Mechanical transfer of Theileria orientalis: possible roles of biting arthropods, colostrum and husbandry practices in disease transmission.

BACKGROUND: The intracellular protozoal parasite Theileria orientalis has rapidly spread across South-eastern Australia, substantially impacting local cattle industries since 2006. Haemaphysalis longicornis appears to be a biological vector in the endemic regions. Mechanical transfer of blood by biting arthropods, in colostrum or iatrogenic transmission though husbandry procedures is another possible mode of transmission. This study assesses the risk of these mechanical modes of transmission. METHODS: Blood was collected from a T. orientalis Ikeda positive Angus steer, and was inoculated into

A longitudinal assessment of the serological response to Theileria parva and other tick-borne parasites from birth to one year in a cohort of indigenous calves in western Kenya.

Tick-borne diseases are a major impediment to improved productivity of livestock in sub-Saharan Africa. Improved control of these diseases would be assisted by detailed epidemiological data. Here we used longitudinal, serological data to determine the patterns of exposure to Theileria parva, Theileria mutans, Babesia bigemina and Anaplasma marginale from 548 indigenous calves in western Kenya. The percentage of calves seropositive for the first three parasites declined from initial high levels due to maternal antibody until week 16, after which the percentage increased until the end of the stu

Equine babesiosis: seroprevalence, risk factors and comparison of different diagnostic methods in Jordan.

The purposes of this study were to estimate the seroprevalence and distribution of horse piroplasmosis, to evaluate risk factors associated with the occurrence of the disease and to compare the different diagnostic methods used for this disease. A total of 253 clinically normal horses were sampled, and a collection form was completed for each horse from five of six different climatic zones of Jordan. The sixth zone was not sampled because it did not include horse population. Competitive enzyme-linked immunosorbent assay (cELISA) revealed 37 horses (14.6%) positive for Theileria equi, and none

Treatment of East Coast fever of cattle with a combination of parvaquone and frusemide.

Pulmonary oedema is a common sign of East Coast fever (ECF, Theileria parva infection) of cattle. A trial was conducted on farms in Uganda to compare a product containing both the antitheilerial compound parvaquone and the diuretic compound frusemide with one containing only parvaquone, in the treatment of ECF. The trial involved 40 clinical cases of ECF, some of them complicated by other infections, in cattle of all ages and on several farms. Confirmed cases were treated with either parvaquone+frusemide (P+F) or parvaquone alone (P). Survival after treatment with P+F was 77% compared with 71%

Phylogeny of sheep and goat Theileria and Babesia parasites.

The phylogenetic relationship of Theileria and Babesia species infecting sheep and goats on the basis of their 18S RNA gene structure was addressed in the present study. For this purpose, the complete sequences of the small ribosomal RNA genes of a panel of sheep and goat piroplasm isolates, including T. lestoquardi, T. ovis, T. separata, B. ovis, B. motasi, B. crassa and several novel species, were sequenced and compared. The classification based on the established phylogenetic tree corresponded with traditional systematics and revealed that sheep/goat piroplasm species are of polyphyletic or

Live vaccines against hemoparasitic diseases in livestock.

Live vaccines against hemoparasitic diseases in livestock are based on parasites derived from culture (Theileria annulata), from blood of infected animals (Babesia bovis, Babesia bigemina, Anaplasma centrale, (attenuated) Anaplasma marginale and Cowdria ruminantium), and from ticks (Theileria parva). The T. annulata attenuated cultured schizont vaccine is safe for all varieties of cattle. Blood derived vaccines are recommended mainly for young cattle, the age limit varying with the different vaccines and breeds of cattle. In older animals, monitoring of the individual response is needed. Immun

The effects of oxytetracycline on Theileria parva in vitro.

When bovine peripheral blood leucocytes were infected with Theileria parva sporozoites, immediate treatment with oxytetracycline (OTC) inhibited the development of sporozoites to mature schizonts. The extent of inhibition was dependent on drug concentration and duration of treatment. Concentrations of 5 micrograms/ml OTC, or higher, for 8 days completely inhibited the establishment of schizonts and their ability to transform host cells. A cytostatic effect on schizont-infected cell lines was found with three tetracyclines and was also demonstrated on uninfected lymphoblasts. The parasites were

Salivary gland of the tick vector of East Coast fever. IV. Cell type selectivity and host cell responses to Theileria parva.

Responses of cells in the tick salivary gland to parasitism by Theileria parva were studied by electron microscopy. The gland is composed of three distinct types of acini (I, II, III) which together include ten or more different cell types. Of some 30 infected cells observed in the present study, all were E-cells of acinus III. The parasite thus exhibits a high degree of selectivity for acinus and cell type. The glandular cell invaded undergoes massive hypertrophy and accumulates glycogen deposits in its cytoplasm which may serve as an energy source for the growing intracellular parasite. As s

[New findings on the immunology of protozoan blood parasitoses in domestic animals].

Intensive research in the field of the immunology of protozoan blood parasitoses has led to a further clarification of the interrelations between parasites and hosts. Progress in the analysis of the antigen character of the parasites and investigations of the immune response form the basis for extending serological diagnosis and improved vaccines. The effectiveness of the vaccines against babesioses could be increased. Vaccines have been tested successfully against tropical theileriosis and East Coast fever. It is only with African trypanosomes that immunoprophylaxis seems to be impossible at

Molecular evidence of co-infections with Anaplasma and Theileria species in domestic sheep and goats from Alentejo, Portugal.

Anaplasma spp. and piroplasmids are tick-borne pathogens with worldwide distribution, affecting domestic animals, wildlife, and humans. Although several tick-borne species have relevance in animal health and can cause economic losses in livestock farms, little is known about the epidemiological situation in small ruminants in Portugal. We aimed to assess the infection prevalence, molecular characterization, co-infection patterns and risk factors of Anaplasma spp. and piroplasmids in sheep and goat farms from the Alentejo region (south-central Portugal). A total of 290 blood samples collected i
