Universal cryo-thermal sculpting of transition metal dichalcogenides: simultaneous phase and size engineering for energy electrocatalysis.

The global quest for a sustainable energy matrix is fundamentally a crisis of material constraints where success relies on structural control at the atomic scale. Herein, we report a universal, physical protocol: cyclic rapid annealing and cooling (CRAC) that simultaneously orchestrates phase-engineering and dimensional reduction. By subjecting bulk TMDs (MoS2, MoSe2, and CoSe2) to extreme, repetitive cryo-thermal shocks, we induce a controlled structural fracturing that yields zero-dimensional (0D) nanoparticles with locked metastable phases (1T/cubic) through rapid kinetic quenching. Decoding MOF-Enzyme-Substrate Compatibility for Universal Biocatalyst Immobilization.

Metal-organic frameworks (MOFs) are widely used for enzyme immobilization due to their tunable pore structures and high stability. However, whether a relatively universal MOF carrier can support diverse enzymatic reactions remains unclear. Herein, we systematically evaluated 120 MOF-enzyme-substrate combinations using ten representative MOFs, three hydrolases (CRL, CALB, and BsEst), and four ester substrates, and then extended the identified carriers to three additional enzymes spanning catalase, oxidase, and protease reactions (GOx, Typ, and BlCAT), examining each with a single substrate. Universal CAR-T cell therapy: mechanisms and evasion strategies of bidirectional immune rejection.

While autologous chimeric antigen receptor (CAR)-T cell therapies have revolutionized the treatment of hematological malignancies, their widespread clinical application remains constrained by lengthy manufacturing cycles, prohibitive costs, and variable patient-derived cell quality. Consequently, "off-the-shelf" universal allogeneic CAR-T cells have emerged as a highly anticipated alternative. However, the clinical translation of these allogeneic therapies faces formidable immunological bottlenecks, principally driven by bidirectional immune rejection: graft-versus-host disease (GVHD) and host

Water-Stable MIL-MOFs Developed Through a Novel Sacrifice-Protection Strategy Inspired by Butterfly Wings' Scales for Long-Term Turn-On Fluorescence Sensing of H2S.

Metal-organic frameworks, which are the desired candidates for biosensing application due to their tunable properties, are significantly hindered by their rapid degradation in aqueous solutions, as well as the loss of their inherent fluorescence. Most studies aim to improve the hydrophobicity of materials by modifying their contact angle, thereby enhancing water stability. However, water droplets poorly adhere to the surface of hydrophobic materials, limiting their application for direct contact and detection in aqueous environments. Drawing inspiration from the sacrificial protection mechanis

Dissecting immunological mechanisms underlying influenza viral nucleoprotein-induced mucosal immunity against diverse viral strains.

The nucleoprotein (NP) of type A influenza virus (IAV) is highly conserved across all virus strains, making it an attractive candidate antigen for universal vaccines. While various studies have explored NP-induced mucosal immunity, here we interrogated the mechanistic differences between intramuscular (IM) and intranasal (IN) delivery of a recombinant adenovirus carrying NP fused with a bifunctional CD40 ligand. Despite being less effective than IM delivery in inducing systemic cellular immune responses and antibody-dependent cellular cytotoxicity (ADCC), IN immunization elicited superior anti

Cost-Effective Modular Biosensor for SARS-CoV-2 and Influenza A Detection.

A modular, multi-purpose, and cost-effective electrochemical biosensor based on a five-stranded four-way junction (5S-4WJ) system was developed for SARS-CoV-2 (genes S and N) and Influenza A virus (gene M) detection. The 5S-4WJ structure consists of an electrode-immobilized universal stem-loop (USL) strand, two auxiliary DNA strands, and a universal methylene blue redox strand (UMeB). This design allows for the detection of specific nucleic acid sequences using square wave voltammetry (SWV). The sequence-specific auxiliary DNA strands (m and f) ensure selectivity of the biosensor for target re

Universal anti-influenza vaccines based on viral HA2 and M2e antigens.

Aquatic birds are the main reservoir of influenza A viruses (IAVs). These viruses can infect humans repeatedly and cause acute respiratory disease with potential of spread in the form of epidemics. In addition, avian influenza viruses that overcome the interspecies barrier and adapt to humans can cause a world-wide pandemic with severe consequences to human health. Therefore, scientists are focused on the development of a "universal" vaccine with a broad protective efficacy, i.e. against different subtypes of influenza A viruses and not only against the currently co-circulating human epidemic

Risk factors for admission to hospital with laboratory-confirmed influenza in young children: birth cohort study.

We determined risk factors for influenza hospital admission in children aged <2 years to guide the design of paediatric vaccination programmes. We linked all singleton live births in Scotland from 2007 to 2015 to hospital administrative data and influenza laboratory reports. Cox proportional hazard models were used to identify birth and family risk factors for influenza admissions. There were 1115 influenza admissions among 424 048 children. 85.1% of admitted children were born at term and were not in a high-risk group. Presence of an older sibling was strongly associated with increased risk of

Double-end blocker and split input mediated CRISPR-Cas12a system for modular single-base mutation detection with low abundance.

Traditional CRISPR-Cas12a mutation detection systems are limited by poor single-base specificity, target-specific crRNA redesign, and insufficient sensitivity for low-abundance mutations, restricting their clinical liquid biopsy applications. Herein, we developed a crRNA-universal, sensitive and specific CRISPR-Cas12a detection platform, termed DESIC (double-end blocker and split-input mediated CRISPR-Cas12a system), for single-base mutation detection. The DESIC system adopts two key structural designs: double-end blocker (DEB) and duplicated split-input (SIN). The DEB spatially isolates crRNA

Development of a standardized routine workflow for entomological and toxicological analysis using larvae of two forensically relevant fly species (Diptera, Calliphoridae).

For more than four decades, no standardized study design or workflow has been established for systematic entomotoxicological research in forensic casework. This study aimed to develop a universal rearing protocol for fly larvae grown on drug-spiked tissue surrogate and a standardized workflow for the qualitative analysis of pharmacologically active compounds (PACs) in insects of different developmental stages. Larvae of Protophormia terraenovae and Lucilia sericata were reared on minced meat serving as tissue surrogate. First-instar larvae were transferred to minced meat spiked with diazepam,

Universal Fully Integrated Wearable Sensor Arrays for the Multiple Electrolyte and Metabolite Monitoring in Raw Sweat, Saliva, or Urine.

Fully integrated wearable sensors are capable of dynamically, directly, and independently tracking biomarkers in raw noninvasive biofluids without any other equipment or accessories by integrating the unique on-body monitoring feature with the special complete functional implementation attribute. Sweat, saliva, and urine are three important noninvasive biofluids, and changes in their biomarkers hold great potential for revealing physiological conditions. However, it is still a challenge to design single fully integrated wearable sensor arrays (FIWSAs) that are universally able to concurrently

A qualitative assessment of a dog program for youth offenders in an adult prison.

OBJECTIVE: Dog programs occurring in prison tend to generate positive public opinion and media portrayals, yet they often lack empirical validation. This is the first study to examine the key themes that drive a dog program aimed at incarcerated youth serving time in an adult prison. DESIGN AND SAMPLE: A sample of male youth (n = 31) were provided survey questions on a rescue dog program. A grounded qualitative approach was employed with open-ended responses. RESULTS: The following themes emerged: the symbolism of the dog its role in therapy, perceptions of positive behaviors and rehabilitat

Potential costs and benefits of newborn screening for severe combined immunodeficiency.

OBJECTIVE: Severe combined immunodeficiency (SCID) is a rare, treatable disorder of the immune system. The incidence is unknown but may be more common than published estimates because infants frequently die of infection before diagnosis. SCID is a candidate for universal newborn screening, so there is a need to determine under which circumstances screening would be cost-effective. STUDY DESIGN: We assumed a screening program for SCID would use T-cell lymphopenia as the screening criterion and performed a cost-utility analysis comparing universal screening with screening only those with a fami

OBJECTIVES: In the clinical laboratory, identification of Streptococcus pneumoniae can be confused with other streptococci. Conventional biochemical tests such as optochin sensitivity and bile solubility can give inconsistent results. This report presents a method to distinguish true S. pneumoniae from other upper respiratory tract streptococci when conventional tests fail. DESIGN AND METHODS: We used arbitrarily primed polymerase chain reaction with the single primer M13 universal as a method to distinguish S. pneumoniae from other upper respiratory tract streptococci. RESULTS: The fingerpr
