 2014 Endocrine Society guidelines and targeted NGS gene panels. METHODS: Patients with proven PPGLs were enrolled. A gene list covering 17 suscept
