Biophysical and structural characterization of a zinc-responsive repressor of the MarR superfamily.

The uptake of zinc, which is vital in trace amounts, is tightly controlled in bacteria. For this control, bacteria of the Streptococcaceae group use a Zn(II)-binding repressor named ZitR in lactococci and AdcR in streptococci, while other bacteria use a Zur protein of the Ferric uptake regulator (Fur) superfamily. ZitR and AdcR proteins, characterized by a winged helix-turn-helix DNA-binding domain, belong to the multiple antibiotic resistance (MarR) superfamily, where they form a specific group of metallo-regulators. Here, one such Zn(II)-responsive repressor, ZitR of Lactococcus lactis subsp…

Natural sun drying process of Rugao sausage: Microbial community succession and its crucial role in flavor development.

This study systematically analyzes the dynamic changes in physicochemical properties, microbial community, and flavor compounds of Rugao sau … id bacteria, dominated in the sausage. Among them, 10 species, such as Macrococcus caseolyticus and Lactococcus garvieae, became the dominant species. A total of 30 volatile compounds were identified as key flavor substances related to flavor formation during drying. After excluding flavor compounds derived from spice, these included 8 aldehydes, 2 alcohols, …

Integrated Microbiome and Metabolomics Analysis Reveals Dynamic Changes in Raw Camel Milk During Refrigeration Storage.

This study investigated microbial and metabolite dynamics in raw camel milk stored at 4°C. Physicochemical and microbial monitoring identified d 3 as a critical transition point, with samples from d 0, 1, 3, and 6 subjected to 16S rRNA sequencing and metabolomic analysis. The dominant genera shifted from Lactococcus to Pseudomonas. Metabolomic analysis showed that non-volatile metabolites were primarily composed of esters, lipids, and organic acids, with 9 of these metabolites exhibiting a continuous increasing trend. Similarly, the levels of volatile metabolites, including ketones, esters, an…

Gastrointestinal health. The role of pro- and pre-biotics in standard foods.

BACKGROUND: Diarrhoea is a common symptom of gastrointestinal illness, ranking among the 20 most frequent reasons for patient encounters in general practice in Australia. Children under 5 years of age are especially vulnerable to bacterial gastroenteritis such as infections with Campylobacter jejuni. Everyday diet, in particular the regular consumption of pro- and pre-biotics, can help with the maintenance of a healthy gastrointestinal system. OBJECTIVE: This article outlines the potential of 'everyday standard' food items such as cheese to promote healthy gastrointestinal microflora and to p

Transfer of plasmid-mediated resistance to tetracycline in pathogenic bacteria from fish and aquaculture environments.

The transferability of a large plasmid that harbors a tetracycline resistance gene tet(S), to fish and human pathogens was assessed using electrotransformation and conjugation. The plasmid, originally isolated from fish intestinal Lactococcus lactis ssp. lactis KYA-7, has potent antagonistic activity against the selected recipients (Lactococcus garvieae and Listeria monocytogenes), preventing conjugation. Therefore the tetracycline resistance determinant was transferred via electroporation to L. garvieae. A transformant clone was used as the donor in conjugation experiments with three differen…

Bacterial microbiota diversity in sheep milk during refrigerated storage under different processing conditions.

Sheep milk, while nutritionally dense due to high calcium and B vitamins, spoils rapidly due to its diverse microbiota. Contamination risks arise from environmental Listeria or biofilm-forming microbes during milking and handling. This study employed 16 S rRNA gene sequencing to characterize microbial dynamics in sheep milk treated with ultrasound, pasteurization, and ultrasound/pasteurization during refrigerated storage. Analysis of 71 samples yielded 3,301,066 high-quality reads, revealing significant shifts in microbial community composition and diversity. Results revealed that Lactococcus …

Comparison of Different Nisin Separation and Concentration Methods: Industrial and Cost-Effective Perspectives.

Separation and concentration of biotechnological products are the most important steps of purification stage in downstream processing. In the present study, three nisin separation and concentration methods including salting out by ammonium sulfate, solvent extraction (at 20 °C, 4 °C, and - 10 °C) and direct chromatography of culture medium were compared with each other. According to our results, nisin precipitation by ammonium sulfate at 40% saturation was the most efficient method (yield = 90.04%, purification fold = 168.80). Low yield and fold purification values were obtained by solvent ext

The Lactococcus phages showed strong correlations and co-o …

Mugger crocodile is the only crocodile existing in Iran. The present study was aimed to investigate the bacterial flora in oral and cloacal cavities of wild Mugger crocodiles in Negour protected area, Iran. The isolation and molecular characterization of oral and cloacal bacterial flora were performed in 22 Mugger crocodiles captured in Negour protected area, Iran. Ten bacterial species from all oral samples and six bacterial species from all cloacal samples were recovered. The most commonly isolated bacteria in oral samples were Burkholderia contaminans and Lactococcus garvieae, respectively;…

Immune Boosting Effect of Limosilactobacillus Reuteri in Immunocompetent C57BL/6J Mice.

Probiotics are well-known to be directly or indirectly involved in the host immune system. In this study, we analyzed the immune-boosting effects of lactic acid bacteria, including Limosilactobacillus and Lactococcus, in immunocompetent C57BL/6J mice. Three different lactic acid bacteria strains were orally administered to C57BL/6J mice for 8 weeks. Then, liver, spleen, and whole blood were harvested after sacrificing the animals. There were no significant changes in whole-body weight, weight of organs, or complete blood cell count by oral administration of lactic acid bacteria. The frequencie…

Efficacy, toxicity study and antioxidant properties of plantaricin E and F recombinants against enteropathogenic Escherichia coli K1.1 (EPEC K1.1).

Enteropathogenic Escherichia coli (EPEC) is one of the resistance bacteria towards antibiotics and have been raising problem during treatments. Therefore, a new antibiotic candidate is required. Plantaricin E and F recombinant have been successfully produced by a GRAS host Lactococcus lactis. This study was aimed to evaluate the efficacy and toxicity of plantaricin E and F recombinant against EPEC K1.1 infection by in vivo assay. The production of plantaricin E and F recombinants from Lactococcus lactis was conducted and encapsulated. The in vivo study was carried out by inoculating the mice p…

Most enzymes acting in glycolysis and fermentation pathways are PHX in these genomes, but not those involved in the TCA cycle and respiration, suggesting that these organisms have predominantly adapted to grow rapidly in an anaerobic environment.
