In the field of wearable robotics, assessing physical human-exoskeletons interaction (pHEI) is of critical importance in the development of wearable assistive devices. However, in the early stages of development of such devices, involving human testing could raise safety and ethical concerns. To address those limitations, we developed an active dummy leg named Leg Replica. Yet, the realism for pHEI was not adequate when mimicking soft-tissue behavior (compression and shear). The work presented here focused on identifying synthetic soft tissue simulants to enhance the fidelity of the Leg Replic

The extracellular matrix (ECM) has a specific architecture that defines tissue mechanics and helps to guide cell functions. In tissue engineering, it is crucial to be able to mimic the architecture and mechanics of the ECM with biomaterials. However, mimicking the architecture and mechanics of tissues that contain ECM elements oriented at the nanoscale, such as articular cartilage, tendon, intervertebral disc, and cornea, remains challenging. To succeed, an emerging strategy is to orient nanocrystals within materials by using manufacturing techniques such as extrusion printing and magnetic ind…

Current treatments for bone diseases are facing significant limitations, including adverse side effects, high costs and safety concerns. Surgical interventions and biologics (e.g., BMP-2) also present challenges in terms of complications, immunogenicity, and spatiotemporal control. Bone tissue engineering (BTE) uses biomaterials (e.g. hydroxyapatite, biodegradable polymers, hyaluronic acid) to provide structural support and ensure biocompatibility. However, these materials exhibit certain drawbacks, including mismatched degradation rates, inadequate mechanical strength, limited bioactivity, an…

Age related macular degeneration and other retinal degenerative disorders are characterized by disruption of the outer blood retinal barrier (oBRB) with subsequent ischemia, neovascularization, and atrophy. Despite the treatment advances, there remains no curative therapy, and no treatment targeted at regenerating native-like tissue for patients with late stages of the disease. Here we present advances in tissue engineering, focusing on bioprinting methods of generating tissue allowing for safe and reliable production of oBRB as well as tissue reprogramming with induced pluripotent stem cells …

Though crucial for natural bone healing, local calcium ion (Ca2+) and phosphate ion (Pi) concentrations can exceed the cytotoxic limit leading to mitochondrial overload, oxidative stress, and cell death. For bone tissue engineering applications, H2S can be employed as a cytoprotective molecule to enhance mesenchymal stem cell (MSC) tolerance to cytotoxic Ca2+/Pi concentrations. Varied concentrations of sodium hydrogen sulfide (NaSH), a fast-releasing H2S donor, were applied to assess the influence of H2S on MSC proliferation. The results suggested a toxicity limit of 4 mM for NaSH and that 1 m…

Several years have passed since the medical and scientific communities leaned toward tissue engineering as the most promising field to aid bone diseases and defects resulting from degenerative conditions or trauma. Owing to their histocompatibility and non-immunogenicity, bone grafts, precisely autografts, have long been the gold standard in bone tissue therapies. However, due to issues associated with grafting, especially the surgical risks and soaring prices of the procedures, alternatives are being extensively sought and researched. Fibrous and non-fibrous materials, synthetic substitutes, …

Osteonecrosis of the femoral head (ONFH) is a devastating and complicated disease with an unclear etiology. Femoral head-preserving surgeries have been devoted to delaying and hindering the collapse of the femoral head since their introduction in the last century. However, the isolated femoral head-preserving surgeries cannot prevent the natural progression of ONFH, and the combination of autogenous or allogeneic bone grafting often leads to many undesired complications. To tackle this dilemma, bone tissue engineering has been widely developed to compensate for the deficiencies of these surger…

This review paper is motivated by a Back-to-Basics presentation given by the author at the 2022 Orthopaedic Research Society meeting in Tampa, Florida. I was tasked with providing a brief history of research leading up to the introduction of functional tissue engineering (FTE) for tendon and ligament repair. Beginning in the 1970s, this timeline focused on two common orthopedic soft tissue problems, anterior cruciate ligament ruptures in the knee and supraspinatus tendon injuries in the shoulder. Historic changes in the field over the next 5 decades revealed a transformation from a focus more …
