Spread of beta-lactam-resistant Pseudomonas aeruginosa in a cystic fibrosis clinic.

BACKGROUND: Pseudomonas aeruginosa colonisation of the airways of patients with cystic fibrosis (CF) is associated with considerable respiratory morbidity. Although segregation of colonised patients from non-colonised patients to prevent cross-infection has been recommended, there is little evidence that such cross-infection is widespread. We observed that a high proportion of children attending our CF clinic were colonised with P aeruginosa that was resistant to ceftazidime and other beta-lactam antibiotics. We used two genomic fingerprinting techniques to see whether this may have arisen fro

Establishing the detection threshold for Bacillus subtilis in a complex matrix using an inorganic fingerprint approach.

Methods for the detection and characterization of airborne biological warfare agents, such as bacteria, using their DNA or organic composition are fairly well developed. This approach is useful for identifying the type of bacterial strain once the organism has been isolated from the matrix sampled (e.g., dust particles) and can identify genetically related organisms, which might be helpful during a forensic investigation. However, this genetic signature will not reveal information related to the methods used to grow and weaponize the organism. Bacteria will take on an inorganic signature that

The epidemiology of tuberculosis in San Francisco. A population-based study using conventional and molecular methods.

BACKGROUND: The epidemiology of tuberculosis in urban populations is changing. Combining conventional epidemiologic techniques with DNA fingerprinting of Mycobacterium tuberculosis can improve the understanding of how tuberculosis is transmitted. METHODS: We used restriction-fragment-length polymorphism (RFLP) analysis to study M. tuberculosis isolates from all patients reported to the tuberculosis registry in San Francisco during 1991 and 1992. These results were interpreted along with clinical, demographic, and epidemiologic data. Patients infected with the same strains were identified acco…

On simple repetitive DNA sequences and complex diseases.

Simple repetitive DNA sequences are abundantly interspersed in eukaryote genomes and therefore useful in genome research and genetic fingerprinting in plants, fungi and animals, including man. Recently, simple repeats were also identified in some prokaryotic genomes. Hence the same probes can be applied for multilocus DNA fingerprinting in medically relevant bacteria. Simple repeats including composite dinucleotide microsatellites are differentially represented in different compartments of eukaryote genomes. Expanded triplet blocks in and around certain genes may, for example, cause so-called …

Optimization of STR locus enrichment for STR profiling of fragmented DNA.

DNA degradation is a major obstacle in gaining an accurate profile with standard DNA typing technology. Although alternative genotyping strategies such as mini-STRs and SNPs have proven to be more successful in profiling degraded DNA, these approaches also have limitations. Here, we show that locus enrichment by hybridization of degraded genomic DNA with an STR locus-specific biotinylated oligonucleotide is a powerful approach to overcome problems in STR typing of highly degraded DNA. An experimental investigation of factors affecting the efficiency of this method indicates that the choice of

From cold case to conviction: How advanced DNA technologies such as mtDNA sequencing connected two brutal homicides.

Cold case investigations involve revisiting case circumstances and prior evidentiary material. Novel or improved methods better suited to minute, mixed or degraded samples may provide new leads or higher evidential values. Here, we describe a reinvestigation started in 2016 of a 2004 murder case in which the body parts of a young woman were found in five double-bagged rubbish bags in the city of Amsterdam. Resampling and autosomal DNA-profiling of one of the rubbish bags knot areas resulted in a low LR for a man who had had sexual contact just before the victim went missing. This man became th

Derivation and characterization of the iPSC line IITGi001-B from primary human adult dermal fibroblasts.

Adult human dermal fibroblasts purchased from ATCC were reprogrammed using the Y4 combination of episomal plasmids expressing hOCT3/4, short hairpin RNA against p53, hSOX2, hKLF4, hL-MYC, and hLIN28 to generate induced pluripotent stem cells (iPSCs). A representative iPSC clone was extensively characterized to confirm the expression of pluripotency markers, normal karyotype, in vitro trilineage differentiation potential, and genetic identity using DNA fingerprinting. Further, the absence of the integration of episomal plasmids and mycoplasma contamination in the iPSC line was confirmed. The ge…

Isolation and characterization of a novel P-II class snake venom metalloproteinase from Trimeresurus stejnegeri.

Stejnitin, a novel class P-II snake venom metalloproteinase (SVMP) with a molecular weight of about 35kDa, was purified from Trimeresurus stejnegeri venom. The cDNA of stejnitin encoded a polypeptide of 295 amino acid residues which comprises a signal peptide, proprotein, metalloproteinase domain, spacer and disintegrin domain. The protein sequence deduced from cDNA was confirmed by peptide mass fingerprinting analysis. It is highly homologous to the members of subclass P-IIa SVMPs which comprises metalloproteinase and disintegrin together. Results from DNA fragmentation and flow cytometry ana

Two cases of subcutaneous Scedosporium apiospermum infection treated with voriconazole.

Scedosporium apiospermum is a mold that is increasingly being recognized as an opportunistic pathogen in immunocompromised patients, and treatment is complicated by intrinsic resistance to several antifungal agents. In our hospital, two cases of S. apiospermum infection occurring within 2 weeks were successfully treated with voriconazole. Since both patients were infected with an uncommon pathogen, a search for a common nosocomial source was performed. As environmental cultures yielded no S. apiospermum, and random amplified polymorphic DNA (RAPD) fingerprinting showed that the patients' strai

Neonatal pneumococcal sepsis in association with fatal maternal pneumococcal sepsis.

A live male infant was born at 37 weeks' gestation after a normal pregnancy to a 34-year-old mother. The baby developed bacteraemia with Streptococcus pneumoniae and recovered completely following treatment with antibiotics. The mother simultaneously developed bacteraemia with the same organism and died from septic shock. Blood culture isolates from mother and child were both serogroup 23F, and were shown to be identical by DNA fingerprinting. The literature reports rare cases of vaginal carriage and/or endometritis with this organism resulting in neonatal sepsis. Transmission to the neonate m…

STR analysis of human DNA recovered from bathwater and other water samples for forensic identification.

Human DNA is widely distributed in the environment and can be released into water through skin cells, sweat, and other biological materials. In forensic investigations involving drowning or suspected immersion events, bathwater represents an underexplored but potentially informative source of DNA evidence. This study evaluated the feasibility of recovering and analyzing human DNA from bathwater for short tandem repeat (STR) profiling. Bathwater samples were collected from 11 volunteers after 1, 2, 5, and 10 minutes of immersion, as well as from 12 forensic autopsy cases. DNA was concentrated u

Adhesive tape versus swabbing for touch DNA collection: A direct PCR approach with the VeriFiler Express Kit.

This study compares two collection techniques - swabbing and adhesive tape - for obtaining touch DNA samples suitable for direct typing with the VeriFiler® Express Kit. Touch DNA, defined as genetic material deposited on surfaces following human contact, plays an increasingly important role in forensic investigations by enabling the analysis of trace biological evidence for potential suspect identification. Samples were collected from two controlled surfaces: sterile glass and a mobile phone. A total of 100 samples were obtained using both methods and analyzed for DNA quantity and quality, spe

Conclusiveness of DNA Reports in Indian rape cases: procedural challenges and judicial caution.

DNA profiling is considered a scientifically reliable tool for identifying perpetrators and exonerating the innocent. However, its role in Indian rape cases is generally regarded as corroborative rather than conclusive. Judicial scepticism stems from the likelihood of procedural deviations in sample collection, preservation, and handling, along with a preference for traditional evidence such as eyewitness testimony. Inconsistent judicial interpretation, multiplicity of forensic protocols, judicial backlogs, inadequate infrastructure/workforce, and delay tactics compound the problem. Statutory
