In vitro sensitivity to 5-fluorocytosine of strains isolated from patients under treatment for chromomycosis.

In vitro 5-fluorocytosine sensitivity tests of strains were isolated from chromomycosis patients undergoing therapy. Tests were rountinely performed for 0.1, 1.0, 5.0 and 10.0 mug/ml 5-fluorocytosine concentrations. In two cases resistance could be demonstrated up to 60.0 and 100.0 mug/ml. Fungistatic activity of 5-fluorocytosine against the agents of chromomycosis was confirmed. Sensitivity tests are useful to detect resistance which may occur during treatment. They are also helpful for guiding the clinician in the establishment of new therapeutic schedules.

Comparative assessment of the broth microdilution and VITEK 2 systems for antifungal susceptibility testing of Candida auris (Candidozyma auris) Isolates.

BACKGROUND: Candidozyma auris is a multidrug resistant fungal pathogen classified by the World Health Organization (WHO) as a critical priority species. Broth microdilution (BMD) is the recommended reference method for antifungal susceptibility testing (AFST), with interpretation based on tentative breakpoints proposed by the Centers for Disease Control and Prevention (CDC) and, more recently, EUCAST clinical breakpoints published in 2025. However, automated systems such as VITEK 2 currently lack validated susceptibility interpretations for C. auris. This study aimed to evaluate the performanc

Safety Assessment of Lacticaseibacillus rhamnosus OF44, a Previously Isolated Human-origin Probiotic Candidate.

Lacticaseibacillus rhamnosus is one of the most extensively studied and well-characterized probiotic species. The strain L. rhamnosus OF44, originally isolated from a healthy female fecal sample, has shown particular promise for its ability to modulate the vaginal microenvironment through pH reduction and inflammation control. However, its comprehensive safety profile remains unclear. In this study, we systematically evaluated the safety of the previously identified strain L. rhamnosus OF44 through a series of standardized assessments. Genomic analysis (99.9% coverage) revealed the absence of

Discovery of Aromatic Meroterpenoids with Potent Anti-biofilm Activity and Genome-Guided Identification of Two Prenyltransferases.

Aspergillus fungi are renowned for producing various natural products, including meroterpenoids that show significant bioactivities. In this study, chemical investigation of a newly identified Aspergillus fungus led to the discovery of seven aromatic meroterpenoids, including two new ones designated as pantaromerotin (1) and pantanitrimerotin (2). Pantaromerotin is distinguished by containing a rare pyran ring formed from a prenyl unit, which are not observed in the structurally similar compound stocksiloate. In comparison, pantanitrimerotin harbors a rare cyanide group, which is not found in

In vitro characterization of GSK2556286 in combination with bedaquiline and pretomanid against Mycobacterium tuberculosis.

GSK2556286 (GSK286) is a novel drug candidate with a cholesterol-dependent mode of action that has shown promising in vivo potential to improve tuberculosis treatment when combined with bedaquiline (B) and pretomanid (Pa). This study explored the in vitro activity of GSK286 alone and in combination with B and Pa, using extracellular and intracellular dose-response time-kill assays against different Mycobacterium tuberculosis (Mtb) strains. GSK286 demonstrated media-specific activity, inhibiting Mtb growth only in cholesterol- and propionate-containing media, with slight activity against both e

Wild-type MIC distributions and epidemiological cut-off values for ibrexafungerp, manogepix, and rezafungin against WHO fungal priority pathogens.

OBJECTIVE: This study gathered public data regarding the in vitro activity of three novel antifungal agents, ibrexafungerp, manogepix, and rezafungin, against the fungi listed in the WHO fungal priority pathogens list. METHODS: Epidemiological cut-off values (ECVs/ECOFFs) were established for species with sufficient data (≥100 MIC values) and when distributions met further essential criteria. For pathogens in which data did not fulfil one or more essential criteria, tentative ECVs (T-ECVs/T-ECOFFs) were proposed. The values were determined by a comprehensive analysis that included visual insp

The Intricate Nonribosomal Assembly of a Potent Antifungal Lipopeptide from the Burkholderia cepacia Complex.

The Burkholderia cepacia complex (BCC) is a group of Gram-negative bacteria known for their pathogenicity to patients suffering from cystic fibrosis (CF). The BCC-belonging strain B. pyrrocinia BC11 (formerly B. cepacia BC11) produces AFC-BC11, a compound with strong activity against phytopathogenic fungi. In this contribution, we report on the unprecedented N-acyl-tetrapeptide structure and antifungal potency of this natural product. We further provide insights into central steps of its biosynthesis mediated by a nonclassical nonribosomal peptide synthesis machinery lacking condensation domai

Facile Synthesis of Oxazolidinones as Potential Antibacterial Agents.

An efficient microwave-assisted synthesis route for novel oxazolidinone analogues has been developed. The general synthesis of these compounds began with an L-proline-mediated three-component Mannich reaction between commercially available 3-fluoro-4-morpholinoaniline, aqueous formaldehyde and α-hydroxyacetone. This was followed by a one-step cyclisation to form the core structure of oxazolidinone antibiotics which was subsequently derivatized. The novel compounds were evaluated for their antibacterial activity against M. smegmatis. One of the novel oxazolidinone derivatives 18 a1 produced a M

Evaluation of omadacycline in vitro activity against Corynebacterium species.

Corynebacterium spp. are gram-positive bacteria increasingly recognized as pathogens. This study evaluates the MICs of omadacycline, a tetracycline, against 40 clinical Corynebacterium isolates using two methodologies: broth microdilution (BMD) and Liofilchem omadacycline MIC Test Strip (MTS). By BMD, the MIC50, MIC90, and MIC range were 0.5 µg/mL, 1 µg/mL, and 0.12-2.0 µg/mL, respectively. Comparing BMD to MTS, essential agreement (EA, within ±1 doubling dilution of the reference BMD MIC) was 87.5% (95% CI: 73.0%-95.4%).

Tetrachloromaxamycins: Divergent Total Synthesis and Initial Assessments.

Divergent total syntheses of binding pocket and peripherally modified tetrachlorovancomycins, a non-native synthetic glycopeptide, and their evaluation are disclosed. Central to the approach is the synthesis of a single late-stage intermediate that bears a residue 4 thioamide ([Ψ[C(═S)NH]Tpg4]tetrachlorovancomycin (3), LLS 15 steps, 14% overall) as a precursor to either of two key pocket modifications and their pairing with any combination of two peripheral modifications conducted without protecting groups. A stereochemical simplification achieved by the addition of two aryl chlorides removes

Characterization and susceptibility of non-albicans Candida isolated from various clinical specimens in Lebanese hospitals.

BACKGROUND: Invasive fungal infections have presented a challenge in treatment. In the past, it was known that the frontrunner in such infections is Candida albicans with little emphasis placed on non-albicans Candida species (NAC). Studies worldwide have shown a rise in fungal infections attributed to non-albicans Candida species. The aim of this study is to describe the epidemiology of NAC infections along with an overview of resistance in Lebanese hospitals. METHODS: This is a two-year observational multi-central descriptive study. Between September 2016 and May of 2018, a total of 1000 is

Enhanced fungal specificity and in vivo therapeutic efficacy of a C-22-modified FK520 analog against C. neoformans.

Fungal infections cause significant morbidity and mortality globally. The therapeutic armamentarium against these infections is limited, and the development of antifungal drugs has been hindered by the evolutionary conservation between fungi and the human host. With rising resistance to the current antifungal arsenal and an increasing at-risk population, there is an urgent need for the development of new antifungal compounds. The FK520 analogs described in this study display potent antifungal activity as a novel class of antifungals centered on modifying an existing orally active FDA-approved

Using a combination of short- and long-read sequencing to investigate the diversity in plasmid- and chromosomally encoded extended-spectrum beta-lactamases (ESBLs) in clinical Shigella and Salmonella isolates in Belgium.

For antimicrobial resistance (AMR) surveillance, it is important not only to detect AMR genes, but also to determine their plasmidic or chromosomal location, as this will impact their spread differently. Whole-genome sequencing (WGS) is increasingly used for AMR surveillance. However, determining the genetic context of AMR genes using only short-read sequencing is complicated. The combination with long-read sequencing offers a potential solution, as it allows hybrid assemblies. Nevertheless, its use in surveillance has so far been limited. This study aimed to demonstrate its added value for AM
