Paeonol ameliorates postmenopausal hyperlipidemia by inhibiting COX-2 to suppress GRP78-mediated endoplasmic reticulum stress.

Paeonol (Pae), a primary bioactive constituent of the root of Paeonia suffruticosa Andrews, shows therapeutic potential for postmenopausal hyperlipidemia. Ovariectomized ApoE-/- mice fed a high-fat diet and HepG2 cells with palmitate-induced steatosis were employed to evaluate the effects of Pae on lipid metabolism and elucidate the underlying mechanisms. Integrated computational and biophysical approaches established cyclooxygenase-2 (COX-2) as a high-affinity target of Pae, evidenced by a high molecular docking score (-6.3 kcal/mol), stable binding dynamics in molecular dynamics simulations,

Machine learning constructs a ferroptosis related signature for predicting prognosis and drug sensitivity in lung cancer.

PURPOSE: Ferroptosis is a novel form of iron-dependent programmed cell death that is associated with the progression of various tumors and cancer treatment responses. However, its role in the clinical treatment of lung cancer, particularly in lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC), remains poorly understood. This study aims to explore the prognostic value of ferroptosis-related genes in lung cancer and establish a reliable predictive model. METHODS: We collected the GSE4573, TCGA-LUAD, and TCGA-LUSC datasets, comprising a total of 1,271 samples and a ferroptosis ge

Late Recurrence of Breast Cancer Nearly 30 Years After Surgery: Mediastinal and Hilar Lymph Node Metastases Diagnosed Using Endobronchial Ultrasound-Guided Transbronchial Needle Aspiration.

Late recurrence of breast cancer occurring more than two decades after curative surgery is rare, but in the era of prolonged survival, clinicians may increasingly encounter such cases. Mediastinal or hilar recurrence may radiologically mimic primary lung cancer; however, histological confirmation using endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) enables accurate diagnosis with minimal invasiveness. We report the cases of two patients who developed mediastinal and hilar lymph node metastases nearly 30 years after breast cancer surgery, for whom EBUS-TBNA played

17-β-estradiol and phytoestrogens elicit NO production and vasodilatation through PI3K, PKA and EGF receptors pathways, evidencing functional selectivity.

Endothelial cells express multiple receptors mediating estrogen responses; including the G protein-coupled estrogen receptor (GPER). Past studies on nitric oxide (NO) production elicited by estrogens raised the question whether 17-β-estradiol (E2) and natural phytoestrogens activate equivalent mechanisms. We hypothesized that E2 and phytoestrogens elicit NO production via coupling to distinct intracellular pathways signalling. To this aim, perfusion of E2 and phytoestrogens to the precontracted rat mesentery bed examined vasorelaxation, while fluorescence microscopy on primary endothelial cell

Cutaneous toxicities occurring during palbociclib (CDK4/6 inhibitor) and endocrine therapy in patients with advanced breast cancer: a single-centre experience.

PURPOSE: Treatment with Palbociclib, a cyclin-dependent kinase 4/6 inhibitor, has demonstrated significantly improved progression-free survival in patients with hormone receptor-positive, HER2-negative, advanced breast cancer, when used in combination with letrozole or fulvestrant endocrine therapies. However, limited information exists on its cutaneous adverse effects (AE). Hence, we conducted a retrospective cohort study to investigate the prevalence and management of cutaneous AE during palbociclib and endocrine therapy. METHOD: We included 324 adult patients with advanced breast cancer wh

Oestrogenic activity of benzyl salicylate, benzyl benzoate and butylphenylmethylpropional (Lilial) in MCF7 human breast cancer cells in vitro.

Benzyl salicylate, benzyl benzoate and butylphenylmethylpropional (Lilial) are added to bodycare cosmetics used around the human breast. We report here that all three compounds possess oestrogenic activity in assays using the oestrogen-responsive MCF7 human breast cancer cell line. At 3 000 000-fold molar excess, they were able to partially displace [(3)H]oestradiol from recombinant human oestrogen receptors ERalpha and ERbeta, and from cytosolic ER of MCF7 cells. At concentrations in the range of 5 x 10(-5) to 5 x 10(-4 )m, they were able to increase the expression of a stably integrated oest

The selective estrogen receptor modulator bazedoxifene inhibits hormone-independent breast cancer cell growth and down-regulates estrogen receptor α and cyclin D1.

Bazedoxifene (BZA) is a third-generation selective estrogen receptor modulator (SERM) that has been approved for the prevention and treatment of postmenopausal osteoporosis. It has antitumor activity; however, its mechanism of action remains unclear. In the present study, we characterized the effects of BZA and several other SERMs on the proliferation of hormone-dependent MCF-7 and T47D breast cancer cells and hormone-independent MCF-7:5C and MCF-7:2A cells and examined its mechanism of action in these cells. We found that all of the SERMs inhibited the growth of MCF-7, T47D, and MCF-7:2A cell
