Spider Silk-Inspired High-Damping Liquid Crystal Elastomer Fibers Enabled by Semi-Interpenetrating Networks.

Inspired by the β-sheet nanocrystals in natural spider silk, we develop a high-damping polycrystalline-phase liquid crystal elastomer (LCE) fiber enabled by a semi-interpenetrating network. Continuous large-scale fabrication of this crosslinked system is realized using a unique channel-confinement strategy. By innovatively designing the end-group molecular structures of linear polymers, we precisely regulate the liquid-crystal phases within the semi-interpenetrating network fibers. Four distinct liquid-crystal phases are constructed, mimicking the β-sheet nanocrystals of spider silk to enable

Innovative silk protein peptide-modified soy protein isolate-xanthan gum films for food packaging.

A novel packaging material (SXS) composed of silk protein peptide (SPP), xanthan gum (XG), and soy protein isolate (SPI) was developed. The incorporation of SPP significantly enhanced the antioxidant activity of the SXS cling film. The aromatic amino acids in SPP and SPI absorbed ultraviolet light, improving the film's UV resistance. Molecular docking verified the interactions among SPI, XG and SPP, indicating multipoint binding that supports the proposed network-strengthening mechanism. The optimum amount of SPP is 6%, providing the best overall balance between UV barrier properties, mechanic

Silk Fibroin Biomimetic Membranes with Villus-Crypt Architecture for In Vitro Intestinal Epithelium Modeling.

Three-dimensional (3D) intestinal models require physiologically relevant microarchitectures and mechanically supportive matrices to accurately replicate epithelial behavior; however, most existing in vitro systems lack villus-crypt topography and do not provide the material cues needed to guide epithelial organization. In this work, we developed biomimetic silk fibroin (SF) membranes that reproduce the native villus-crypt structure using two complementary cross-linking and processing strategies: chemical cross-linking with BDDE to form soft hydrogels and ethanol-induced β-sheet formation to g

All-silk fibroin-based dual-drug delivery system for antibacterial and antioxidant wound repair.

Chronic and contaminated wounds are characterized by concurrent bacterial infection and excessive reactive oxygen species (ROS), necessitating biomaterials capable of coordinated antimicrobial and antioxidant regulation. In this study, we developed an all-silk fibroin-based dual-drug delivery system through macromolecular assembly of distinct silk micro/nanostructures. Berberine (BBR) was electrostatically immobilized onto negatively charged silk micro/nanofibers to enable rapid antibacterial release, while curcumin (CUR) was encapsulated within silk fibroin nanoparticles formed via PEG-induce

Electrospun silk fibroin nanofibers enriched with metallic nanoparticles and phenolic acids for antimicrobial applications.

Current trends in the development of the next generation of advanced wound dressings are directed toward the use of natural polymeric materials with additional properties such as antimicrobial function, controlled permeability, renewability, easy preparation, wound conformability and biocompatibility. Herein, fibrous hybrid organic-inorganic membranes of silk fibroin (SF) were prepared by electrospinning and were enriched with metal nanoparticles (Ag and Cu NPs) and phenolic compounds: caffeic, rosmarinic, and ellagic acids (CA, RA, EA, respectively) by using inert gas condensation sputtering

3D Cell Culture-Based Hybrid Bioanalytical Platform for Optical Imaging Utilizing Silk Fibroin Sponges.

We present an innovative bioanalytical hybrid platform designed for the preclinical evaluation of cellular characteristics. The system combines a three-dimensional (3D) cell culture grown on an artificial extracellular matrix with a chromatography-inspired array configuration. Sponges, made from the structural protein silk fibroin, serve both as a biomimetic extracellular matrix and as a stationary phase. Silk fibroin sponges were produced in-house using a multistep process involving removal of inherent sericin proteins from raw silk fibers, followed by dissolution and dialysis to purify the f

Highly stretchable polyacrylamide/silk fibroin double network self-adhesive hydrogel for flexible wearable sensors.

Silk fibroin (SF) is a natural biopolymer that is highly suitable for preparing biocompatible composite hydrogel strain sensors due to its excellent biocompatibility, amphiphilicity, and biodegradability. However, the excessive crystallization of β-sheet structures in SF limits the tensile and adhesive properties of pure SF hydrogels, severely restricting their application in hydrogel sensors. This study addresses this problem by introducing polyacrylamide (PAM) into the SF hydrogel system to construct a double-network structure. The flexible PAM chains regulate the β-sheet crystallization in

Toward Flexible Printed Electronics: A Spider-Silk-Inspired, Strong and Tough Thermoplastic Polyamide Elastomer.

The advancement of intelligent and compliant flexible electronic devices demands polymer substrate materials that exhibit both high strength and toughness, which are critical for the performance and durability of printed circuits. Inspired by the structure of spider silk, we designed and synthesized a series of naphthalene-containing thermoplastic polyamide elastomers (TPAE) in this work. The optimized TPAE-N40%-PEG25% exhibits an impressive hydrogen bonding density of up to 41.8%, leading to outstanding mechanical properties: a tensile strength of 37.3 MPa, an elongation at break of 470.8%, a

Silk-based double-sided magnetic microneedles for enhanced targeted retention in the gastrointestinal tract.

Gastrointestinal-retentive drug delivery devices represent a novel oral drug delivery approach capable of significantly enhancing patient compliance and improving local drug delivery efficiency compared to conventional oral formulations. However, developing gastrointestinal-retentive drug delivery devices with high retention capability based on compatible material systems presents significant challenges. Here, we introduce silk-based double-sided magnetic microneedles (SDMMs) designed to facilitate targeted oral drug delivery with precise local retention in the gastrointestinal tract. The SDMM

Silk fibroin hollow microneedle system for sustained transdermal administration of liraglutide: development and characterization.

Liraglutide demonstrates a glucose-dependent hypoglycemic mechanism with significant therapeutic potential for type 2 diabetes mellitus. This study developed a hollow microneedle system featuring dual drug loading mechanisms cavity encapsulated and matrix embedded to achieve sustained release of liraglutide. The cavity structure avoids direct contact between the drug and the silk fibroin shell, as well as the binding state transition caused by hydrophobic interactions. The shell has piercing mechanical properties and provides swelling diffusion channels. The pharmacokinetic data in SD rats sho

A sequential drug delivery system based on silk fibroin scaffold for effective cartilage repair.

Endogenous repair of cartilage defects is a preferential strategy for cartilage repair, but always hindered by insufficient early-stage cells and incomplete cell differentiation at later stages. For in-situ cartilage regeneration, it is crucial to develop a sequential drug release system capable of recruiting endogenous bone marrow mesenchymal stem cells (BMSCs) and promoting their chondrogenic differentiation. Herein, based on our long-term and fruitful research on silk fibroin (SF) porous scaffolds, a cell-free sequential drug delivery SF scaffold was developed. BMSCs affinity peptide PFSSTK

Biomineralized silk fibroin hydrogel with ultra-high strength for supporting bodyweight-bearing bone defects.

With four levels of formations and functional properties similar to biological tissues, protein hydrogel attracts great attention from researchers and clinical practitioners for broad applications. However, hydrogels with high water content usually exhibit poor mechanical properties, which limit their widespread application. In the study, a general and feasible approach was used to enhance the regenerated silk fibroin (RSF) hydrogel's mechanical strength by mimicking the human skeleton biomineralization process in vitro. The final mechanical strength was around 4.5 MPa, and the compressive mod

Tannic Acid-Enabled Antioxidant and Stretchable MXene/Silk Strain Sensors for Diving Training Healthcare.

MXene-based conductive hydrogels hold significant promise as epidermal sensors, yet their susceptibility to oxidation represents a formidable limitation. This study addresses this challenge by incorporating MXene into a tannic acid (TA) cross-linked silk fibroin matrix. The resulting conductive hydrogel (denoted as e-dive) exhibits favorable characteristics such as adjustable mechanical properties, self-healing capabilities (both mechanically and electrically), and strong underwater adhesion. The existence of a percolation network of MXene within the nanocomposites guarantees good electrical c

Dual-network DNA-silk fibroin hydrogels with controllable surface rigidity for regulating chondrogenic differentiation.

Osteoarthritis (OA) is a common joint disease known for cartilage degeneration, leading to a substantial burden on individuals and society due to its high disability rate. However, current clinical treatments for cartilage defects remain unsatisfactory due to the unclear mechanisms underlying cartilage regeneration. Tissue engineering hydrogels have emerged as an attractive approach in cartilage repair. Recent research studies have indicated that stem cells can sense the mechanical strength of hydrogels, thereby regulating their differentiation fate. In this study, we present the groundbreakin
