Ensemble force changes that result from human cardiac myosin mutations and a small-molecule effector.

Cardiomyopathies due to mutations in human β-cardiac myosin are a significant cause of heart failure, sudden death, and arrhythmia. To understand the underlying molecular basis of changes in the contractile system's force production due to such mutations and search for potential drugs that restore force generation, an in vitro assay is necessary to evaluate cardiac myosin's ensemble force using purified proteins. Here, we characterize the ensemble force of human α- and β-cardiac myosin isoforms and those of β-cardiac myosins carrying left ventricular non-compaction (M531R) and dilated cardiomy

Functional changes in spinal circuitry in essential tremor revealed with analysis of intramuscle synergies.

Essential tremor (ET) is one of the most common movement disorders and is marked by centrally generated oscillations that interfere with stable hand function. The contribution of spinal circuitry to impaired movement stability remains largely unexplored. In this study, we assessed intramuscle synergies as a potential biomarker of spinal deficits during accurate cyclic force production in individuals with ET and compared them with those in patients with Parkinson's disease (PD), age-matched controls (AMC), and young adult controls. Motor unit activity was extracted through surface EMG decomposi

Musculoskeletal models determine the effect of a soft active exosuit on muscle activations and forces during lifting and lowering tasks.

Exosuits have the potential to mitigate musculoskeletal stress and prevent back injuries during industrial tasks. This study aimed to 1) validate the implementation of a soft active exosuit into a musculoskeletal model of the spine by comparing model predicted muscle activations versus corresponding surface EMG measurements, and 2) evaluate the effect of the exosuit on peak back and hip muscle forces. Fourteen healthy participants performed squat and stoop lift and lower tasks with boxes of 6 and 10 kg, with and without wearing a 2.7 kg soft active exosuit. Participant-specific musculoskeletal

Computational sensitivity analysis to identify muscles that can mechanically contribute to shoulder deformity following brachial plexus birth palsy.

PURPOSE: Two mechanisms, strength imbalance or impaired longitudinal muscle growth, potentially cause osseous and postural shoulder deformity in children with brachial plexus birth palsy. Our objective was to determine which muscles, via either deformity mechanism, were mechanically capable of producing forces that could promote shoulder deformity. METHODS: In an upper limb computational musculoskeletal model, we simulated strength imbalance by allowing each muscle crossing the shoulder to produce 30% of its maximum force. To simulate impaired longitudinal muscle growth, the functional length

On the Adaptation of Pelvic Motion by Applying 3-dimensional Guidance Forces Using TPAD.

Pelvic movement is important to human locomotion as the center of mass is located near the center of pelvis. Lateral pelvic motion plays a crucial role to shift the center of mass on the stance leg, while swinging the other leg and keeping the body balanced. In addition, vertical pelvic movement helps to reduce metabolic energy expenditure by exchanging potential and kinetic energy during the gait cycle. However, patient groups with cerebral palsy or stroke have excessive pelvic motion that leads to high energy expenditure. In addition, they have higher chances of falls as the center ofmass co

Stability of Nano and Micro Particle Suspensions in Electrolyte Solutions: A Comparative Classical Density Functional Theory and Measurement of Force and Structure in Confined Complex Fluid.

We compare theory to experiment to study the suitability of different classical density functional theory (DFT) strategies to predict interactions and stability in different-size colloid mixtures in an electrolyte solution. Atomic force microscopy (AFM) measures the interaction force between a spherical silica microcolloid and a flat silica substrate across a suspension of silica nanoparticles in a solution of sodium chloride. The calculated and measured oscillatory structural and repulsive electrostatic forces give insight on the stability of microcolloids in suspensions of nanoparticles rele

Nano-samples give higher brittle strength by the Griffith energy principle.

The purpose of this paper is to show that brittle test samples give a huge size effect that can take several different forms depending on the sample geometry, crack position and mode of force application. Sometimes crack equilibrium force depends on sample dimension d or d1/2 and sometimes the force is independent of area, for example in peel or lap joint cracking. This big size effect arises from the potential energy term in the conservation theory, not considered by Griffith but dominating certain cracks. These examples illustrate the fact that strength of a brittle material containing a cra

A cable driven robotic palpation system with contact force sensing based on cable tension observation.

BACKGROUND: Prostate Cancer screening based on manual palpation is subjective. Robotic palpation systems can objectively acquire stiffness conditions of the prostate. METHODS: A 2DoF cable driven robotic system for prostate palpation is proposed. An indirect method to estimate the contact force based on cable tension observation is presented. Kinematic models and a joint angle estimation method to determine the tip position of the probe are derived. Positioning accuracy was verified using an optical marker tracking system and by displacement measurement, respectively. The contact force estima

Tethering effects on first-passage variables of lattice random walks in linear and quadratic focal point potentials.

Diffusion in a confining potential offers a minimal setting to understand the interplay between random motion and deterministic forces driving a particle towards a focal point or potential minimum. In continuous space and time, two extensively studied examples are Brownian motion in a linear (V-shaped) or a quadratic (U-shaped) potential. The deterministic bias towards the minimum is represented, respectively, by a constant force for the former and by an elastic restoring force that increases proportionally with distance for the latter. Surprisingly, unlike Brownian walks, random walks under f

Determinant of Dynamics and Interfacial Forces in Ultraprecision Machining of Optical Freeform Surface through Simulation-Based Analysis.

This study delves into the intricacies of ultraprecision machining, particularly in the context of machining optical freeform surfaces using Diamond Turning Machines (DTMs). It underscores the dynamic relationship between toolpath generation, hydrostatic bearing in DTMs, and the machining process. Central to this research is the innovative introduction of Metal Matrix Composites (MMCs) to replace the traditional materials used in designing linear bearings. This strategic substitution aims to dynamically enhance both the accuracy and the quality of the machined optical freeform surfaces. The st

The Effect of Mechanical Vibration on Osteogenesis of Periodontal Ligament Stem Cells.

INTRODUCTION: Appropriate occlusal forces can prevent ankylosis after tooth replantation or transplantation. However, the "proper occlusal forces" on periodontal ligament (PDL) healing have not yet been defined due to insufficient in vitro studies and uncertain in vitro models. Herein, we presented a mechanical vibration device as an in vitro model to determine such favorable occlusal forces. METHODS: Human periodontal ligament stem cells (hPDLSCs) were exposed to mechanical vibration force with 4 frequencies (30, 90, 150, and 210 rpm). Cell viability and the expression of osteogenic differen

Recovery from acidosis is a robust trigger for loss of force in murine hypokalemic periodic paralysis.

Periodic paralysis is an ion channelopathy of skeletal muscle in which recurrent episodes of weakness or paralysis are caused by sustained depolarization of the resting potential and thus reduction of fiber excitability. Episodes are often triggered by environmental stresses, such as changes in extracellular K+, cooling, or exercise. Rest after vigorous exercise is the most common trigger for weakness in periodic paralysis, but the mechanism is unknown. Here, we use knock-in mutant mouse models of hypokalemic periodic paralysis (HypoKPP; NaV1.4-R669H or CaV1.1-R528H) and hyperkalemic periodic

The effect of the number of single door segments in the posterior cervical canal on the traction of the cervical 5 nerve root.

BACKGROUND: The posterior cervical canal plays a critical role in the protection and function of the cervical nerve roots, particularly the C5 nerve root. Variations in its anatomical structure, notably the number of single-door segments, may impact the traction forces experienced by the C5 nerve root during surgical interventions or pathological conditions. This study aims to quantify how the number of single-door segments affects traction forces on the C5 nerve root. METHODS: A biomechanical model was developed to simulate traction forces on the C5 nerve root based on varying anatomical con

Anisotropic nanorod-integrated magneto-mechano-electric cascade system for neural electrical stimulation.

Piezoelectric materials are now being considered as a potential treatment system for neural electrical stimulation. However, their therapeutic efficacy is limited by insufficient electrical outputs, which are generated by cellular forces and/or tissue motions. Herein, we constructed a high-efficiency magneto-mechano-electric cascade system with magnetic field (MF)-driven wireless electrical stimulation. The anisotropic Fe3O4 nanorod (RFO)-integrated poly(vinylidene fluoride-co-trifluoroethylene) copolymer (PVDF-TrFE) fibrous scaffold (PT-RFO) with enhanced piezoelectric β-phase content was pre

Elucidating transmission dynamics and host-parasite-vector relationships for rodent-borne Bartonella spp. in Madagascar.

Bartonella spp. are erythrocytic bacteria transmitted via arthropod vectors, which infect a broad range of vertebrate hosts, including humans. We investigated transmission dynamics and host-parasite-vector relationships for potentially zoonotic Bartonella spp. in invasive Rattus rattus hosts and associated arthropod ectoparasites in Madagascar. We identified five distinct species of Bartonella (B. elizabethae 1, B. elizabethae 2, B. phoceensis 1, B. rattimassiliensis 1, and B. tribocorum 1) infecting R. rattus rodents and their ectoparasites. We fit standard epidemiological models to species-s

Intramuscular Nanofat Injection Promotes Inflammation-Induced Gastrocnemius Regeneration in a Syngeneic Rat Sciatic Nerve Injury Model.

BACKGROUND: Mechanical emulsification of adipose tissue to concentrate protein and stromal cell components (ie, nanofat) has gained considerable interest in clinical practice. Although the regenerative potential of nanofat has largely been used in aesthetic applications, these effects have considerable potential in reconstruction as well. Here, the authors investigated the therapeutic properties of nanofat injected directly into the denervated gastrocnemius after a sciatic nerve injury in Lewis rats. METHODS: Muscle denervation was induced by transecting and immediately repairing the sciatic

Finite element analysis of stress in removable lower complete denture under vertical and oblique occlusal forces.

Frequent fractures of complete removable dentures during mastication in the anterior zone cannot be explained by bite forces in this area, because occlusal forces on the incisors are small. It appears that the mechanism of fracture must be from lateral mastication forces. The hypothesis of the study was that, under stable support of a complete removable denture on the foundation, its fracture in the anterior segment is possible due to stress generated under masticatory forces typical for denture wearers. This study analyzes stress distribution in removable complete dentures (designed in Exocad

Neural substrates associated with the acquisition of fine finger force control.

BACKGROUND: Precise finger force control is essential for performing everyday tasks such as writing, buttoning, and eating. While the neurophysiological basis of cognitive skill learning has been extensively studied, much less is known about the neural mechanisms supporting the acquisition of fine motor skills involving finger force control. This study aimed to investigate changes in cortical oscillatory activity and event-related potentials (ERPs) associated with the acquisition of fine finger force control. RESULTS: Eighteen right-handed healthy young adults practiced a visual target-matchi

Electro-Mechanical Ionic Channel Modeling for Uterine Contractions and Oxytocin Effect during Pregnancy.

Uterine contractions during normal pregnancy and preterm birth are an important physiological activity. Although the cause of preterm labor is usually unknown, preterm birth creates very serious health concerns in many cases. Therefore, understanding normal birth and predicting preterm birth can help both newborn babies and their families. In our previous work, we developed a multiscale dynamic electrophysiology model of uterine contractions. In this paper, we mainly focus on the cellular level and use electromyography (EMG) and cell force generation methods to construct a new ionic channel mo

Zinc Supplementation Sustains Diaphragm Contractility and Preserves SERCA2a Expression in Aged Female Rats with Type 2 Diabetes.

Diabetes mellitus (DM) is a chronic metabolic disease characterized by hyperglycemia, and the diaphragm-the primary respiratory muscle-is adversely affected by this diabetic process. The aim of this study is to investigate the effects of zinc sulfate (ZnSO4) treatment on diaphragm muscle contractile dynamics, calcium homeostasis, apoptosis, and fibrosis in an 18-month-old female Type 2 diabetic rat model. Thirty-two 18-month-old female Wistar rats were randomly divided into four groups: Control (CON), CON + ZnSO4, Diabetes Mellitus (DM), and DM + ZnSO4. The DM model was induced by a high-fat d
