Astrocyte-neurone crosstalk mediates dexmedetomidine sedation.

As the clinical applications of dexmedetomidine expand, advances in neuroscientific research have provided further insights into its mechanisms of action. We provide a focused overview of the role that astrocytes play in the changes in neurotransmission that accompany the sedative action of dexmedetomidine. After activation of astrocytic α2A-adrenergic receptors, the subsequent rise in cytosolic calcium increases the permeability of bestrophin-1 channels, facilitating the release of γ-aminobutyric acid (GABA) that stimulates extrasynaptic GABAA receptors on glutamatergic neurones. This signall…

Advances in the role of astrocytes in glioma progression and therapy.

Glioblastoma is the most common malignant primary intracranial tumor, characterized by high invasiveness, rapid progression, and poor sensitivity to radiotherapy and chemotherapy. Given the limited efficacy of current treatments, novel therapeutic strategies are urgently needed. Astrocytes play pivotal roles in glioma initiation and progression by promoting tumor proliferation, invasion, therapeutic resistance, and immune evasion. Reactive astrocytes are increasingly recognized as dynamic, heterogeneous cell states rather than fixed subtypes. This review discusses representative astrocyte prog…

Astrocytic Accumulation at the Choroid Plexus Attachment Region.

Immune cell entry into the central nervous system (CNS) under neuroinflammatory conditions is commonly discussed in relation to established barrier interfaces, including the blood-brain barrier and the choroid plexus epithelium. In addition, an alternative anatomical route at the choroid plexus attachment region has been proposed, yet its structural organization remains poorly defined. Here, we provide a detailed descriptive analysis of the choroid plexus attachment region and identify glial structures reminiscent of a glia limitans. We analyzed the morphology of the choroid plexus attachment

Astrocyte dysfunction in hydrocephalus: Neuroinflammation and glymphatic impairment.

Hydrocephalus is a common neurological disorder characterized by pathological dilation of the ventricular system. Its pathogenesis involves multiple factors, including cerebrospinal fluid (CSF) dynamic imbalance (encompassing overproduction, circulation obstruction, and impaired absorption) and neuroinflammatory responses. Astrocytes, as core components of the neurovascular unit, mediate CSF transport via their specialized end-foot structures and the glymphatic system formed by perivascular spaces (PVS). Simultaneously, they serve as key effector cells in neuroinflammatory regulation, particip…

Primary Cilia of Astrocytes in the Retina in Development and Disease.

PURPOSE: Defects in primary cilia and ciliary signaling are associated with an array of neurodegenerative diseases. Primary cilia are sensory organelles that transmit and regulate various cellular communication pathways, including the significant Sonic hedgehog and Wnt signal pathways. Extensive studies have shown that neuronal cilia in the central nervous system contribute to early patterning, neuronal maturation, and survival. The behavior of cilia on astrocytes in glaucoma and aging remains poorly understood. This study used rodent and human retinal astrocytes to examine primary cilia and t…

Astrocyte Reactivity by Alcohol Dependence in the Central Amygdala.

Astrocytes play essential roles in maintaining brain homeostasis and in contributing to synaptic functions, but, in response to injury, infection, or disease, astrocytes can downregulate their homeostatic and physiological functions while increasing neuroinflammatory responses. The central amygdala (CeA) is important for stress responsivity and the development of alcohol (ethanol) dependence. Using a multi-omics approach in Aldh1l1-EGFP/Rpl10a mice and the chronic intermittent ethanol two-bottle choice (CIE-2BC) model, we have characterized the translational response of CeA astrocytes, as well…

Astrocytic lipid dysregulation as an early driver of neurodegeneration.

Astrocytes have traditionally been cast as supportive glia, but they are increasingly recognized as metabolic hubs that regulate cholesterol synthesis, fatty acid detoxification, lipid droplet dynamics and redox homeostasis in the CNS. Neurons have a limited intrinsic capacity for lipid storage and detoxification and rely heavily on astrocytes to maintain a safe lipid environment. Emerging evidence indicates that dysregulation of astrocytic lipid homeostasis precedes overt neuronal degeneration in a range of neurodegenerative diseases, including Alzheimer disease, Parkinson disease, amyotrophi…

Astrocyte response in Alzheimer's disease: Good or bad?

Alzheimer's disease (AD) is the most common age-associated neurodegenerative disease globally. The pathogenesis of AD is related to the progressive neuronal apoptosis due to progressive accumulation of extracellular amyloid beta (Aβ) peptide and intracellular neurofibrillary tangles (NFTs) which caused by hyperphosphorylation of tau protein. The accumulation of Aβ and NFTs in AD also affects the supporting cells such as microglia, astrocytes, and oligodendrocytes Predominantly, in response to oxidative stress, neuroinflammation, synaptic failure, and neuronal apoptosis in AD, astrocytes become…

Advances in Astrocyte Research on Neuronal Death and Regeneration.

The onset and progression of central nervous system (CNS) disorders are frequently associated with aberrant neuronal death. In addition to the classical forms of cell death such as apoptosis and necrosis, neurons can also undergo alternative modes of death, including ferroptosis, cuproptosis, and ammonia-induced cell death, all of which may involve the participation of astrocytes. Neuronal death is an irreversible process and plays a central role in the pathogenesis of numerous CNS diseases. We found that astrocytes exhibit the capacity to regenerate into neurons, a characteristic that may pav…

Biomarkers of neuroaxonal and astrocytic pathology in autoimmune encephalitis.

BACKGROUND AND OBJECTIVES: Autoimmune encephalitis (AE) is an immune mediated central nervous system disorder that results in significant symptoms and disability including cognitive dysfunction. Biomarkers of neuro-axonal (neurofilament light, NfL) and astrocytic (glial fibrillary acidic protein, GFAP) pathology in autoimmune encephalitis (AE) are altered in the cerebrospinal fluid and serum in AE. However, their relationship to disease phase and outcomes, particularly cognition, is not well established. Understanding their association with the AE disease course may enhance monitoring and prog

Astrocyte-derived factors regulate CNS myelination.

The role that astrocytes play in central nervous system (CNS) myelination is poorly understood. We investigated the contribution of astrocyte-derived factors to myelination and revealed a substantial overlap in the secretomes of human and rat astrocytes. Using in vitro myelinating co-cultures of primary retinal ganglion cells and cortical oligodendrocyte precursor cells, we discovered that factors secreted by resting astrocytes, but not reactive astrocytes, facilitated myelination. Soluble brevican emerged as a new enhancer of developmental myelination in vivo, CNS and its absence was linked t…

The role of astrocyte in neuroinflammation in traumatic brain injury.

Traumatic brain injury (TBI), a significant contributor to mortality and morbidity worldwide, is a devastating condition characterized by initial mechanical damage followed by subsequent biochemical processes, including neuroinflammation. Astrocytes, the predominant glial cells in the central nervous system, play a vital role in maintaining brain homeostasis and supporting neuronal function. Nevertheless, in response to TBI, astrocytes undergo substantial phenotypic alternations and actively contribute to the neuroinflammatory response. This article explores the multifaceted involvement of ast…

Cuprizone feeding induces swollen astrocyte endfeet.

The cuprizone model is a widely used model to study the pathogenesis of multiple sclerosis (MS). Due to the selective loss of mature oligodendrocytes and myelin, it is mainly being used to study demyelination and the mechanisms of remyelination, as well as the efficiency of compounds or therapeutics aiming at remyelination. Although early investigations using high dosages of cuprizone reported the occurrence of hydrocephalus, it has long been assumed that cuprizone feeding at lower dosages does not induce changes at the blood-brain barrier (BBB). Here, by analyzing BBB ultrastructure with high

Mechanisms of Zika astrocyte infection and neuronal toxicity.

OBJECTIVES: Zika virus (ZIKV) has become an epidemic in several countries and was declared a major public health issue by the WHO. Although ZIKV infection is asymptomatic or shows mild fever-related symptoms in most people, the virus can be transmitted from a pregnant mother to the fetus, resulting in severe brain developmental abnormalities, including microcephaly. Multiple groups have identified developmental neuronal and neuronal progenitor compromise during ZIKV infection within the fetal brain, but little is known about whether ZIKV could infect human astrocytes and its effect on the deve…

Microglia and astrocyte responses to neuropathogenic protozoan parasites.

Cerebral toxoplasmosis and cerebral malaria are two important neurological diseases caused by protozoan parasites. In this review, we discuss recent findings regarding the innate immune responses of microglia and astrocytes to Toxoplasma and Plasmodium infection. In both infections, these tissue-resident glial cells perform a sentinel function mediated by alarmin crosstalk that licenses adaptive type 1 immunity in the central nervous system. Divergent protective or pathogenic effects of type 1 activation of these astrocytes and microglia are revealed depending on the inherent lytic potential o…
