Development of silk fibroin/collagen film containing GI-20 peptide-loaded PLGA nanoparticles against corneal herpes simplex virus-1.

Herpes simplex virus-1 (HSV-1) is the primary cause of infectious blindness. Despite impressive therapeutic outcomes of conventional treatments, HSV-1 drug resistance can be easily developed. Thus, more constructive strategies should be implemented. Led by this inspiration, this work describes the potential utility of a biodegradable silk fibroin/collagen (SF/Col) film combined with GI-20-loaded poly lactic-co-glycolic acid (PLGA) nanoparticle to provide efficient and sustained delivery platform for synthetic GI-20 peptide against HSV-1. A non-irritant film containing 90 % SF and 10 % Col inco

Unveiling the synergy: Biocompatible alginate-cellulose hydrogel loaded with silk fibroin and zinc ferrite nanoparticles for enhanced cell adhesion, and anti-biofilm activity.

Combining a biocompatible hydrogel scaffold with the cell-supportive properties of silk fibroin (SF) and the unique functionalities of ZnFe2O4 nanoparticles creates a promising platform for advanced nanobiomaterials. The research is centered on synthesizing a natural hydrogel using cellulose (Cellul) and sodium alginate (SA) combined with SF and zinc ferrite nanoparticles. A range of analytical and biological assays were conducted to determine the biological and physicochemical properties of the nanobiocomposite. The hemolysis and 2,5-diphenyl-2H-tetrazolium bromide (MTT) assays indicated that

Beta-cyclodextrin modified mesoporous bioactive glass nanoparticles/silk fibroin hybrid nanofibers as an implantable estradiol delivery system for the potential treatment of osteoporosis.

Osteoporosis, a systemic skeletal disease prevalent in elderly women, is associated with post-menopausal estrogen deficiency. Although systemic administration of exogenous estradiol (E2) reduced fragility fractures, the treatment has adverse effects. Localized delivery technologies of E2 could be utilized to circumvent the systemic adverse effects of systemic administration. In this study, a localized E2 delivery system is developed. Mesoporous bioactive glass nanoparticles (MBGNPs) with inherent osteogenic properties are modified with β-cyclodextrin (CD-MBGNPs) to enhance their affinity for E

Fabrication Scheme for Obtaining Transparent, Flexible, and Water-Insoluble Silk Films from Apparently Dissolved Silk-Gland Fibroin of Bombyx mori Silkworm.

Films from silk fibroin protein are one of the most promising biomaterials because of their exquisite balance between mechanical properties and biocompatibility. Numerous schemes have been proposed for processing fibroin film, utilizing liquid silk fibroin (LSF) or regenerated silk fibroin (RSF). The films cast from LSF or RSF in the solution state are water-soluble, and therefore require postproduction treatment inducing β-sheet formation, to render them insoluble in water. Many kinds of postproduction treatments, using alcohol-water solution, water vapor, or controlled temperature, have been

Performance Evaluation of Highly Uniform Astragalus Polysaccharide/Silk Fibroin Microspheres Fabricated via Microfluidics as Biodegradable Fillers: Collagen Regeneration and Tissue Reaction.

Skin aging results from the synergistic assault of extrinsic environmental factors and intrinsic biological processes. While traditional microsphere implants promote skin rejuvenation through collagen regeneration, their clinical utility is hampered by non-biodegradability and risks such as granuloma formation. To overcome these limitations, this study employed microfluidic technology to develop highly uniform, novel injectable, highly bioactive, and degradable Astragalus polysaccharide/silk fibroin (APS/SF) composite microspheres for subcutaneous filling to promote collagen regeneration. Overcoming silk fibroin electrospinning limitations: ultra-high surface area nanofibers and microfilms through ethyl cellulose-assisted high-speed spinning.

Silk fibroin (SF) is a fundamental building block for the development of advanced biomaterials. SF nanofibers are ideal for industrial and biomedical applications due to their excellent biocompatibility, mechanical strength, and tunable biodegradability. However, conventional electrospinning suffers from jet instability, low fiber yield, and poor spinnability, which restricts its performance and scalability. In this study, we present a high-speed, controllable electrospinning process coupled with a simple, environmentally friendly post-treatment using ethanol. SF nanofibers were produced by bl

Injectable Mn-Icariin-functionalized silk fibroin/PEG hydrogels restore redox homeostasis and reprogram osteogenic-angiogenic coupling for diabetic bone regeneration.

Bone defect repair under hyperglycemic conditions is limited by sustained oxidative stress, chronic low-grade inflammation and disrupted osteoimmune-vascular coupling. In this study, we constructed an injectable and biodegradable silk fibroin/PEG hydrogel incorporating MnO2 honeycomb nanospheres and liposome-encapsulated icariin (FG/Mn@Ica) to rebuild a bioactive and mechanocompatible microenvironment for diabetic bone regeneration. The hydrogel presented an interconnected porous morphology, elastic-dominant rheology, adaptive compressive behavior and controlled degradation with the sustained

Enhanced pharmacokinetic approach for anastrozole via macromolecule-based silk fibroin nanoparticles incorporated in situ injectables for oestrogen-positive breast cancer therapy.

Breast cancer (BC) is a substantial reason for cancer-related mortality among women across the globe. Anastrozole (ANS) is an effective orally administered hormonal therapy for oestrogen-positive (ER+) BC treatment. However, several side effects and pharmacokinetic limitations restricted the uses of ANS in BC therapy. Therefore, this investigation developed an in situ gelling injectable-loaded silk fibroin (SF)-ANS NPs, which offers sustained drug release and improved pharmacokinetic properties compared to conventional oral formulations at the targeted site. The optimised in situ gel (ISG) inc

Crosslinker-free silk fibroin/κ-carrageenan bioinks incorporating RGD-functionalized, cell-laden microcarriers for 3D cartilage bioprinting: Formulation and cytocompatibility.

One of the key challenges in 3D bioprinting is developing bioinks that ensure both structural integrity and biocompatibility. This study introduces a novel strategy that combines microcarrier technology with 3D bioprinting to improve cell viability and mechanical strength without using toxic crosslinkers, focusing on cartilage tissue engineering. Poly(butylene adipate-co-terephthalate) (PBAT) microcarriers (∼100 μm) were functionalized with RGD peptides to enhance cell attachment and seeded with ATDC5 chondroprogenitor cells. These were incorporated into bioinks composed of silk fibroin (SF, 2

Disrupting Mycobacterium smegmatis biofilm using enzyme-immobilized rifampicin loaded silk fibroin nanoparticles for dual anti-bacterial and anti-biofilm action.

Biofilm formation is a major challenge in the treatment of tuberculosis, leading to poor treatment outcomes and latent infections. The complex and dense extracellular polymeric substances (EPS) of the biofilm provides safe harbour for bacterium enabling persistence against anti-TB antibiotics. In this study, we demonstrated that rifampicin-encapsulated silk fibroin nanoparticles immobilized with antibiofilm enzymes can disrupt the Mycobacterium smegmatis biofilm and facilitate the anti-bacterial action of Rifampicin (RIF). The EPS of M.smegmatis biofilm predominantly comprised of lipids (48.8

An interference screw made using a silk fibroin-based bulk material with high content of hydroxyapatite for anterior cruciate ligament reconstruction in a rabbit model.

Upgradation is still in need for the clinically applied interference screws in anterior cruciate ligament reconstruction for more reliable fixation. Silk fibroin bulk materials offer a promising opportunity for this application except lacking osteoinductivity to some extent. Here we report a novel silk-based bulk material with high content of hydroxyapatite-silk fibroin (HA-SF) hybrid particles, which is prepared via a dual-network hydrogel. This composite bulk material possesses a compression modulus of 3.2 GPa, comparable to that of the natural compact bone, and presents satisfactory cytocom

Intratesticular Peptidyl Prolyl Isomerase 1 Protein Delivery Using Cationic Lipid-Coated Fibroin Nanoparticle Complexes Rescues Male Infertility in Mice.

Male infertility is a multifactorial condition. Unexplained male infertility is often caused by spermatogenesis dysfunction. Knockout of Pin1, an important regulator of cell proliferation and differentiation, produces male infertility phenotypes such as testicular immaturity and azoospermia with spermatogonia depletion and blood-testis barrier (BTB) dysfunction. Gene therapy has been clinically considered for the treatment of male infertility, but it is not preferred because of the risks of adverse effects in germ cells. Direct intracellular protein delivery using nanoparticles is considered a

Sustained Release of TPCA-1 from Silk Fibroin Hydrogels Preserves Keratocyte Phenotype and Promotes Corneal Regeneration by Inhibiting Interleukin-1β Signaling.

Corneal injury due to ocular trauma or infection is one of the most challenging vision impairing pathologies that exists. Many studies focus on the pro-inflammatory and pro-angiogenic effects of interleukin-1β (IL-1β) on corneal wound healing. However, the effect of IL-1β on keratocyte phenotype and corneal repair, as well as the underlying mechanisms, is not clear. This study reports, for the first time, that IL-1β induces phenotype changes of keratocytes in vitro, by significantly down-regulating the gene and protein expression levels of keratocyte markers (Keratocan, Lumican, Aldh3a1 and CD

Improving glycemic control in model mice with type 2 diabetes by increasing superoxide dismutase (SOD) activity using silk fibroin hydrolysate (SFH).

Islet cell dysfunction in type 2 diabetes is primarily attributed to the increased apoptosis of pancreatic beta cells. Silk fibroin hydrolysate (SFH) has an effect on blood in type 2 diabetes model mice (C57BL/KsJ-db/db). However, its exact mechanism is unknown. The type 2 diabetes model mice were randomly divided into non-diabetic mice (ND), diabetic mice (DB), and diabetic mice treated with silk fibroin hydrolysate (DB-SFH). The results showed that SFH significantly decreased fasting blood glucose and hemoglobin A1c (HbA1c). The oral glucose tolerance and insulin tolerance were significantly

Silk fibroin scaffolds promote formation of the ex vivo niche for salivary gland epithelial cell growth, matrix formation, and retention of differentiated function.

Salivary gland hypofunction often results from a number of causes, including the use of various medications, radiation for head and neck tumors, autoimmune diseases, diabetes, and aging. Since treatments for this condition are lacking and adult salivary glands have little regenerative capacity, there is a need for cell-based therapies to restore salivary gland function. Development of these treatment strategies requires the establishment of a system that is capable of replicating the salivary gland cell "niche" to support the proliferation and differentiation of salivary gland progenitor cells

Bioprinting of transparent and adhesive corneal patches: Integrating photo-crosslinkable dopamine-conjugated silk fibroin and decellularized cornea matrix for sutureless tissue integration and regeneration.

Corneal injuries, a leading cause of visual impairment, are traditionally addressed through tissue transplantation. However, challenges such as donor shortages, graft rejection, and complications from suturing often limit their effectiveness. Current corneal adhesives frequently fall short in both adhesion strength and biocompatibility. We present an innovative solution: a photocurable hydrogel that integrates dopamine-conjugated methacrylated silk fibroin (d-MSF) with a decellularized corneal matrix (DCM). This hydrogel combines advanced materials to create a bioadhesive system that offers su
