A large-scale procedure for purification and concentration of Japanese encephalitis virus vaccine by a continuous-flow isopycnic banding technique in a sucrose density gradient solution, using a K-III zonal centrifuge rotor, is presented. The quality of zonal-purified vaccine was compared with commercial and Japanese National Institutes of Health reference vaccines for antigenicity, immunodiffusion, and allergic encephalitogenicity tests to show its high purity.

111In-oxine "pure" granulocyte scanning allows an exact localisation of infiltrated bowel in Crohn's disease and ulcerative colitis. Fecal excretion of labeled granulocytes yields a good parameter of disease activity. In comparison to 111In-oxine "mixed" leukocyte scanning, this method is more specific in assessing disease activity and more sensitive in localising diseased bowel. Radiation exposure to blood cells is not as high as in "mixed" leukocyte scanning. Pure granulocytes were isolated by discontinuous density gradient centrifugation of an isolated "mixed" leukocyte fraction.

Platelets of patients with thrombocytosis following splenectomy, in chronic granulocytic leukaemia and in polycythaemia vera were separated into five fractions by centrifugation in discontinuous Ficoll density gradient. Platelet volume, content of protein and enzyme activities of lactic dehydrogenase, phosphoglycerate kinase and glyceraldehyde phosphate dehydrogenase were distinctly higher for the three groups in the heavy fraction IV compared with the light fraction I. With regard to the platelet volume, however, these differences were compensated almost completely like in the normal persons.

OBJECTIVE: To evaluate pregnancy rates (PRs) in women undergoing artificial insemination with sperm alternately prepared by one of five techniques: sperm washing, Percoll gradient centrifugation, swim-up, swim-down, or refrigeration/heparin treatment.
