Optimized aptamer-based next generation biosensor for the ultra-sensitive determination of SARS-CoV-2 S1 protein in saliva samples.

COVID-19 is an infectious disease caused by the SARS-CoV-2 virus, which rapidly spread worldwide and resulted in a pandemic. Efficient and sensitive detection techniques have been devised since the onset of the epidemic and continue to be improved at present. Due to the crucial role of the SARS-CoV-2 S1 protein in facilitating the virus's entry into cells, efforts in detection and treatment have primarily centered upon this protein. In this study, a rapid, ultrasensitive, disposable, easy-to-use, cost-effective next generation biosensor based on optimized aptamer (Optimer, OPT) was developed b

Development and Analytical Evaluation of a Point-of-Care Electrochemical Biosensor for Rapid and Accurate SARS-CoV-2 Detection.

The COVID-19 pandemic has underscored the critical need for rapid and accurate screening and diagnostic methods for potential respiratory viruses. Existing COVID-19 diagnostic approaches face limitations either in terms of turnaround time or accuracy. In this study, we present an electrochemical biosensor that offers nearly instantaneous and precise SARS-CoV-2 detection, suitable for point-of-care and environmental monitoring applications. The biosensor employs a stapled hACE-2 N-terminal alpha helix peptide to functionalize an in situ grown polypyrrole conductive polymer on a nitrocellulose m

Bioimpedance spectroscopy for breast cancer-related lymphedema assessment: clinical practice guidelines.

PURPOSE: Breast cancer-related lymphedema (BCRL) represents a significant concern for patients following breast cancer treatment, and assessment for BCRL represents a key component of survivorship efforts. Growing data has demonstrated the benefits of early detection and treatment of BCRL. Traditional diagnostic modalities are less able to detect reversible subclinical BCRL while newer techniques such as bioimpedance spectroscopy (BIS) have shown the ability to detect subclinical BCRL, allowing for early intervention and low rates of chronic BCRL with level I evidence. We present updated clini

Classification of Hydration in Clinical Conditions: Indirect and Direct Approaches Using Bioimpedance.

Although the need to assess hydration is well recognized, laboratory tests and clinical impressions are impractical and lack sensitivity, respectively, to be clinically meaningful. Different approaches use bioelectrical impedance measurements to overcome some of these limitations and aid in the classification of hydration status. One indirect approach utilizes single or multiple frequency bioimpedance in regression equations and theoretical models, respectively, with anthropometric measurements to predict fluid volumes (bioelectrical impedance spectroscopy-BIS) and estimate fluid overload base

Label-free electrochemical impedance immunosensor based on modified screen-printed gold electrodes for the diagnosis of canine visceral leishmaniasis.

Leishmaniasis is a disease with high impact on public health in many countries. Visceral leishmaniasis (VL) is a vectorial zoonosis, with dogs as primary reservoirs in the domestic environment. VL presents high morbidity, mortality and importance in epidemiology in the American continent. In the present study, the first label-free electrochemical impedance immunosensor using screen-printed electrodes (SPEs) for the detection of anti-Leishmania infantum antibodies was developed. The soluble antigens of L. infantum were immobilized on an SPE by a 3-mercaptopropionic acid monolayer. Electrochemic

Novel electrochemical sensing platform for ultrasensitive detection of cardiac troponin I based on aptamer-MoS2 nanoconjugates.

Cardiac troponin I (cTnI) is a specific and sensitive biomarker for the early diagnosis of acute myocardial infarction and for the subsequent clinical treatments. In this work, novel electrochemical sensing platform for sensing of cTnI based on aptamer-MoS2 nanoconjugates was proposed. For comparison, core-shell Au@SiO2@Au nanoparticles were also used for sensing of cTnI. The sensing schemes and electrochemical responses of the proposed sensors were investigated by electrochemical impedance spectroscopy (EIS) in 5.0 mM K3[Fe(CN)6]/K4[Fe(CN)6] (1:1) solution containing 0.1 M KCl, respectively.

A novel three-dimensional biosensor based on aluminum oxide: application for early-stage detection of human interleukin-10.

Immunosensors based on electrolyte-oxide-semiconductors (EOS) have been extensively researched over the last few decades. By electrochemical impedance spectroscopy (EIS) the specific molecular biorecognition of the antibody-antigen (Ab-Ag) can be detected providing an alternative quantitative system to immunoassay techniques. The electrochemical variations from a fabricated immunosensor can provide quantitative values for the analyte of interest at reduced costs and analysis time. In this context, a novel EOS substrate based on aluminum oxide (Al2O3) grown by atomic layer deposition on silicon

The effect of the hydrophilic spacer length on the functionality of a mercury-supported tethered bilayer lipid membrane.

A biomimetic membrane consisting of a thiolipid monolayer tethered to a mercury electrode, with a dioleoylphosphatidylcholine (DOPC) monolayer on top of it, was fabricated. The thiolipid, referred to as DPOL, consisted of an octaethyleneoxy (OEO) chain terminated at one end with a lipoic acid residue and covalently linked at the other end to two phytanyl chains. The functionality of this biomimetic membrane, referred to as a tethered bilayer lipid membrane (tBLM), was tested by incorporating gramicidin and alamethicin and verifying their ion channel activity. Advantages and drawbacks with resp

Impedance-based cell culture platform to assess light-induced stress changes with antagonist drugs using retinal cells.

This Article describes an unprecedented, simple, and real-time in vitro analytical tool to measure the luminous effect on the time responses function of retinal ganglion cells (RGC-5) by electric cell substrate impedance sensing (ECIS) system. The ECIS system was used for the continuous measurement of different color light-induced effects on the response of cells that exposed to protective drugs. The measurement suggests that the association of photo-oxidative stress was mediated by reactive oxygen species (ROS), which plays a critical role that leads to cell stress, damages, and retinopathy,

Impedance spectroscopy and conductometric biosensing for probing catalase reaction with cyanide as ligand and inhibitor.

In this work, a new biosensor was prepared through immobilization of bovine liver catalase in a photoreticulated poly (vinyl alcohol) membrane at the surface of a conductometric transducer. This biosensor was used to study the kinetics of catalase-H(2)0(2) reaction and its inhibition by cyanide. Immobilized catalase exhibited a Michaelis-Menten behaviour at low H(2)0(2) concentrations (<100mM) with apparent constant K(M)(app)=84±3mM and maximal initial velocity V(M)(app)=13.4μS min(-1). Inhibition by cyanide was found to be non-competitive and inhibition binding constant K(i) was 13.9±0.3μM. T

Femtosecond Optical Kerr Effect in Alzheimer's Brain Tissue.

The optical Kerr effect (OKE) spectroscopy is a powerful technique to study ultrafast dynamics of a material's response to an optical field. In this study, we investigated OKE in Alzheimer's disease (AD) and normal human brain tissues from the hippocampus and Brodmann areas 9 and 17. The Kerr signals from both AD and normal samples exhibited a distinct double-peak profile, corresponding to the electronic and plasma mechanisms. Analysis of plasma relaxation and dielectric response times revealed that conductivity in the AD hippocampus was approximately 46%-61% higher than in normal tissue, due

Surface-Enhanced Raman Spectra Promoted by a Finger Press in an All-Solid-State Flexible Energy Conversion and Storage Film.

Electrochemically up-regulated surface-enhanced Raman spectroscopy (E-SERS) effectively increases Raman signal intensities. However, the instrumental requirements and the conventional measurement conditions in an electrolyte cell have hampered its application in fast and on-site detection. To circumvent the inconveniences of E-SERS, we propose a self-energizing substrate that provides electrical potential by converting film deformation from a finger press into stored electrical energy. The substrate combines an energy conversion film and a SERS-active Ag nanowire layer. A composite film prepar

Hemocompatibility of stent materials: alterations in electrical parameters of erythrocyte membranes.

BACKGROUND: It is presently unknown if stents used in the correction of artery stenosis are fully hemocompatible or if their implantation causes alterations at the level of the plasma membrane in red blood cells. METHODS: We addressed this important issue by measuring the passive electrical properties of the erythrocyte membrane before and after stent insertion by means of dielectric relaxation spectroscopy in the radiowave frequency range in a series of patients who were undergoing standard surgical treatment of arterial disease. RESULTS: Our findings provide evidence that full hemocompatib

Simple and rapid mercury ion selective electrode based on 1-undecanethiol assembled Au substrate and its recognition mechanism.

A simple and rapid mercury ion selective electrode based on 1-undecanethiol (1-UDT) assembled Au substrate (Au/1-UDT) has been well constructed. 1-UDT was for the purpose of generating self-assembled monolayer on gold surface to recognize Hg2+ in aqueous solution, which had a working concentration range of 1.0×10-8-1.0×10-4molL-1, with a Nernst response slope of 28.83±0.4mV/-pC, a detection limit of 4.5×10-9molL-1, and a good selectivity over the other tested cations. Also, the Au/1-UDT possessed good reproducibility, stability, and short response time. The recovery obtained for the determinat

Solvent structure and dynamics of ion pairs between a ruthenium photocatalyst and a countercation studied with infrared spectroscopy.

The strength of the ionic interaction between a counterion and photosensitizer can alter photocatalyst excited state stabilities and redox potentials, ultimately determining catalytic efficiency. Counterion identities are known to directly influence ionic interaction strength; however, recent literature indicates that the structure of the solvation shell around photosensitizer-counterion pairs can play a similarly important role in dictating photocatalytic properties. As such, the capacity to discriminate between different ionic solvation structures is imperative for understanding and optimizi

Non-Invasive Skin Cancer Diagnosis by Electrical Impedance Spectroscopy: Biophysics, Devices, Clinical Evidence, and Future Directions.

Skin cancer represents a growing global health burden. Current diagnostic pathways combine clinical examination and dermoscopy with histopathological confirmation; however, overlap between benign and malignant lesions can create diagnostic uncertainty and lead to potentially avoidable biopsies. Electrical impedance spectroscopy (EIS) has emerged as a non-invasive technique with potential for portable and cost-efficient implementation that quantifies the dielectric contrast between malignant and healthy tissue, providing objective information that may support clinical decision-making.
