Effect of cyclophosphamide on lymphokine production in MRL/lpr.Yaa mice.

The Y chromosome (Yaa gene) from autoimmune BXSB mice has been shown to be responsible for the acceleration of autoimmune symptoms when transferred to MRL/lpr mice. We examined the pathological, serological and immunological characteristics of MRL/lpr. Yaa mice and the suppressive effect of cyclophosphamide (CP) on the mice. MRL/lpr. Yaa mice spontaneously developed a massive lymphadenopathy characterized by hypergammaglobulinemia, the presence of several autoantibodies, and autoimmune disease. In MRL/lpr. Yaa mice, IL-2, IL-4 and IL-5 production in concanavalin A (Con A)-stimulated splenocyte…

The effect of cyclophosphamide on the eruption of impeded and resected incisors in rats.

It is known that a single dose of cyclophosphamide (40 mg.kg-1 intraperitoneal) slows the eruption of unimpeded incisors but has no effect on impeded incisors. In this study incisors that were impeded when cyclophosphamide was given were subsequently made unimpeded. When they were unimpeded they erupted more slowly than the controls. Therefore the changes cyclophosphamide produces which slow eruption are formed in impeded incisors but have an effect only in unimpeded incisors. It has been reported that cyclophosphamide slows the eruption of resected incisors when it is given before the resecti…

Transplacental teratogenesis and mutagenesis in mouse fetuses treated with cyclophosphamide.

We studied transplacental fetotoxicity, teratogenicity, and mutagenicity in Swiss Webster mice following different doses of cyclophosphamide (CP; 0, 5, 10, 15, or 20 mg/kg), a well-known mutagen/teratogen, on day 12 of gestation. The fetal survival and weight on day 18 of gestation decreased significantly with increasing CP dose (P less than 0.01). The CP-treated fetuses were also dysmorphic (e.g., shortened limbs, digital defects, cleft palate, open eyes, and hydrocephaly) and the percentage of dysmorphology increased with increasing CP doses (P less than 0.01). To evaluate mutagenesis, a sep…

Synchronization of plasmapheresis and pulse cyclophosphamide therapy in pemphigus vulgaris.

Long-lasting and complete remission was obtained in a 48-year-old patient with refractory pemphigus vulgaris by an experimental treatment protocol that tries to synchronize plasmapheresis with subsequent pulse cyclophosphamide therapy. The rationale of the approach tries to utilize the plasmapheresis-induced increased proliferation of pathogenic cell clones for partial deletion of these clones through application of maximum pulse immunosuppression treatment during the period of assumed maximum proliferation and, thus, maximum vulnerability of the antibody-producing cells. The treatment schedul…

Cyclophosphamide in the male rat: behavioral effects in the adult offspring.

Cyclophosphamide in a daily dose of 10 mg/kg was injected intraperitoneally in male Wistar rats for 15 days. Two rats each time were killed for testis examination at regular intervals within 100 days following treatment (over 2 spermatogenetic cycles). One hundred days after the end of the treatment, the other rats, whose spermatogenesis had recovered in the meantime, were mated with 3-month-old females. Offspring were evaluated in regard to the mean number per litter, sex ratio, frequency of gross external malformations, growth pattern, mortality in the first 4 months of life and reproductive…

Cyclophosphamide (Cytoxan). A review on relevant pharmacology and clinical uses.

Cyclophosphamide (Cytoxan; Cy) is an alkylating agent with cytotoxic and immunosuppressive activities. The parent compound is inactive in vitro and exerts its biologic activity through metabolites, mainly phosphoramide mustard generated by hepatic microsomal enzymes. The exact mode of cytotoxic and immunosuppressive action of Cy at cellular level is not completely understood. Myelosuppression, hemorrhagic cystitis, alopecia, and gonadal damage are the main toxic effects. Available data suggest that Cy has carcinogenic potential in humans. Cy is widely used for cancer chemotherapy. As an immuno…

Cloning of African trypanosomes in mice immunosuppressed by cyclophosphamide treatment.

The use of cyclophosphamide-treated mice for purpose of cloning the African trypanosomes was assessed. C3HeB/FeJ mice were injected with 200 mg/kg cyclophosphamide (CY) 24-72 hours prior to infection with a single trypanosome isolated from Trypanosoma rhodesiense infected blood samples. All CY-treated mice exhibited depressed parasite-exposure, which was a period of time sufficient to grow trypanosomes from a single organism to a fulminating parasitemia. Cloning efficiency was routinely 45% in these animals. Thus, our study demonstrates that CT-treated mice are a convenient and efficient vehic…

Differential effects of cyclophosphamide on Coxiella burnetii infection in mice.

Treatment of mice with a single dose (200 mg/kg body weight) of cyclophosphamide (CPA) 3 days before intraperitoneal (i. p.) administration of Coxiella burnetii did not substantially affect either the yield of C. burnetii from mouse spleen or the splenomegaly induced by the infection. Administration of the same CPA dose 2 days after i. p. infection resulted in an increased yield of C. burnetii from the spleen and in a reduction of the spleen size as compared to CPA-untreated and CPA-pretreated infected mice. The yields of C. burnetii from the blood and peritoneum were similarly affected. The r…

Role of glutathione in the metabolism-dependent toxicity and chemotherapy of cyclophosphamide.

The role of glutathione in the biological effects of cyclophosphamide (CP) was evaluated by investigating the following: effect of CP on hepatic glutathione levels; relationship between hepatic glutathione depletion (repletion) and the binding of [chloroethyl-3H]CP and [4-14C]CP to hepatic macromolecules; effects of interaction between CP (or acrolein) and diethyl maleate (a classical glutathione depletor), and/or between CP and cysteine on the binding of labeled CP to hepatic macromolecules, on the induction of hematuria, on the content of hepatic cytochrome P-450, on weight gain in rats, on …

Bolus corticosteroids and cyclophosphamide for initial treatment of Wegener's granulomatosis.

Wegener's granulomatosis, a necrotizing vasculitis involving primarily the upper and lower respiratory tract and the kidney, may be rapidly fatal. We describe two children with Wegener's granulomatosis in whom we observed dramatic improvement within 72 hours following bolus corticosteroid therapy combined with cyclophosphamide.

The effects of cyclophosphamide, nitrosomethylurea and benzo(a)pyrene on the rat incisor.

Wistar rats given a single intraperitoneal injection of 75 mg cyclophosphamide revealed dental anomalies in the form of shortening or lengthening of incisors, and development of supernumerary teeth. Rats given additionally 5 mg/kg b. w. of a nitrosomethylurea solution over a period of four months showed the same dental anomalies, but with relatively shorter latency times. In a supplementary investigation examining the effects of benzo(a)pyrene and nitrosomethylurea applied locally to the organon dentale, neoplastic lesions and dental dysplasia were seen.

Influence of cyclophosphamide and 4-ketocyclophosphamide on mouse limb development.

Many studies have been performed on the in vivo teratogenicity of cyclophosphamide, and there is uncertainty whether the parent compound or P-450 generated alkylating metabolite(s) is the proximal teratogen(s). We have examined the influence of cyclophosphamide and a metabolite, 4-ketocyclophosphamide, on mouse limb development. Pregnant mice were injected with 10, 15 or 20 mg/kg of cyclophosphamide on days 9 to 11 of gestation. Hindlimb buds were maximally sensitive to 20 mg/kg of cyclophosphamide at 9 A.M. on day 11, and the predominant malformations formed were preaxial ectrodactyly and hem…

Effect of cyclophosphamide on experimental Nocardia asteroides infection in mice.

It was found that cells of Nocardia asteroides GUH-2 (virulent) were approximately 10 times more virulent than cells of N. asteroides 14759 (intermediate) and greater that 500 times more virulent than N. asteroides 10905 (avirulent) cells when early-stationary-phase cultures suspended in saline were injected intravenously into "normal" mice. There appeared to be a specific organ tropism for each strain. Thus, N. asteroides GUH-2 infected primarily the kidneys, N. asteroides 14759 infected the lungs and heart, and N. asteroides 10905 (in large doses) infected the lungs. Cyclophosphamide treatme…
