Natural Product Inspired N-Terminal Hsp90 Inhibitors: From Bench to Bedside?

The 90 kDa heat shock proteins (Hsp90) are responsible for the conformational maturation of nascent polypeptides and the rematuration of denatured proteins. Proteins dependent upon Hsp90 are associated with all six hallmarks of cancer. Upon Hsp90 inhibition, protein substrates are degraded via the ubiquitin-proteasome pathway. Consequentially, inhibition of Hsp90 offers a therapeutic opportunity for the treatment of cancer. Natural product inhibitors of Hsp90 have been identified in vitro, which have served as leads for the development of more efficacious inhibitors and analogs that have enter

Special method to prepare quantum dot probes with reduced cytotoxicity and increased optical property.

Quantum dots (QDs) are widely used in the life sciences because of their novel physicochemical properties. However, the cytotoxity of these nonoparticles have attracted great attention recently because this has not been well resolved. Four probes were synthesized by chemical coupling and protein denaturation with CdSeZnS, CdTe QDs, and transferrin. Sodium dodecyl sulfate polyacrylamide gel electrophoresis and capillary electrophoresis were used to verify the conjugation of these luminescent probes. The cytotoxicity of these four luminescent probes and the original QDs were evaluated in HeLa ce…

Efficient in vitro refolding and characterization of a new peptide from the scorpion Buthotus saulcyi venom produced in Escherichia coli.

The selective toxicity of depressant scorpion neurotoxins to insects is useful in studying the insect sodium channel gating, as well as being relevant to several other applications. In order to carry out structure/activity studies, the functional expression of such polypeptides is required. In the work reported here, the cDNA of a new peptide from the venom of the scorpion Buthotus saulcyi was cloned and sequenced. It codes for a 64 residues peptide (BsaulI) with 8 highly-conserved cysteines. This peptide shares high sequence similarity with depressant insect toxins of other scorpion species.

Variation of CI-2 Conformers upon Addition of Methanol to Water: An IMS-MS-Based Thermodynamic Analysis.

Chymotrypsin inhibitor 2 (CI-2) is a well-studied, textbook example of a cooperative, two-state, native ↔ denatured folding transition. A recent hybrid ion mobility spectrometry (IMS)/mass spectrometry (MS) thermal denaturation study of CI-2 (the well-studied truncated 64-residue model) in water reported evidence that this two-state transition involves numerous (∼41) unique native and non-native (denatured) solution conformations. The characterization of so many, often low-abundance, states is possible because of the very high dynamic range of IMS-MS measurements of ionic species that are prod

Differential oxidation of surimi actomyosin by malondialdehyde and heme: spectroscopy and molecular simulation.

Pro-oxidative factors compromise surimi gel quality by inducing protein denaturation and aggregation. This study elucidated the interactions between malondialdehyde (MDA) and hemin with surimi actomyosin through multispectral analysis and molecular dynamics (MD) simulations. The results suggested that MDA and hemin induced actomyosin oxidation by altering protein conformation, surface hydrophobicity, and the microenvironment of aromatic residues, especially under low-pH/high-temperature conditions. The MD results confirmed that MDA weakly interacted with myosin owing to its flexible backbone a…

Fabrication and characterization of whey protein-tannic acid-ferrous ion complexes with improved iron stability and bioavailability.

Conventional food iron fortification strategies face great challenges due to the instability and lower bioavailability of Fe2+. This study fabricated a whey protein isolate-tannic acid-ferrous ion (WTF) complex and systematically investigated its formation mechanism and characteristics. The results indicated that tannic acid non-covalently bound to whey protein, and Fe2+ synergistically promoted the structural reorganization. This interaction led to a decrease in protein α-helix content accompanied by an increase in the proportion of β-structure, along with a decrease in thermal denaturation t

A multistep platform identifies spleen-tropic lipid nanoparticles for in vivo T cell-targeted delivery of gene-editing proteins.

Lipid nanoparticles (LNPs) are a promising nonviral delivery system for gene-editing proteins, but optimal formulations remain underexplored. Unlike messenger RNA-based approaches, ribonucleoprotein delivery enables immediate genome editing without relying on endogenous translation. However, intracellular delivery remains a major challenge due to protein size, charge variability, and susceptibility to denaturation and degradation. Here, we present a multistep screening platform to optimize LNP formulations for gene-editing protein delivery, focusing on in vivo T cell targeting. Through in vitr

A magnetic bifunctional endotoxin removal nano-agent for the efficient elimination of endotoxins in recombinant protein preparation.

Endotoxin contamination, as one of the most significant challenges in recombinant protein production by Escherichia coli, represents a critical biosafety concern and greatly hinders the biomedical application of recombinant proteins. Conventional methods, such as extreme-condition inactivation and chromatography-based separation, are plagued by issues including protein denaturation, low efficiency, and operational complexity in endotoxin removal. In this work, we developed a novel magnetic bifunctional endotoxin removal nano-agent (MagBER) with a multi-layered structure, consisting of a superp…

Molecular insights into freeze-induced quality deterioration in surimi: Mechanisms and mitigation strategies.

Surimi, a concentrated myofibrillar protein product derived from fish muscle, serves as a fundamental raw material in a wide range of processed aquatic foods. Although freezing and frozen storage are essential for ensuring microbial safety and extending shelf life, it inevitably leads to quality deterioration. This review critically examines the molecular mechanisms underlying freeze-induced quality degradation in surimi, emphasizing the roles of ice crystal formation, protein denaturation, and lipid oxidation. Furthermore, recent advances in novel technologies, including liquid nitrogen freez…

High early post-mortem temperature induces activation of AMP-activated protein kinase and development of pale, soft and exudative characteristics in turkey muscles.

This study investigated the effect of early post-mortem temperature on development of a turkey muscle's pale, soft and exudative (PSE) characteristics. Muscles obtained at 20 min post-mortem were incubated at 0, 20 and 40 °C until 4 h post-mortem and then stored at 4 °C. During incubation, the 40 °C group had greater rate of pH decline, lactate accumulation and R-values increase than other groups. Moreover, AMP-activated protein kinase (AMPK) activation at 1h post-mortem was higher in the 40 °C group than the other groups. At 24 h post-mortem, the 40 °C group had higher L* values and drip loss

Molecular modelling indicates that the pathological conformations of prion proteins might be beta-helical.

Creutzfeldt-Jakob disease, kuru, scrapie and bovine spongiform encephalopathy are diseases of the mammalian central nervous system that involve the conversion of a cellular protein into an insoluble extracellular isoform. Spectroscopic studies have shown that the precursor protein contains mainly alpha-helical and random-coil conformations, whereas the prion isoform is largely in the beta conformation. The pathogenic prion is resistant to denaturation and protease digestion and can promote the conversion of the precursor protein to the pathogenic form. These properties have yet to be explained

Concerted Deoligomerization and Nonfunctional Reassembly of the Hexameric Proteasomal ATPase Mpa upon Chemical and Thermal Perturbation.

Mycobacterium tuberculosis Mpa is an AAA+ ATPase that unfolds and translocates substrate proteins during proteasomal degradation. Mpa spontaneously assembles into a homohexamer driven by interdomain interactions, even in the absence of a nucleotide. To dissect the mechanisms underlying its oligomerization and deoligomerization, we perturbed the system using chemical and thermal denaturants and monitored the oligomeric states by far-UV CD, fluorescence, calorimetry, state-of-the-art single-particle mass photometry, and ATPase activity assays. Equilibrium chemical denaturation resulted in gradua

Temperature-dependent structural changes of Parkinson's alpha-synuclein reveal the role of pre-existing oligomers in alpha-synuclein fibrillization.

Amyloid fibrils of α-synuclein are the main constituent of Lewy bodies deposited in substantial nigra of Parkinson's disease brains. α-Synuclein is an intrinsically disordered protein lacking compact secondary and tertiary structures. To enhance the understanding of its structure and function relationship, we utilized temperature treatment to study α-synuclein conformational changes and the subsequent effects. We found that after 1 hr of high temperature pretreatment, >80°C, α-synuclein fibrillization was significantly inhibited. However, the temperature melting coupled with circular dichroism

Intrinsic fluorescent detection of tau conformation and aggregation.

The polymerization of the microtubule-associated protein tau into paired helical filaments (PHFs) is one of the hallmarks of Alzheimer's disease. Insights into the prerequisites and kinetics of the polymerization was obtained by the in vitro analysis of this process. In the past, fluorescent dyes were used to stain amyloidogenic material in histology and later on similar dyes were used in in vitro studies as well. To circumvent the flaws of extragenous dyes, namely the alteration of the polymerization kinetic or incompatibility with other chemical compounds needed for stability analysis, we ap

Enhancing lactoferrin stability in ethanol aqueous systems through oligosaccharide modulation: Mechanisms and functional implications.

To improve protein stability in ethanol aqueous systems, this study investigated the effects of three oligosaccharides-oligofructose (FOS), oligogalactose (GOS), and oligoxylose (XOS)-on the conformational and thermal stability of lactoferrin (LF). Results demonstrated that these oligosaccharides significantly increased LF's thermal denaturation temperature (Tm) and elevated the energy barrier for conformational changes in ethanol aqueous systems, thereby enhancing thermostability. Specifically, FOS, GOS, and XOS raised the Tm of LF from 69.1 ± 0.4 °C to 79.8 ± 1.3 °C, 73.7 ± 0.6 °C, and 76.8

Crowder Environments Control Interfacial Dynamics and Topological Stability in a Knotted Protein.

Knotted proteins are unique biomolecules where backbone entanglement imposes intrinsic constraints on structure and dynamics. The interaction of such a topologically restrained protein with molecular crowders remains unexplored. Using molecular dynamics simulations, we investigate the effects of polyethylene glycol (PEG) and dextran on the structure, interactions, and topological dynamics of the trefoil-knotted enzyme AOTCase. Persistent, heterogeneous accumulation of dextran near the protein surface is observed, whereas PEG exhibits a transient interaction pattern. The distinct interfacial be

Fish sarcoplasmic proteins as a high value marine material for wound dressing applications.

Fish sarcoplasmic proteins (FSP) constitute around 25-30% of the total fish muscle protein. As the FSP are water soluble, FSP were isolated from fresh cod (Gadus morhua) by centrifugation. By SDS-PAGE, it was possible to determine the composition of FSP extracts (FSP-E). The FSP-E undergo denaturation at 44.12 ± 2.34° C, as characterized by differential scanning calorimetry thermograms (DSC). The secondary structure of FSP-E is mainly composed by α-helix structure, as determined by circular dichroism. The cytocompatibility of FSP-E, at concentrations ranging from 5 to 20 mg/mL, was investigate
