Balancing act: The dynamic relationship between nutrient availability and plant defence.

Plants depend heavily on soil nutrients for growth, development and defence. Nutrient availability is crucial not only for sustaining vital biochemical processes but also for mounting effective defences against a diverse array of pathogens. Macronutrients such as nitrogen, phosphorus and potassium significantly influence plant defence mechanisms by providing essential building blocks for the synthesis of defence compounds, immune signalling and physiological responses like stomatal regulation. Micronutrients like zinc, copper and iron are essential for balancing reactive oxygen species and oth

Arabidopsis AtERF014 acts as a dual regulator that differentially modulates immunity against Pseudomonas syringae pv. tomato and Botrytis cinerea.

ERF transcription factors play critical roles in plant immune responses. Here, we report the function of AtERF014, a nucleus-localized transcriptional activator, in Arabidopsis immunity. Expression of AtERF014 was induced by Pseudomonas syringae pv. tomato (Pst) and Botrytis cinerea (Bc). AtERF014-overexpressing (OE) plants displayed increased Pst resistance but decreased Bc resistance, whereas AtERF014-RNAi plants exhibited decreased Pst resistance but increased Bc resistance. After Pst infection, expression of salicylic acid (SA)-responsive genes AtPR1 and AtPR5 in AtERF014-OE plants and of

Long-term evaluation of mucosal and systemic immunity and protection conferred by different polio booster vaccines.

Oral polio vaccine (OPV) and Inactivated Polio Vaccine (IPV) have distinct advantages and limitations. IPV does not provide mucosal immunity and introduction of IPV to mitigate consequences of circulating vaccine-derived polio virus from OPV has very limited effect on transmission and OPV campaigns are essential for interrupting wild polio virus transmission, even in developed countries with a high coverage of IPV and protected sewer systems. The problem is magnified in many countries with limited resources. Requirement of refrigeration for storage and transportation for both IPV and OPV is al

Transcriptome Analysis Reveals a Comprehensive Virus Resistance Response Mechanism in Pecan Infected by a Novel Badnavirus Pecan Virus.

Pecan leaf-variegated plant, which was infected with a novel badnavirus named pecan mosaic virus (PMV) detected by small RNA deep sequencing, is a vital model plant for studying the molecular mechanism of retaining green or chlorosis of virus-infected leaves. In this report, PMV infection in pecan leaves induced PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI). PMV infection suppressed the expressions of key genes of fatty acid, oleic acid (C18:1), and very-long-chain fatty acids (VLCFA) biosynthesis, indicating that fatty acids-derived signaling was one of the important def

Phytohormone-microbiome interactions and epigenetic regulation of plant stress responses and priming.

Plants continuously face fluctuating environmental conditions, requiring tightly coordinated regulatory systems to balance growth and stress responses. This review synthesizes current knowledge on phytohormones as central integrators of plant-microbiome interactions, highlighting their dual role as internal regulators and ecological gatekeepers that shape microbiome assembly and function. Phytohormones, such as auxins, cytokinins, gibberellins, abscisic acid, ethylene, salicylic acid, and jasmonates, dynamically regulate plant development, immunity, and rhizosphere chemistry, thereby influenci

Plant Peptide Hormones: Three Decades of Discovery and Emerging Paradigms.

Plant peptide hormones have reshaped our understanding of intercellular signaling in plants. Once considered absent in plants, peptide-based signaling pathways emerged as key regulators of plant growth, development, environmental responses and immunity. A paradigm shift occurred with the discoveries of Systemin and Phytosulfokine (PSK) in the 1990s as well as the identifications of receptor-like kinases (RLKs) regulating important biological processes. Over the past three decades, a repertoire of peptide hormone families has been expanded enormously, with deeper understandings of peptide modif

Unseen Struggles: How Plant-Parasitic Nematodes Manipulate ROS Signaling in Host Plants.

Reactive oxygen species (ROS) play a central role in plant defense, especially during interactions with plant-parasitic nematodes (PPNs). These molecules act as early signals that activate immune responses and help reinforce plant cell walls to block nematode invasion. However, PPNs have evolved specialized effector proteins (small, secreted molecules, typically proteins, that enter host cells to directly suppress immunity and manipulate host processes), which they secrete into host tissues and cells to interfere with ROS production and signaling. These effectors can suppress ROS bursts, detox

Canonical ETI-Dependent and -Independent Pathways Mediate Autoimmunity Caused by Loss of CBP60b Clade Function.

Calmodulin-Binding Protein 60 (CBP60), a plant-specific atypical transcription factor family, plays pivotal roles in reprogramming plant immunity. Loss of function of the CBP60b and CBP60b/c/d/e/f subfamily constitutively activates immune responses (autoimmunity). However, the specific mechanisms and pathways underlying this autoimmunity remain elusive. Here, through EMS mutagenesis of the cbp60b mutant, we isolated a dominant suppressor mutant that suppresses the cbp60b autoimmune phenotype. Sequencing identified a point mutation in CBP60g (designated cbp60g-3) that causes an aspartate-to-asp…

TGACG-BINDING FACTOR 1 (TGA1) and TGA4 regulate salicylic acid and pipecolic acid biosynthesis by modulating the expression of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g).

Salicylic acid (SA) and pipecolic acid (Pip) play important roles in plant immunity. Here we analyzed the roles of transcription factors TGACG-BINDING FACTOR 1 (TGA1) and TGA4 in regulating SA and Pip biosynthesis in Arabidopsis thaliana. We quantified the expression levels of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g), which encode two master transcription factors of plant immunity, and the accumulation of SA and Pip in tga1-1 tga4-1 mutant plants. We tested whether SARD1 and CBP60g are direct targets of TGA1 by chromatin immunoprecipitation-p…

Pipecolic Acid Orchestrates Plant Systemic Acquired Resistance and Defense Priming via Salicylic Acid-Dependent and -Independent Pathways.

We investigated the relationships of the two immune-regulatory plant metabolites, salicylic acid (SA) and pipecolic acid (Pip), in the establishment of plant systemic acquired resistance (SAR), SAR-associated defense priming, and basal immunity. Using SA-deficient sid2, Pip-deficient ald1, and sid2 ald1 plants deficient in both SA and Pip, we show that SA and Pip act both independently from each other and synergistically in Arabidopsis thaliana basal immunity to Pseudomonas syringae. Transcriptome analyses reveal that SAR establishment in Arabidopsis is characterized by a strong transcriptiona

The Small GTPase RHO1 Regulates ROS Homeostasis and Differentially Responds to Pathogens Infection via Interaction With Aquaporin TIP1;1 in Nicotiana.

Small G-proteins are crucial regulators in plant growth and environmental responses. However, the mechanistic basis underlying their regulation of plant immunity to different pathogens is still poorly understood. Here, we showed that the small GTPase Ras homologous 1 (RHO1) responded differently to infection by distinct pathogens. Overexpression of RHO1 increased the plant susceptibility to chilli veinal mottle virus (ChiVMV) and necrotrophic Botrytis cinerea infections but enhanced plant resistance to the biotrophic bacterium Pseudomonas syringae. Further exploration revealed that there exist…

Verticillium dahliae Secretory Aspartyl Protease VdSAP Targets Cotton GhARP to Modulate Plant Defence and Defoliation.

Verticillium dahliae isolates causing devastating vascular wilt in cotton plants can be divided into defoliating and nondefoliating pathotypes. The mechanisms underlying how V. dahliae uses secretory proteins to manipulate plant physiological processes and suppress immunity in cotton plants have received renewed research focus over the past several years. Here, we describe the role of a secretory protein named VdSAP (secretory aspartyl protease) from the defoliating V. dahliae strain XJ592 in virulence and defoliation. Deletion of VdSAP in strain XJ592 led to significantly reduced virulence an
