Plant-Derived Natural Products and Selective Apoptosis: A Cancer-Cell Vulnerability-State Framework from Redox Imbalance to Membrane-Ion Dysregulation.

Plant-derived natural products remain a major source of anticancer chemical diversity, yet much of the preclinical literature still describes their activity through compound-centered readouts such as IC50 values, reactive oxygen species generation, mitochondrial depolarization, BCL-2-family remodeling, caspase activation, and PI3K/Akt or NF-κB inhibition. Although informative, these mechanisms do not fully explain why the same phytochemical induces apoptosis in some cancer cells, and cytostasis or adaptation in others, while normal cells frequently activate cytoprotective responses. Rather tha

AS03 adjuvant enhances the magnitude, persistence, and clonal breadth of memory B cell responses to a plant-based COVID-19 vaccine in humans.

Vaccine adjuvants increase the breadth of serum antibody responses, but whether this is due to the generation of antigen-specific B cell clones with distinct specificities or the maturation of memory B cell clones that produce broadly cross-reactive antibodies is unknown. Here, we longitudinally analyzed immune responses in healthy adults after two-dose vaccination with either a virus-like particle COVID-19 vaccine (CoVLP), CoVLP adjuvanted with AS03 (CoVLP+AS03), or a messenger RNA vaccination (mRNA-1273). CoVLP+AS03 enhanced the magnitude and durability of circulating antibodies and antigen-

Myrtus communis and Celastrol enriched plant cell culture extracts control together the pivotal role of Cutibacterium acnes and inflammatory pathways in acne.

INTRODUCTION: Acne is a multifactorial inflammatory disease of the pilosebaceous unit in which Cutibacterium acnes is one of the main triggers. A strong predominance of C. acnes phylotype IA1 is present in acne skin with higher biofilm organization and virulence, promoting local immuno-inflammation, especially the Th17 pathway. OBJECTIVES: We evaluated the single and combined pharmacological properties of the plant extracts, Myrtus communis (Myrtacine®) and Celastrol enriched plant cell culture (CEE) extracts on the C. acnes/Th17 pathway. METHODS: The effect of Myrtacine® on the virulence of

Screening of plant-based natural compounds as an inhibitor of FtsZ from Salmonella Typhi using the computational, biochemical and in vitro cell-based studies.

Salmonella Typhi is emerging as a drug-resistant pathogen, particularly in developing countries. Hence, the progressive development of new antibiotics against novel drug targets is essential to prevent the spread of infections and mortality. The cell division protein FtsZ is an ideal drug target as the cell wall synthesis in bacteria is driven by the dynamic treadmilling nature of the FtsZ. The polymerization of the FtsZ provides the essential mechanical constricting force and flexibility to modulate the cell wall synthesis. Any alteration in FtsZ polymerization leads to the bactericidal or ba

Potential of Medicinal Plants as Neuroprotective and Therapeutic Properties Against Amyloid-β-Related Toxicity, and Glutamate-Induced Excitotoxicity in Human Neural Cells.

Alzheimer's disease (AD) and Parkinson's disease (PD) are notorious neurodegenerative diseases amongst the general population. Being age-associated diseases, the prevalence of AD and PD is forecasted to rapidly escalate with the progressive aging population of the world. These diseases are complex and multifactorial. Among different events, amyloid β peptide (Aβ) induced toxicity is a well-established pathway of neuronal cell death, which plays a vital function in AD. Glutamate, the major excitatory transmitter, acts as a neurotoxin when present in excess at the synapses; this latter mechanism

Extract from Aphloia theiformis, an edible indigenous plant from Reunion Island, impairs Zika virus attachment to the host cell surface.

The mosquito-borne Zika virus (ZIKV) belongs to the flavivirus genus of the Flaviviridae family. Contemporary epidemic strains of ZIKV are associated with congenital malformations in infants, including microcephaly, as well as Guillain-Barré syndrome in adults. A risk of human-to-human transmission of ZIKV is also well documented. A worldwide research effort has been undertaken to identify safe and effective strategies to prevent or treat ZIKV infection. We show here that extract from Aphloia theiformis, an edible endemic plant from Indian Ocean islands, exerts a potent antiviral effect agains

Plumbagin, a plant-derived naphthoquinone metabolite induces mitochondria mediated apoptosis-like cell death in Leishmania donovani: an ultrastructural and physiological study.

Naphthoquinones are known to exhibit a broad range of biological activities against microbes, cancer and parasitic diseases and have been widely used in Indian traditional medicine. Plumbagin is a plant-derived naphthoquinone metabolite (5-hydroxy-2-methyl-1,4-naphthoquinone) reported to inhibit trypanothione reductase, the principal enzyme and a validated drug target involved in detoxification of oxidative stress in Leishmania. Here, we report the mechanistic aspects of cell death induced by plumbagin including physiological effects in the promastigote form and ultrastructural alterations in

Novel plant virus-based vaccine induces protective cytotoxic T-lymphocyte-mediated antiviral immunity through dendritic cell maturation.

Currently used vaccines protect mainly through the production of neutralizing antibodies. However, antibodies confer little or no protection for a majority of chronic viral infections that require active involvement of cytotoxic T lymphocytes (CTLs). Virus-like particles (VLPs) have been shown to be efficient inducers of cell-mediated immune responses, but administration of an adjuvant is generally required. We recently reported the generation of a novel VLP system exploiting the self-assembly property of the papaya mosaic virus (PapMV) coat protein. We show here that uptake of PapMV-like part

Reorganization of Mitochondrial Function and Architecture in Response to Plant-Derived Alkaloids: Anatabine, Anabasine, and Nicotine, Investigated in SH-SY5Y Cells and in a Cellular Model of Parkinson's Disease.

AIMS: Nicotine, anatabine, and anabasine are the most prevalent alkaloids in Nicotiana species. While nicotine is the main addictive ingredient in tobacco products, it was also shown to have neuroprotective properties. Mitochondria appear to be one of the targets of nicotine in the cell. These multifunctional organelles are also the first responders to various cellular stresses. Thus, we characterized the impact of tobacco alkaloids on these organelles. METHODS: We investigated the effects of structurally similar alkaloids, anatabine, anabasine, and nicotine, on mitochondrial function in SH-S

Comparison on removal performance of virus, antibiotic-resistant bacteria, cell-associated and cell-free antibiotic resistance genes, and indicator chemicals by ozone in the filtrated secondary effluent of a sewage treatment plant.

Due to the widespread appearance of viruses, antibiotic-resistant bacteria (ARBs), and antibiotic resistance genes (ARGs) in the aquatic environment, more powerful oxidation processes such as ozonation are needed to enhance the efficiency of their inactivation and removal during wastewater treatment. However, information is lacking on the elimination rates of viruses, ARBs, cell-associated ARGs (ca-ARGs), and cell-free ARGs (cf-ARGs) during ozonation. This study examined the kinetics and dose-dependent inactivation of a virus (MS2 coliphage) and an ARB (Ampicillin-resistant [AmpR] E. coli) and

Identification of plant cells in black pigments of prehistoric Spanish Levantine rock art by means of a multi-analytical approach. A new method for social identity materialization using chaîne opératoire.

We present a new multi-analytical approach to the characterization of black pigments in Spanish Levantine rock art. This new protocol seeks … a primary raw material for pigment production, identified by means of the archaeobotanical study of plant cells. For the second goal, black charcoal pigments were replicated in the laboratory by using different raw materials and binders and by reproducing two main chaînes opératoires. The comparative study of the structure and preservation of plant tissues of …

Contribution of the putative clathrin adaptor AP-1 subunit β to cell wall integrity and virulence factor secretion in the plant pathogen Botrytis cinerea.

In filamentous fungi, the secretory pathway plays a key role in polarized growth by delivering vesicles carrying lipids and enzymes required for cell wall synthesis and nutrition. In pathogenic species, it also supports the delivery of virulence factors. In metazoa and yeast, secretory vesicle formation occurs at the Golgi apparatus and endosomes through a complex and still partially characterized machinery involving clathrin and adaptor proteins such as AP-1. In filamentous fungi, the molecular mechanisms controlling these processes remain poorly understood. Using a mutant underexpressing the

Extraction Strategy and C18 Solid-Phase Fractionation Shape Phenolic Profiles, Antioxidant Capacity, and Cancer Cell Antiproliferative Activity of Selected Medicinal Plants.

Medicinal plants are a rich source of biologically active compounds, including phenolic acids, flavonoids, ellagitannins and other secondary metabolites. However, the contribution of specific groups of phenolic compounds to antiproliferative activity remains insufficiently clarified. This study extends our previous crude-extract screening by evaluating whether C18 solid-phase extraction (SPE) fractions with different phenolic profiles are associated with different antiproliferative responses. In parallel, extraction strategies were compared to assess method-dependent changes in phenolic recove

Screening of promising chemotherapeutic candidates from plants against human adult T-cell leukaemia/lymphoma (X): withanolides from Physalis pruinosa.

Adult T-cell leukaemia/lymphoma (ATL) is a malignancy of mature peripheral T-lymphocytes caused by human T-cell lymphotropic virus type I (HTLV-I). Administering conventional chemotherapeutic regimens used against malignant lymphomas to patients with ATL results in poor therapeutic outcomes. Previously, we isolated withanolides from the aerial parts of Physalis pruinosa and observed potent activity against leukaemia cell lines. The most effective compound showed selective toxicity against HTLV-1-infected T-cell lines compared with normal peripheral blood mononuclear cells following analysis of
