Gastrointestinal tract epithelial changes associated with taxanes: marker of drug toxicity versus effect.

BACKGROUND: Microscopic findings associated with paclitaxel (Taxol) chemotherapy toxicity were described years ago but whether they are specific for toxicity remains unclear. Further, epithelial changes associated with taxanes can mimic high-grade dysplasia (HGD) in non-neoplastic gastrointestinal (GI) tract mucosa. Similar changes associated with colchicine are only seen in patients with toxicity. METHODS: GI tract specimens were reviewed (221 total; 93 esophageal, 55 gastric cardiac, 48 oxyntic, 7 antral, 8 small bowel, 6 colonic, 3 appendiceal, 1 anal) from 71 patients (63M, 8F), 38 to 84

[A case of esophageal cancer patient who developed tumor lysis syndrome after chemotherapy].

We report here a case of esophageal cancer patient who developed tumor lysis syndrome after chemotherapy. A 57- year-old man received esophagectomy for advanced esophageal cancer. CT study revealed multiple lymphnode metastases, multiple bone metastases and multiple lung metastases after 15 months from the operation. So, chemotherapy was performed for the patient. Eleven days following the administration of docetaxel: 30 mg/m2 and nedaplatin: 30 mg/m2, the patient developed TLS (serum creatinine: 8.57 mg/dL, uric acid: 11.0 mg/dL, serum potassium: 6.3 mEq/L, serum phosphorus: 7.18 mg/dL). Docetaxel: a therapeutic option in the treatment of cutaneous angiosarcoma: report of 9 patients.

BACKGROUND: Effective treatment options are limited for patients with cutaneous angiosarcoma (AS). Docetaxel, a member of the taxane family of drugs, reportedly has been effective in the treatment of lung, head and neck, and breast cancers. Another taxane drug, paclitaxel, reportedly had unique activity in the treatment of AS of the scalp and neck and acquired immunodeficiency syndrome-related Kaposi sarcoma. Therefore, the authors hypothesized that docetaxel may be of value in the treatment of cutaneous AS that is resistant to conventional therapy. However, there were only 3 case reports of t

Prevention of chemotherapy-induced alopecia using an effective scalp cooling system.

Alopecia is a distressing side-effect of cancer treatment. Taxanes (TX), anthracyclines (ANR) and etoposide (ET) have been consistently associated with significant alopecia. We studied an effective scalp cooling system, the Penguin Cold Cap system, for the prevention of chemotherapy-induced alopecia in 70 patients receiving chemotherapy, including one of the following major alopecia-causing agents: Group A, TX-based regimes (without ANR); Group B, TX+ANR; Group C, ANR-based regimes (without TX); Group D, ET-based regimes. Protection from hair loss was achieved by maintaining scalp temperatures

Treatment Patterns and Attrition in Metastatic Castration-Resistant Prostate Cancer.

IMPORTANCE: Rapid expansion of the therapeutic landscape for metastatic castration-resistant prostate cancer (mCRPC) has increased the complexity of treatment selection and sequencing. However, large, contemporary studies describing treatment patterns and attrition rates in clinical treatment of mCRPC are lacking. OBJECTIVE: To investigate treatment patterns and attrition rates in patients with mCRPC from 2021 to 2025. DESIGN, SETTING, AND PARTICIPANTS: This retrospective multicenter cohort study used the US-based, electronic health record-derived, deidentified Flatiron Health Research Datab

CRYOTHERAPY FOR THE PREVENTION OF TAXANE-INDUCED PERIPHERAL NEUROPATHY IN BREAST CANCER: A PROSPECTIVE MULTICENTER STUDY.

BACKGROUND: Chemotherapy-induced peripheral neuropathy (CIPN) is a common dose-limiting toxicity of taxane-based chemotherapy in breast cancer. Cryotherapy using cooling gloves and socks has been investigated as a preventive strategy, although evidence remains inconsistent. This study evaluated the effect of preventive cryotherapy on neuropathy-related pain and symptom burden in patients receiving taxane-containing chemotherapy. METHODS: This prospective multicenter controlled study included patients with stage I-IV breast cancer treated with taxane-based chemotherapy between March 2023 and S

FROM INFUSION REACTION TO IMMUNE CASCADE: A CASE OF SEQUENTIAL TAXANE AND CAPECITABINE TOXICITIES IN TRIPLE-NEGATIVE BREAST CANCER.

This case report describes a 61-year-old female with stage IIIc triple-negative breast cancer (TNBC) who experienced a sequence of distinct treatment-related toxicities: a hypersensitivity reaction to paclitaxel, taxane-induced pneumonitis during subsequent docetaxel therapy, and radiation recall dermatitis (RRD) triggered by capecitabine administered after the completion of radiotherapy. The pneumonitis was successfully managed with corticosteroids, and the RRD resolved with topical therapy and dose modification. This case highlights the complex interplay of immune-mediated toxicities and und

Pembrolizumab plus lenvatinib combination therapy for advanced endometrial carcinoma.

INTRODUCTION: Advanced and recurrent endometrial carcinoma remains a difficult diagnosis to treat due to the limited and ineffective available treatment options following platinum and taxane chemotherapy. Patients who are microsatellite stable (MSS) or mismatch repair proficient (pMMR) have even poorer outcomes with fewer effective therapies. Fortunately, recent Phase Ib/II and Phase III trials have demonstrated that combination pembrolizumab and lenvatinib resulted in improved ORR, PFS, and OS than currently used therapies in this setting. AREAS COVERED: In this article, we review the histor

Investigation of Low Dose Cabazitaxel Potential as Microtubule Stabilizer in Experimental Model of Alzheimer's Disease: Restoring Neuronal Cytoskeleton.

BACKGROUND: Neuronal Microtubule (MT) tau protein, providing cytoskeleton to neuronal cells, plays a vital role, including maintenance of cell shape, intracellular transport, and cell division. Tau hyperphosphorylation mediated MT destabilization results in axonopathy, additionally neurotransmitter deficit and ultimately causing Alzheimer's disease. Pre-clinically, streptozotocin (3mg/kg, 10μl/ unilateral, ICV) stereotaxically mimics the behavioral and neurochemical alterations similar to Alzheimer's tau pathology resulting in MT assembly defects further lead to neuropathological cascades. Taxane-based chemotherapy and risk of breast cancer-related lymphedema: Protocol for a systematic review and meta-analysis.

BACKGROUND: Many studies were performed to explore the correlation between taxane-based chemotherapy and the risk of breast cancer-related lymphedema (BCRL), however, with inconsistent results. Hence, the purpose of this study is to evaluate whether taxane-based chemotherapy is a risk factor for BCRL. METHODS: A comprehensive systematic search of clinical trials published in the PubMed, Embase and the Cochrane Library databases will be conducted to identify eligible studies up to the date of December 31, 2018. We will employ risk ratios with 95% confidence intervals (95% CIs) to estimate the

Single-eye trial of a topical carbonic anhydrase inhibitor versus intravitreal bevacizumab for the treatment of taxane drug-induced cystoid macula oedema.

Taxanes are a class of microtubule stabilising agents used to treat a wide range of malignancies. Taxane drug-induced cystoid macula oedema (TDICMO) is a known but rare complication of therapy. First reported with Docetaxel in 2003 and Paclitaxel in 2007, there are currently less than 20 cases of TDICMO in the literature. Although most cases resolve following taxane cessation, several authors have tried using carbonic anhydrase inhibitors or intravitreal bevacizumab to accelerate resolution or when taxane therapy cannot be discontinued. We report the first published case of TDICMO treated with
