Spacecraft Maximum Allowable Concentrations for Hydrogen Fluoride.

BACKGROUND: Spacecraft maximum allowable concentrations (SMACs) provide guidance on allowable chemical exposures for nominal and emergency situations aboard spacecraft. SMACs are set to mitigate or preclude potential crew health effects and performance degradation. Hydrogen fluoride (HF) gas is highly irritating. Inhaled HF produces irritation primarily in the upper respiratory tract. HF is not routinely present in spacecraft atmospheres. However, it can be produced in spacecraft due to overheating or combustion events involving materials containing fluorinated organics.METHODS: Toxicological …

Hydrogen sulfide in ageing, longevity and disease.

Hydrogen sulfide (H2S) modulates many biological processes, including ageing. Initially considered a hazardous toxic gas, it is now recognised that H2S is produced endogenously across taxa and is a key mediator of processes that promote longevity and improve late-life health. In this review, we consider the key developments in our understanding of this gaseous signalling molecule in the context of health and disease, discuss potential mechanisms through which H2S can influence processes central to ageing and highlight the emergence of novel H2S-based therapeutics. We also consider the major ch…

Acute myocardial injury following hydrogen sulfide poisoning.

BACKGROUND: Hydrogen sulfide poisoning can cause severe myocardial injury, but the damage is subtle and can be easily misdiagnosed. This report presents the dynamic observation of myocardial injury associated with hydrogen sulfide poisoning. CASE REPORT: Two young men presented with symptoms of "lightning-like" death immediately after entering a tank. They were found and rescued in 20 min at a time when they were already in a coma. Case 1 had no spontaneous breathing and pulse, while case 2 had spontaneous breathing and a pulse. Upon transfer to a local hospital, case 1 received continuous ca…

Hydrogen sulfide and vascular regulation - An update.

BACKGROUND: Hydrogen sulfide (H2S) is considered to be the third gasotransmitter after carbon monoxide (CO) and nitric oxide (NO). It plays an important role in the regulation of vascular homeostasis. Vascular remodeling have has proved to be related to the impaired H2S generation. AIM OF REVIEW: This study aimed to summarize and discuss current data about the function of H2S in vascular physiology and pathophysiology as well as the underlying mechanisms. KEY SCIENTIFIC CONCEPTS OF REVIEW: Endogenous hydrogen sulfide (H2S) as a third gasotransmitter is primarily generated by the enzymatic pa…

Diet and Hydrogen Sulfide Production in Mammals.

Significance: In recent times, it has emerged that some dietary sulfur compounds can act on mammalian cell signaling systems via their propensity to release hydrogen sulfide (H2S). H2S plays important biochemical and physiological roles in the heart, gastrointestinal tract, brain, kidney, and immune systems of mammals. Reduced levels of H2S in cells and tissues correlate with a spectrum of pathophysiological conditions, including heart disease, diabetes, obesity, and altered immune function. Recent Advances: In the last decade, researchers have now begun to explore the mechanisms by which diet…

Data-Driven Identification of Hydrogen Sulfide Scavengers.

Hydrogen sulfide (H2 S) is an important signaling molecule whose up- and down-regulation have specific biological consequences. Although significant advances in H2 S up-regulation, by the development of H2 S donors, have been achieved in recent years, precise H2 S down-regulation is still challenging. The lack of potent/specific inhibitors for H2 S-producing enzymes contributes to this problem. We expect the development of H2 S scavengers is an alternative approach to address this problem. Since chemical sensors and scavengers of H2 S share the same criteria, we constructed a H2 S sensor datab…

Hydrogen sulfide signaling in mitochondria and disease.

Hydrogen sulfide can signal through 3 distinct mechanisms: 1) reduction and/or direct binding of metalloprotein heme centers, 2) serving as a potent antioxidant through reactive oxygen species/reactive nitrogen species scavenging, or 3) post-translational modification of proteins by addition of a thiol (-SH) group onto reactive cysteine residues: a process known as persulfidation. Below toxic levels, hydrogen sulfide promotes mitochondrial biogenesis and function, thereby conferring protection against cellular stress. For these reasons, increases in hydrogen sulfide and hydrogen sulfide-produc…

Autonomous Temporal Control of Hydrogen Sulfide Delivery.

Hydrogen sulfide (H2S) is dichotomous in nature as it is lethal at higher concentrations, but at lower concentrations it shows a more cytoprotective nature. Due to its involvement in many physiological processes, it has recently risen to significance as a gasotransmitter alongside carbon monoxide (CO) and nitric oxide (NO). Experimentation using H2S is thus important to generate in vitro and in vivo models for this ubiquitous gasotransmitter. However, laboratory studies involving H2S are challenging due to the special handling conditions required to work with such a toxic gas. The use of chemi…

Esterase-sensitive trithiane-based hydrogen sulfide donors.

1,3,5-Trithiane functionalized with esterase-sensitive ester groups on the methylene linkers was developed as a class of enzymatic hydrolysis-based hydrogen sulfide (H2S) donors. The amount of H2S released from the donors was dependent on the number of ester bonds. The donors release H2S in a controllable manner in the presence of an enzyme.

Effects of intramolecular hydrogen bonds on lipophilicity.

Intramolecular hydrogen bonds (IMHBs) affect the lipophilicity and physicochemical properties of small organic molecules and drug candidates. Considering IMHBs in drug design is an important strategy for improving the lipophilicity and physicochemical properties of drug candidates. However, how IMHBs affect the lipophilicity and partition coefficients are not well understood. Based on the relationship between the effect of IMHBs on the experimental partition coefficients and the H-bonding capabilities of H-bond forming atoms, we explore how IMHBs affect water/n-octanol partition coefficients, …

Hydrogen-Deuterium Exchange Profiles of Polyubiquitin Fibrils.

Ubiquitin and its polymeric forms are conjugated to intracellular proteins to regulate diverse intracellular processes. Intriguingly, polyub … fibrils has been reported so far. Here, we identify the core of M1-linked diubiquitin fibrils from hydrogen-deuterium exchange experiments using solution nuclear magnetic resonance (NMR) spectroscopy. Intriguingly, intrinsically flexible regions became highly solvent-protected in the fibril structure. These results indicate that polyubiquitin fibrils are …

Hydrogen Peroxide: A Potential Wound Therapeutic Target?

Hydrogen peroxide (H2O2) is a topical antiseptic used in wound cleaning which kills pathogens through oxidation burst and local oxygen production. H2O2 has been reported to be a reactive biochemical molecule synthesized by various cells that influences biological behavior through multiple mechanisms: alterations of membrane potential, generation of new molecules, and changing intracellular redox balance, which results in activation or inactivation of different signaling transduction pathways. Contrary to the traditional viewpoint that H2O2 probably impairs tissue through its high oxidative pro…

Hydrogen sulfide in stroke: Protective or deleterious?

Hydrogen sulfide is believed to be a signalling molecule in the central nervous system. It is known to increase rapidly following an ischemic insult in experimental stroke. Is it protective or deleterious? This review surveys the relevant information available in the literature. It appears that there is no definitive answer to this question at present. Current evidence seems to suggest that the presence of H2S in the ischemic brain may either be deleterious or protective depending on its concentration, deleterious when high and protective when low. Therefore, it can be inferred that either an …

Biological chemistry of hydrogen sulfide and persulfides.

Hydrogen sulfide (H2S) has been traditionally considered to be a toxic molecule for mammals. However, it can be formed endogenously and exert physiological effects with potential health benefits. H2S can partition two-fold in biological membranes and traverse them rapidly, diffusing between compartments. H2S reactivity has similarities to that of thiols, although it is less nucleophilic than thiols and it can form different products. H2S can react with oxidants derived from the partial reduction of oxygen, but direct scavenging is unlikely to explain H2S protective actions. Important effects a…
