Serum IgA antibodies to three Aspergillus fumigatus antigen fractions were determined by enzyme-linked immunosorbent assay (ELISA) in 13 patients with aspergilloma, 23 patients with aspergillus lung infiltrates, 8 patients with allergic broncho-pulmonary aspergillosis (ABPA), and 113 healthy blood donors. All three groups of patients had higher median serum IgA antibody levels against the three antigen fractions than healthy subjects. Thirty six (82%) of 44 patients had IgA antibody values to at least one antigen fraction above the 95 percentile of the blood donor group. Patients with aspergil

A procedure was developed to type California serogroup viruses by an antibody-capture, enzyme-linked immunosorbent assay. Seven California serogroup members from North America were distinguished, including snowshoe hare, La Crosse, California encephalitis, San Angelo, Jamestown Canyon, Keystone, and trivittatus. Extensive cross-reactions were observed between Jamestown Canyon and the closely related South River strain. The enzyme-linked immunosorbent assay method was successfully applied to the typing of 77 California serogroup viruses isolated in Canada, including 61 snowshoe hare, 13 Jamesto

A semiautomated, kinetic-dependent, enzyme-linked immunosorbent assay (K-ELISA) was adapted to detect serum antibodies to Legionella pneumophila. In a comparative study, 158 human serum samples (79 pairs) were tested by K-ELISA and the standard indirect immunofluorescence assay for determination of antibody levels to L. pneumophila serogroup 1. K-ELISA determinations were made by using a serogroup-specific antigen or a preparation (unfractionated antigen) which contained both common antigen and serogroup-specific reactivity. There was good correlation between the immunofluorescence assay and t

An enzyme-linked immunosorbent assay (ELISA) was developed for determination of the IgA, IgM and IgG antibody responses against a lipopolysaccharide antigen representing Shigella sonnei phase I bacteria. Two or more sera from 33 patients infected with Shigella sonnei were collected during a 12 month period after onset of the disease. Convalescent sera from 56 patients with other enteric infections (salmonellosis, yersiniosis, campylobacteriosis) and sera drawn from 40 healthy blood donors served as controls. Twenty-eight of the 33 patients (85%) had at least one serum specimen where two or thr

An enzyme-linked immunosorbent assay (ELISA) for the detection of antibody to cysticerci of Taenia solium has been developed that employs a pork muscle antigen control for the cysticercus test antigen, somewhat improving the serological distinction between infected and uninfected subjects. Serum antibody to cysticercus was detected in 79% of classical neurocysticercosis patients from Mexico, and in 61% of a group of cysticercosis patients with an unusually rapid invasion of the central nervous system in an endemic focus of disease in Irian Jaya. Antibody was absent in a group of healthy Americ

The kinetics of the release of a liver enzyme, glutamate dehydrogenase, into the serum of Fasciola hepatica infected rats was studied. The results support previous work that has shown that barriers to reinfection with this parasite are met before any pronounced liver damage takes place. The presence of this enzyme in serum can be used as an indicator of resistance to reinfection.

An immunoenzymatic diagnostic technique applicable to cutaneous leishmaniasis is described. The antigen used (Leishmania tropica major) was equally useful in diagnosing visceral and mucocutaneous forms of the disease. The criteria for positivity were defined by using groups of negative controls, and the specificity of the reaction was evaluated by using sera from patients with various diseases. Among these, sera from patients with lepromatous leprosy, tuberculosis, or African trypanosomiasis strongly cross-reacted with leishmania antigen. Examining serial dilutions of the sera facilitated the

Antibody levels in a small group of Sudanese patients with clinically diagnosed mycetoma, and control groups were measured by countercurrent immunoelectrophoresis (CIE) and enzyme linked immunosorbent assay (ELISA). Antigens were prepared from the following organisms: Streptomyces somaliensis, Actinomadura madurae, A. pelletieri, Nocardia brasiliensis and Madurella mycetomi. Positive reactions were obtained in clinical cases against the homologous antigens in both CIE and ELISA; all heterologous and control sera negative in CIE but produced some reaction particularly against N. brasiliensis an

We previously described a reverse transcriptase-PCR using flavivirus genus-conserved and virus species-specific amplimers (D. W. Trent and G. J. Chang, p. 355-371, in Y. Becker and C. Darai; ed., Frontiers of Virology, vol. 1, 1992). Target amplification was improved by redesigning the amplimers, and a sensitive enzyme-linked immunosorbent assay (ELISA) technique has been developed to detect amplified digoxigenin (DIG)-modified DNA. A single biotin motif and multiple DIG motifs were incorporated into each amplicon, which permitted amplicon capture by a biotin-streptavidin interaction and detec

BACKGROUND: Thyroid disorders affect millions of people around the world, mainly women. Measurement of hormones is critical in their identification, treatment, and monitoring; Therefore, the methods must be as reliable and valid as possible.
METHODS: We designed a cross-sectional study to compare thyroid-stimulating hormone (TSH) and free tetraiodothyronine (fT4) concentrations using ELISA (Enzyme-Linked Immunosorbent Assay) and LFA (Lateral Flow Assay) in human serum samples. We obtained 96 serum samples that were evaluated with a commercial kit for ELISA (Accubind Monobite, Lake Forest, Cal

Two rapid, sensitive immunoassays based on monoclonal antibody for detecting clothianidin were developed and applied in agricultural samples: a quantitative enzyme-linked immunosorbent assay (ELISA) and a semiquantitative gold immunochromatographic assay (GICA). Under optimal conditions, the half-maximal inhibition concentration (IC50) and the limit of detection (LOD, IC10) of clothianidin were 25.6 and 3.8 ng mL(-1) for ELISA. GICA using colloidal gold-MAb probe had a visual detection limit of 8 ng mL(-1), and the results can be judged by the naked eye within 10 min. The cross-reactivities of

Toscana virus (TOSV) is an arthropod-borne virus, transmitted to humans by Phlebotomus spp. Sandflies, which causes neurological diseases such as aseptic meningitis and meningoencephalitis. The commercial enzyme-linked immunosorbent assay (ELISA) is used widely to detect anti-TOSV IgG and IgM antibodies and to allow for rapid diagnosis of infection (Diesse Diagnostica Senese, Siena, Italy). Recently, an immunochromatographic assay (ICA) was developed for human anti-TOSV IgG or IgM detection by InBios International (Seattle, WA, USA). A comparison of the two diagnostic assays was performed on o

In placental tissue, human and rat aromatases convert testosterone to estradiol, which was prioritized for investigation over others due to its susceptibility to endocrine disruption by mycotoxins, yet their effects remain unclear. The objective of this study is to investigate the effects of mycotoxins on human and rat aromatase, including inhibition strength, mechanism, and structural-activity relationships (SAR). Screening was conducted to evaluate mycotoxin inhibition using placental microsomes by HPLC-MS/MS. IC50 and Ki values were determined as well as effects on estradiol secretion in hu

In the event of a radiological incident, a fast and accurate biological dosimetry (biodosimetry) method for evaluating people who have been potentially exposed to ionising radiation is crucial. Among the many biodosimetry methods available, the immunodetection of phosphorylated H2AX (γ-H2AX) stands as a promising method to be used in the triage of patients exposed to radiation. Currently, the most common way to measure γ-H2AX levels is through fluorescence microscopy. In this pilot study, we assessed the feasibility of using an enzyme-linked immunosorbent assay (ELISA) for quantifying γ-H2AX f
