Association of three genetic loci with uric acid concentration and risk of gout: a genome-wide association study.

BACKGROUND: Hyperuricaemia, a highly heritable trait, is a key risk factor for gout. We aimed to identify novel genes associated with serum uric acid concentration and gout. METHODS: Genome-wide association studies were done for serum uric acid in 7699 participants in the Framingham cohort and in 4148 participants in the Rotterdam cohort. Genome-wide significant single nucleotide polymorphisms (SNPs) were replicated in white (n=11 024) and black (n=3843) individuals who took part in the study of Atherosclerosis Risk in Communities (ARIC). The SNPs that reached genome-wide significant associat

Genome-wide association study of clinically defined gout identifies multiple risk loci and its association with clinical subtypes.

OBJECTIVE: Gout, caused by hyperuricaemia, is a multifactorial disease. Although genome-wide association studies (GWASs) of gout have been reported, they included self-reported gout cases in which clinical information was insufficient. Therefore, the relationship between genetic variation and clinical subtypes of gout remains unclear. Here, we first performed a GWAS of clinically defined gout cases only. METHODS: A GWAS was conducted with 945 patients with clinically defined gout and 1213 controls in a Japanese male population, followed by replication study of 1048 clinically defined cases an

Genome-Wide Association Study of Pterygium Versus Pinguecula Identifies Loci Associated with Lesion-Type Divergence.

PURPOSE: Both pterygium and pinguecula are ocular surface lesions associated with shared environmental risk factors, including ultraviolet (UV) radiation exposure. However, most pingueculae do not progress to pterygium, and while prior genetic studies suggest a genetic contribution to pterygium susceptibility, the difficulty of controlling for shared UV exposure has been a limiting factor. Here, we use array-based data and whole genome sequencing (WGS)-based data from the multiancestry All of Us Research Platform to perform a case-case genome-wide association study directly comparing individua…

Genetic architecture of sleep in a genome wide association study of device measured sleep traits.

Sleep is essential for health and regulated by genetic and environmental factors. We perform genome-wide association studies of device-measured sleep duration, efficiency, and accelerometer-derived rapid eye movement (REM) and non-rapid eye movement (NREM) sleep in 80,013 UK Biobank participants. We identify 20 autosomal loci, 12 of which have not been previously reported, including genome-wide significant associations for REM and NREM sleep duration. MEIS1 shows strong opposing effects on REM and NREM durations and is intolerant to loss-of-function mutations, suggesting an essential role in t

Genome-wide association study reveals two novel genetic loci associated with chronic lung allograft dysfunction.

BACKGROUND: Chronic lung allograft dysfunction (CLAD) leads to declining respiratory function and high mortality, representing the main barrier to long-term survival in lung transplantation (LT). We performed the first genome-wide association study (GWAS) investigating donor's and recipient's genetic factors associated with CLAD. METHOD: We genotyped 392 donor-recipient pairs from the multicentric Cohort in Lung Transplantation. We tested 4.5 million SNPs for association with CLAD using multivariable logistic regression models corrected for age, sex, initial disease and genetic ancestry. Thre…

Genome-Wide Association Study of Chronic Venous Insufficiency and Lymphedema in the Million Veteran Program.

BACKGROUND: Lymphedema is a chronic condition characterized by the accumulation of fluid due to impaired lymphatic drainage, frequently occurring secondary to chronic venous insufficiency (CVI), malignancy, or obesity. Despite known environmental and clinical risk factors, the genetic contributors to lymphedema and CVI have been understudied. METHODS: We conducted a multipopulation genome-wide association study in participants of the Million Veteran Program without cancer to identify genetic variants associated with CVI, lymphedema, and their cooccurrence. Individuals were categorized into 3 …

Genome-Wide Association Study in European Beech (Fagus sylvatica L.) for Drought Stress Related Traits.

Genome-wide association studies (GWAS) aim to identify loci associated with different traits, which could be evaluated in subsequent studies and later be prioritized in individual and seed stand selection. In trees, traits are measured at maturity or are only available after a long time, and juvenile-mature correlations are often unknown. Therefore, it is a promising approach to study natural populations directly. Data were collected from approximately 100 adult beech trees per stand in five locations in the Postăvarul Massif, South-Eastern Romanian Carpathians (Carpații de Curbură) along an a

Cross-ancestry genome-wide association study identifies implications of SORL1 in cerebral beta-amyloid deposition.

GWAS of Alzheimer's disease have been predominantly based on European ancestry cohorts with clinically diagnosed patients. Increasing the ancestral diversity of GWAS and focusing on imaging brain biomarkers for Alzheimer's disease may lead to the identification of new genetic loci. Here, we perform a GWAS on cerebral β-amyloid deposition measured by PET imaging in 3,885 East Asians and a cross-ancestry GWAS meta-analysis with data from 11,816 European participants. Our GWAS analysis replicates known loci (APOE4, CR1, and FERMT2) and identifies a novel locus near SORL1 that is significantly ass

Genome-Wide Association Study of Hypoglycemia in Adults With Diabetes in the Million Veteran Program.

UNLABELLED: Hypoglycemia is a preventable adverse treatment effect in diabetes patients, but genetic markers to identify those with increased susceptibility are lacking. We performed a case/control genome-wide association study (GWAS) of hypoglycemia in U.S. Million Veteran Program (MVP) participants with medication-treated diabetes. Case participants had an outpatient random serum/plasma glucose <70 mg/dL or an emergency department visit for hypoglycemia. GWAS was stratified by race/ethnicity, adjusted for age at MVP enrollment, sex, and top 10 population-specific principal components, follow…

Integrating Expression Quantitative Trait Loci and Genome-Wide Association Study Identifies Druggable Genes for Tinnitus.

BACKGROUND: The investigation of druggable target genes through large-scale expression quantitative trait loci (eQTL) and genome-wide association study (GWAS) data has demonstrated promise across various diseases. This approach has yet to be explored in the context of tinnitus. METHODS: We obtained cis-eQTL data for 3,453 druggable genes from eQTLGen. Tinnitus phenotype derived from the UK Biobank was used as the discovery cohort. A large-scale Mendelian randomization (MR) analysis was conducted to investigate the inferred causal relationships between the 3,453 druggable genes and tinnitus. R…

Genome-Wide Association Study Identifies a Locus at 17p13 to Be Associated With Intestinal Malrotation.

BACKGROUND: Intestinal malrotation is a congenital malformation of the embryonic gut that may cause midgut volvulus either in children or adults. Our understanding of its genetic background stems from reports of syndromic or familial forms and no genome-wide association studies (GWAS) have been reported. We perform the first GWAS to identify common variants associated with this malformation. METHODS: Subjects were enrolled and genotyped as part of the Mass General Brigham Biobank and individuals with diagnosis of intestinal malrotation were identified. Single nucleotide polymorphisms (SNPs) w

Genome wide association study and genomic risk prediction of age related macular degeneration in Israel.

The risk of developing age-related macular degeneration (AMD) is influenced by genetic background. In 2016, the International AMD Genomics Consortium (IAMDGC) identified 52 risk variants in 34 loci, and a polygenic risk score (PRS) from these variants was associated with AMD. The Israeli population has a unique genetic composition: Ashkenazi Jewish (AJ), Jewish non-Ashkenazi, and Arab sub-populations. We aimed to perform a genome-wide association study (GWAS) for AMD in Israel, and to evaluate PRSs for AMD. Our discovery set recruited 403 AMD patients and 256 controls at Hadassah Medical Cente…

A Genome-Wide Association Study Reveals a Novel Susceptibility Locus for Pancreas Divisum at 3q29.

INTRODUCTION: Pancreas divisum (PD) is a common congenital anomaly of the pancreas, but its genetic basis remains unknown. The purpose of this genome-wide association study was to identify genetic loci associated with PD. METHODS: Using the Mass General Brigham Biobank, patients diagnosed with PD were identified. Quality control and imputation were performed using standard approaches. Single nucleotide polymorphisms (SNPs) with minor allele frequency (MAF) ≥ 5% were tested for association with PD using mixed linear model-based association analysis. The significance threshold was set at 5 × 10…

Interpreting elevated liver blood test results through a genetic lens: A genome-wide association study.

BACKGROUND AND AIMS: Individuals with genetic polymorphisms in UGT1A1 exhibit bilirubin levels that belie their risk of liver disease (Gilbert's syndrome) but it is not known if this phenomenon extends to other common liver blood tests (LBTs). METHODS: A genome-wide association analysis of 10 LBTs was conducted using the UK biobank. Polygenic scores (PGS) were created from discordant loci (e.g. loci associated with the LBT but not associated with cirrhosis morbidity risk). Participants were assigned to a low, intermediate or high PGS for each LBT. A high PGS approximates Gilbert's syndrome (i

Genome-wide association study identifies human genetic variants associated with fatal outcome from Lassa fever.

Infection with Lassa virus (LASV) can cause Lassa fever, a haemorrhagic illness with an estimated fatality rate of 29.7%, but causes no or mild symptoms in many individuals. Here, to investigate whether human genetic variation underlies the heterogeneity of LASV infection, we carried out genome-wide association studies (GWAS) as well as seroprevalence surveys, human leukocyte antigen typing and high-throughput variant functional characterization assays. We analysed Lassa fever susceptibility and fatal outcomes in 533 cases of Lassa fever and 1,986 population controls recruited over a 7 year pe
