[Serological and Genotypic Analysis of a Family with B(A) Subtype].

OBJECTIVE: To perform blood group typing and genotyping on the proband with weak ABO antigen expression and his family members, thereby clarifying the ABO phenotype and the molecular genetic basis of the weak antigen expression. METHODS: Serological ABO blood group typing was performed on the proband and four family members. PCR amplification of exons 6 and 7 of the ABO gene was carried out followed by direct sequencing analysis, and haplotypes were further confirmed by clonal sequencing. RESULTS: Direct sequencing results revealed a c.640A>G single-nucleotide heterozygous mutation in exon 7

Treatment of Acute Hemolytic Transfusion Reaction due to ABO-Incompatible Red Blood Cell Transfusion: A Case Report, Review of the Literature and Recommendation.

Although acute hemolytic transfusion reaction (AHTR) due to ABO-incompatible red blood cell transfusion is a rare complication in high-income countries, it is considered a medical emergency because of the severe intravascular hemolysis that can lead to major morbidity or death. AHTR can cause intravascular hemolysis, acute renal failure, disseminated intravascular coagulation (DIC), and shock, and therefore treatment should be prompted early and directed to prevent these complications. We present a case of AHTR due to ABO-incompatible red blood cell transfusion successfully treated with high-d

Pure red cell aplasia after major or bidirectional ABO incompatible hematopoietic stem cell transplantation: to treat or not to treat, that is the question.

Pure red cell aplasia (PRCA) is a complication related to major or bidirectional ABO mismatched hematopoietic stem cell transplantation. This disorder is characterized by anemia, reticulocytopenia, and the absence or virtual absence of erythroid progenitors, other causes such as infections, hemolysis, disease relapse, or drug toxicity having been excluded. Patients with PRCA may become RBC transfusion dependent for long periods, suffering an important long-term iron overload, alloimmunization, and transfusion reactions. The persistence of recipient isoagglutinins against donor ABO antigens pro

Acute Intravascular Hemolysis Following an ABO Non-Identical Platelet Transfusion: A Case Report and Literature Review.

BACKGROUND Platelet transfusion is a common clinical practice required for therapeutic purposes in the setting of symptomatic thrombocytopenia, and, in some cases, prophylactically for asymptomatic thrombocytopenia. Crossmatch compatibility is not routinely done for platelet transfusions, and transfusion of ABO non-identical platelets has been adapted as an acceptable clinical practice. Acute intravascular hemolysis due to ABO non-identical platelets is a rare but clinically significant entity. Our case report reinforces the importance of a vigilant clinical approach in case of ABO non-identic

Seasonal variability is not observed in the rates of high anti-A and anti-B titers in plasma, apheresis platelet, and whole blood units tested by different methods.

BACKGROUND: ABO-incompatible platelet transfusions are common, and transfusions with ABO-incompatible plasma are increasing with the use of group A plasma and group O whole blood (WB) in emergencies. Many centers screen blood products for anti-A and/or anti-B titers to help prevent hemolysis from ABO-incompatible transfusions, yet titer methods and definition of high titers are not standardized. STUDY DESIGN AND METHODS: This international multicenter study collected data on anti-A and anti-B titer practices for plasma, apheresis platelet (AP), and WB units from January 2015 through December

An expeditious synthesis of blood-group antigens, ABO histo-blood group type II antigens and xenoantigen oligosaccharides with amino type spacer-arms.

Blood group oligosaccharides are one of the most clinically important antigen families and they may also act as secondary ligands for bacterial toxins from Escherichia coli and Vibrio cholerae. Herein we report the synthesis of spacered (sp = CH2CH2CH2NH2) glycosides of A antigen {α-D-GalNAc-(l→3)-[α-L-Fuc-(l→2)]-β-D-Gal-}, B antigen{α-D-Gal-(l→3)-[α-L-Fuc-(l→2)]-β-D-Gal-}, LewisX{α-D-Gal-(l→4)-[α-L-Fuc-(l→3)]-β-D-GlcNAc-}, A type-II {α-D-GalNAc-(l→3)-[α-L-Fuc-(l→2)]-β-D-Gal-(1→4)-β-D-GlcNAc-}, B type-II {α-D-Gal-(l→3)-[α-L-Fuc-(l→2)]-β-D-Gal-(1→4)-β-D-GlcNAc-}, H type-II{α-L-Fuc-(l→2)-β-D-Gal

Neonatal haemochromatosis with reversible pituitary involvement.

Neonatal haemochromatosis is a rare alloimmune gestational disease with a high mortality. The hallmark of neonatal haemochromatosis is severe neonatal liver failure associated with extrahepatic siderosis. Thus far, no pituitary dysfunction has been reported to result from the tissue damage associated with extrahepatic siderosis. The present report describes a neonate with neonatal haemochromatosis and secondary hypothyroidism associated with pituitary iron deposition. Both the conditions were successfully treated by ABO-incompatible liver transplantation. Pituitary gland dysfunction is another

Delayed intravascular haemolysis following multiple asymptomatic ABO-incompatible red blood cell transfusions in a patient with hepatic failure.

ABO-incompatible red blood cell (RBC) transfusions have rarely been associated with delayed haemolysis. However, we report the case of a 75-year-old man (blood type O) with hepatic disease, who received 5 units of incompatible type B RBCs over 8 days. The patient did not develop symptomatic or biochemical evidence of haemolysis until 7-8 days after the first incompatible RBC unit. The patient had a low anti-B antibody titre (1 : 64) prior to the first transfusion. The onset of haemolysis was temporally associated with an increase in anti-B and the infusion of fresh-frozen plasma. In conclusion

Renal transplantation across the ABO barrier.

Some patients with end-stage renal disease do not have suitable living related donors due to ABO incompatibility. Crossing the ABO barrier has long been considered an absolute contraindication to kidney transplantation. Although transplantation using A2 kidneys has been performed in a few centers over the past two decades with variable success, the presence of blood group antigens can cause immediate or early graft loss due to preformed antibody-mediated hyperacute rejection. Over the past few years, novel protocols designed to reduce anti-donor antibodies have allowed successful transplantati

Acalculous cholecystitis in bone marrow transplant patients.

BACKGROUND: Acalculous cholecystitis (ACC) is an uncommon disorder of the biliary tract, accounting for approximately 6% of acute cholecystitis cases. In this study, cholecystitis was seen in 8 of 770 bone marrow transplant recipients, with ACC occurring in five (63%). METHODS: Records of 592 allogenic and 150 autologous BMT patients were reviewed for risk factors associated with ACC. RESULTS: Only the number of blood transfusions administered and the use of total parenteral nutrition were associated with ACC development. ACC occurred in 4 of 42 (9%) allogeneic recipients who required exchan

High throughput human genotyping for variants associated with malarial disease outcomes using custom targeted amplicon sequencing.

Malaria has exhibited the strongest known selective pressure on the human genome in recent history and is the evolutionary driving force behind genetic conditions, such as sickle-cell disease, glucose-6-phosphatase deficiency, and some other erythrocyte defects. Genomic studies (e.g., The 1000 Genomes project) have provided an invaluable baseline for human genetics, but with an estimated two thousand ethno-linguistic groups thought to exist across the African continent, our understanding of the genetic differences between indigenous populations and their implications on disease is still limite
