Uncovering the neural control of laryngeal activity and subglottic pressure in anaesthetized rats: insights from mesencephalic regions.

To assess the possible interactions between the dorsolateral periaqueductal gray matter (dlPAG) and the different domains of the nucleus ambiguus (nA), we have examined the pattern of double-staining c-Fos/FoxP2 protein immunoreactivity (c-Fos-ir/FoxP2-ir) and tyrosine hydroxylase (TH) throughout the rostrocaudal extent of nA in spontaneously breathing anaesthetised male Sprague-Dawley rats during dlPAG electrical stimulation. Activation of the dlPAG elicited a selective increase in c-Fos-ir with an ipsilateral predominance in the somatas of the loose (p < 0.05) and compact formation (p < 0.01

Evaluation of an in vitro assay to screen for the immunotoxic potential of chemicals to fish.

A wide variety of environmental contaminants has been shown to disrupt immune functions of fish and may compromise their defense capability against pathogens. Immunotoxic effects, however, are rarely considered in ecotoxicological testing strategies. The aim of this study was to systematically evaluate the suitability of an in vitro immuno-assay using selected fish immune parameters to screen for chemicals with known immunotoxic potential and to differentiate them from non-immunotoxicants. Non-stimulated and lipopolysaccharide-stimulated head kidney leukocytes of rainbow trout (Oncorhynchus my

A low concentration of ethanol reduces the chemiluminescence of human granulocytes and monocytes but not the tumor necrosis factor alpha production by monocytes after endotoxin stimulation.

The ability of polymorphonuclear neutrophils (PMNs) and monocytes (Mphi) to produce reactive oxygen species (ROS) has been related closely to their potential in the killing of microorganisms. Ethanol has been shown to impair the generation of ROS in these phagocytes after stimulation with some immunogens and to increase the susceptibility of alcohol abusers to infectious diseases. As endotoxemia is common in alcohol abusers, we investigated the effect of ethanol (21.7 mmol/liter) on the luminol-amplified chemiluminescence of PMNs and Mphi after endotoxin stimulation and the release of tumor ne

[Autoimmunological receptor diseases].

Specific chemical signals are recognized by cell receptors which initiate an appropriate biologic response. Autoimmune reactions against hormone receptors can lead to failure of receptor function. Several mechanisms are responsible for either abnormal stimulation or blockade of the receptor-mediated intracellular events. Immunoglobulins which compete with hormone binding to receptors have been found in Graves' disease, in myasthenia gravis, in acanthosis nigricans with insulin resistance, and in a seizure disorder with primary IgA deficiency.

Opioid peptides in the rabbit carotid body: identification and evidence for co-utilization and interactions with dopamine.

The rabbit carotid body is a catecholaminergic organ that contains dopamine and norepinephrine in a proportion of nearly 5:1. Chronic (15 days) carotid sinus nerve denervation or superior cervical ganglionectomy did not modify the carotid body dopamine content (5-6 nmol/mg of protein, equivalent to 250 pmol per carotid body), but sympathectomy reduced by approximately 50% the norepinephrine content. The carotid body has also a very high content of opioid activity (250 equivalent pmol of Leu-enkephalin/mg of protein) as measured by a radioreceptor assay that detects preferentially delta-opioid

[Effects of chronic pancreatitis on salivary secretion].

In chronic pancreatitis, increased concentrations of Na+ and Cl- in sweat have been attributed to increased parasympathetic drive. It was postulated that similar changes might occur in saliva. To avoid masking increased parasympathetic drive, saliva was collected without stimulation. In patients with chronic pancreatitis, there were significant increases of concentrations of Na+ and Cl- in basal salivary concentrations. These differences disappeared when salivary secretion was stimulated with citric acid. Anatomic or neurologic modification of the salivary glands seemed unlikely as stimulated

Emetic activity of N-substituted norapomorphines.

Norapomorphine and ten of its N-substituted derivatives were prepared by modifications of procedures described earlier. In a dog emesis test the N-ethyl and N-n-propyl compounds had minimum effective doses of 0.00025 and 0.0005 mg/kg, respectively, when administered iv, sc, or im. In a modified Irwin mouse profile screen the minimum effective iv dose was 0.013 mg/kg for the N-ethyl and 0.0024 mg/kg for the N-n-propyl compound; percutaneous absorption was also observed in mice. All compounds examined caused the stereotyped apomorphine behavior syndrome but hypotensive effects were not serious.

Response of genioglossus muscle to increasing chemical drive in sleeping obstructive apnea patients.

STUDY OBJECTIVES: Subjects with a collapsible upper airway must activate their pharyngeal dilators sufficiently in response to increasing chemical drive if they are to maintain airway patency without arousal from sleep. Little is known about the response of pharyngeal dilators to increasing chemical drive in these subjects. We wished to determine, in obstructive apnea patients, the response of the genioglossus to increasing chemical drive and the contribution of mechanoreceptor feedback to this response. DESIGN: Physiological study. SETTING: University-based sleep laboratory. PATIENTS: 20 p

A heart-on-a-chip simultaneously monitoring contractility and troponin secretion under dynamic mechanical stimulation.

Mechanical stimulation can remodel the physiological mechanical microenvironment of cardiomyocytes and influence their maturation and injury progression. However, the dynamic process by which cardiomyocytes transition from adaptive maturation to injury remains unclear, because conventional endpoint assays cannot continuously track cellular states. Here, we developed a heart-on-a-chip platform that reconstructs physiological strain and fluidic microenvironments. The chip integrates traction force microscopy (TFM) and a cardiac troponin I (cTnI) biosensor, enabling synchronous monitoring of card

Respiratory responses to aortic and carotid chemoreceptor activation in the dog.

Respiratory responses arising from both chemical stimulation of vascularly isolated aortic body (AB) and carotid body (CB) chemoreceptors and electrical stimulation of aortic nerve (AN) and carotid sinus nerve (CSN) afferents were compared in the anesthetized dog. Respiratory reflexes were measured as changes in inspiratory duration (TI), expiratory duration (TE), and peak averaged phrenic nerve activity (PPNG). Tonic AN and AB stimulations shortened TI and TE with no change in PPNG, while tonic CSN and CB stimulations shortened TE, increased PPNG, and transiently lengthened TI. Phasic AB and

Chemistry and Effects of Brainstem Acting Drugs.

BACKGROUND: Brain is the most sensitive organ, whereas brainstem is the most important part of Central Nervous System (CNS). It connects the brain and the spinal cord. However, a myriad of drugs and chemicals affects CNS with severe resultant effects on the brainstem. METHODS: In view of this, a number of literature were assessed for information on the most sensitive part of brain, drugs and chemicals that act on the brainstem and clinical benefit and risk assessment of such drugs and chemicals. RESULTS: Findings have shown that brainstem regulates heartbeat, respiration and because it conne

Host heparan sulfate promotes ACE2 super-cluster assembly and enhances SARS-CoV-2-associated syncytium formation.

SARS-CoV-2 infection causes spike-dependent fusion of infected cells with ACE2 positive neighboring cells, generating multi-nuclear syncytia that are often associated with severe COVID. To better elucidate the mechanism of spike-induced syncytium formation, we combine chemical genetics with 4D confocal imaging to establish the cell surface heparan sulfate (HS) as a critical stimulator for spike-induced cell-cell fusion. We show that HS binds spike and promotes spike-induced ACE2 clustering, forming synapse-like cell-cell contacts that facilitate fusion pore formation between ACE2-expresing and

The collagenolytic effects of the traditional Chinese medicine preparation, Han-Dan-Gan-Le, contribute to reversal of chemical-induced liver fibrosis in rats.

Han-Dan-Gan-Le (HDGL), a Chinese herb preparation composed of Stephaniat tetrandra, Salvia miltorrhiza, Radix paeoniae, Astragalus membranaceus, and Ginkgo biloba, has been used to treat human liver fibrosis. This study was designed to examine the therapeutic effect of HDGL on chemical-induced liver fibrosis in adult Wistar rats. Liver fibrosis was produced in rats by carbon tetrachloride (1.2 ml CCl(4)/kg, 2 times/week, after an initial dose of 5.0 ml CCl(4)/kg, sc), plus a diet of 20% fat, 0.05% cholesterol (continuous) and 30% alcohol in the drinking water ad libitum (every other day) for 8

Local lymph node assay responses to paraphenylenediamine: intra- and inter-laboratory evaluations.

The murine local lymph node assay (LLNA) is a method for the prospective identification of skin sensitizing chemicals. Proliferative responses induced in lymph nodes draining the site of topical application of the test chemical are measured and those chemicals that induce a stimulation index of three or more compared with concurrent vehicle-treated controls are considered to have the potential to cause skin sensitization. Dose-response data from the LLNA may be used to derive an estimate of relative skin sensitizing potency, based upon derivation of the concentration of chemical required to ca

Rhythmical and non-rhythmical spontaneous activity recorded from the central cut end of the sinus nerve.

1. Two types of spontaneous nervous activities have been recorded from the central cut end of the carotid sinus nerve of the cat.2. One type was composed of nerve potentials that exhibited a respiratory or cardiac rhythm, whose rate of firing was depressed by the pressor response to adrenaline, and that were found to arise from post-ganglionic fibres of the superior cervical ganglion.3. The other type of activity consisted of non-rhythmical nerve potentials whose rate of discharge increased 10-30 sec after the injection of adrenaline. The activity of these fibres also increased when the arteri

Direct mechanical stimulation triggers hatching in Trichuris suis eggs.

Hatching of Trichuris suis eggs within the host intestine is essential for the establishment of infection; however, the mechanisms underlying its induction remain incompletely understood. Although previous studies have suggested the involvement of intestinal microbiota and chemical factors, the contribution of physical cues has not been systematically examined. In the present study, we employed an in vitro hatching system to investigate the environmental signals required for egg hatching. Under host body temperature conditions (38 °C), hatching was not induced in the absence of physical stimul

Recombinant gonadotrophin-based, ovarian hyperstimulation syndrome-free stimulation of the high responder: suggested protocol for further research.

Ovarian stimulation of the high responder remains controversial. The narrow margin between a too low gonadotrophin dose (and abnormal oestradiol rise) and excessive stimulation (risk of OHSS) is a challenging situation. This study describes an OHSS-free protocol, based on the use of recombinant gonadotrophins for stimulation and gonadotrophin-releasing hormone (GnRH) antagonist to prevent premature LH surge. In six patients with a previous history of excessive response, stimulation was started on day 3 of cycle with recombinant FSH. When follicular diameter reached 13-14 mm, a daily injection
