The Untapped Opportunities of Addressing Lifecourse Growth in Mammographic Breast Density and Breast Cancer Prevention in Non-Hispanic Black Women.

This commentary discusses the importance of adiposity and lifecourse growth with mammographic breast density (MBD) in non-Hispanic Black (NHB) women. Although high MBD is a well-established risk factor for breast cancer, the determinants of MBD in NHB women remain understudied. Although adiposity at the time of mammography is most strongly associated with MBD, adiposity as early as ages 10 and 18 years is also independently associated with MBD. The commentary emphasizes the need for future research to identify biological mechanisms underlying the associations of adulthood adiposity and lifecou

Mammographic breast density and breast cancer risk in the Saudi population: a case-control study using visual and automated methods.

OBJECTIVE: This study aims to establish risk of breast cancer based on breast density among Saudi women and to compare cancer prediction using different breast density methods. METHODS: 1140 pseudonymised screening mammograms from Saudi females were retrospectively collected. Breast density was assessed using Breast Imaging Reporting and Data System (BI-RADS) density categories and visual analogue scale (VAS) of 285 cases and 855 controls matched on age and body mass index. In a subset of 160 cases and 480 controls density was estimated by two automated methods, Volpara Density™ and predicted

Studies of parenchymal texture added to mammographic breast density and risk of breast cancer: a systematic review of the methods used in the literature.

This systematic review aimed to assess the methods used to classify mammographic breast parenchymal features in relation to the prediction of future breast cancer. The databases including Medline (Ovid) 1946-, Embase.com 1947-, CINAHL Plus 1937-, Scopus 1823-, Cochrane Library (including CENTRAL), and Clinicaltrials.gov were searched through October 2021 to extract published articles in English describing the relationship of parenchymal texture features with the risk of breast cancer. Twenty-eight articles published since 2016 were included in the final review. The identification of parenchyma

Mammographic breast density and breast cancer risk in a Mediterranean population: a nested case-control study in the EPIC Florence cohort.

PURPOSE: Mammographic breast density (MBD) has been consistently associated with breast cancer (BC) risk, and at the same time it is modulated by established BC risk factors related to reproductive and hormonal history and to lifestyle. We aimed to evaluate the association between the clinical breast imaging reporting and data system (BI-RADS), a qualitative MBD classification used in clinical setting, and BC risk through a case-control nested in the EPIC Florence cohort where baseline information on reproductive history, lifestyle and anthropometry were collected. METHODS: The study includes

Using clinical factors and mammographic breast density to estimate breast cancer risk: development and validation of a new predictive model.

BACKGROUND: Current models for assessing breast cancer risk are complex and do not include breast density, a strong risk factor for breast cancer that is routinely reported with mammography. OBJECTIVE: To develop and validate an easy-to-use breast cancer risk prediction model that includes breast density. DESIGN: Empirical model based on Surveillance, Epidemiology, and End Results incidence, and relative hazards from a prospective cohort. SETTING: Screening mammography sites participating in the Breast Cancer Surveillance Consortium. PATIENTS: 1,095,484 women undergoing mammography who had

Global mammographic asymmetry and short-term breast cancer risk by breast density: a nationwide screening cohort of 5.5 million women.

Global mammographic asymmetry (GA) is generally considered benign, and its association with subsequent breast cancer risk is unclear. We examined whether GA on screening mammography predicts short-term and long-term breast cancer and whether this varies by Breast Imaging Reporting and Data System (BI-RADS) breast density. In this retrospective cohort study using the Korean National Health Insurance Service screening programme, we included women aged ≥40 years who underwent screening mammography in 2009-2010 and had no prior breast cancer. GA and BI-RADS density were recorded on baseline mammog

Comparison between software volumetric breast density estimates in breast tomosynthesis and digital mammography images in a large public screening cohort.

OBJECTIVES: To compare software estimates of volumetric breast density (VBD) based on breast tomosynthesis (BT) projections to those based on digital mammography (DM) images in a large screening cohort, the Malmö Breast Tomosynthesis Screening Trial (MBTST). METHODS: DM and BT images of 9909 women (enrolled 2010-2015) were retrospectively analysed with prototype software to estimate VBD. Software calculation is based on a physics model of the image acquisition process and incorporates the effect of masking in DM based on accumulated dense tissue areas. VBD (continuously and categorically) was

Differential detection by breast density for digital breast tomosynthesis versus digital mammography population screening: a systematic review and meta-analysis.

BACKGROUND: We examined whether digital breast tomosynthesis (DBT) detects differentially in high- or low-density screens. METHODS: We searched six databases (2009-2020) for studies comparing DBT and digital mammography (DM), and reporting cancer detection rate (CDR) and/or recall rate by breast density. Meta-analysis was performed to pool incremental CDR and recall rate for DBT (versus DM) for high- and low-density (dichotomised based on BI-RADS) and within-study differences in incremental estimates between high- and low-density. Screening settings (European/US) were compared. RESULTS: Pool

Adjuvant tamoxifen-induced mammographic breast density reduction as a predictor for recurrence in estrogen receptor-positive premenopausal breast cancer patients.

Tamoxifen is known to reduce the risk of breast cancer in women at high risk and also reduces mammographic breast density (MD) in a preventive setting. We investigated the efficacy of MD reduction (MDR) for predicting recurrence in estrogen receptor (ER)-positive patients in an adjuvant setting. A total of 1,066 ER-positive breast cancer patients who were enrolled in this study underwent curative surgery and received adjuvant tamoxifen for at least 2 years at our institution between January 2003 and December 2008. Using a computerized system, a single radiologist reviewed all mammograms and cl

DensiThAI: A Multi-View Deep Learning Framework for Breast Density Estimation Using Infrared Images.

Breast tissue density is an established biomarker of breast cancer risk and an important determinant of mammographic sensitivity. Density assessment is typically performed using X-ray mammography. In this study, we investigate whether breast tissue density-related information may be reflected in infrared thermal images using artificial intelligence. The underlying hypothesis is that fibroglandular and adipose tissues differ in thermophysical and physiological properties, potentially giving rise to subtle surface temperature patterns. We propose DensiThAI, a multi-view deep learning framework t

Paclitaxel prodrug loaded high-density lipoprotein nanoparticles for chemoimmunotherapy of triple-negative breast cancer.

Insufficient tumor-specific immunogenicity and cytotoxic T lymphocytes infiltration limit the therapeutic efficacy of combined chemotherapy and immune checkpoint blockade for triple-negative breast cancer (TNBC). To address this limitation, we developed a paclitaxel prodrug-loaded synthetic high-density lipoprotein nanoparticle (P-sHDL) that both targets tumors and induces immunogenic cell death (ICD) to potentiate immune checkpoint blockade. Leveraging scavenger receptor class B type 1-mediated uptake, P-sHDL efficiently accumulated at tumor sites and triggered ICD in tumor cells, thereby enh

Semiautomated magnetic resonance imaging-based breast density measurement from routine sequences: comparison with mammography.

AIM: High breast density increases breast cancer risk and decreases mammographic sensitivity, yet prior magnetic resonance imaging (MRI)-based density assessments have relied on specialized sequences or vendor-dependent software. This study aimed to evaluate a semiautomated workflow for MRI-based breast density measurement using routinely acquired MRI sequences and compare its diagnostic performance and concordance with visual and software-based mammographic classifications. MATERIALS AND METHODS: In this single-center retrospective study, we analyzed 113 women (mean age: 60.8 ± 13.6 years) w
