Biosynthesis, enzymatic basis, and transcriptional regulation of benzylisoquinoline alkaloids in medicinal plants.

Benzylisoquinoline alkaloids (BIAs) are a class of structurally diverse secondary metabolites, with over 2,500 identified variants classified into seven major categories based on their core skeletons. These compounds exhibit significant pharmacological activities - including antitumor, antibacterial, and anti-inflammatory effects - and serve as important sources of clinical drugs such as morphine, berberine, and magnoflorine. Currently, BIAs are primarily obtained through plant cultivation and extraction. However, factors such as low in-planta content, seasonal constraints, and environmental v…

Liquid chromatography-tandem mass spectrometry analysis of pyrrolizidine alkaloids in Polish honey.

INTRODUCTION: Pyrrolizidine alkaloids (PAs) are among the most widespread naturally occurring toxins. Since they are primarily produced by flowering plants, there is a high likelihood of their transfer into honey. Given the toxic properties of PAs, their presence in honey can significantly compromise the safety of the product. Therefore, monitoring the occurrence of PAs in honey is essential. MATERIAL AND METHODS: A total of 65 honey samples collected in Poland were analysed for the presence of 42 PAs. The samples included multifloral, acacia, rapeseed and forest honeys. Pyrrolizidine alkaloi…

Divergent Total Syntheses of Bisnicalaterine Alkaloids Enabled by a Stereocontrolled Geissoschizol Synthesis.

Bisnicalaterine alkaloids are structurally complex and biologically significant bisindole natural products. Herein, we report the first enantioselective total syntheses of bisnicalaterines B, C, and their N4' epimers using an eight-step longest linear sequence. Central to this strategy is a site- and stereoselective bioinspired Friedel-Crafts-type coupling between the eburnamine and geissoschizol fragment, enabling a highly selective formation of the bisindole core. The geissoschizol framework is efficiently assembled using a stereocontrolled Bischler-Napieralski cyclization/reduction sequence…

The underexplored pulmonary toxicity of pyrrolizidine alkaloids: Emerging hazard from food contamination.

Pyrrolizidine alkaloids (PAs) are the most widespread plant-derived toxins globally. Humans are frequently exposed to PAs through ingestion of PA-contaminated food products, with chronic dietary exposure estimated at up to 48.4 ng/kg body weight/day, posing a significant global health threat. Aside from the well-documented hepatotoxicity, dietary exposure to PAs is strongly implicated in the development of pulmonary arterial hypertension (PAH), a serious and often fatal disease with limited treatment options. However, the underlying molecular mechanisms remain poorly defined. This review provi…

Fungal ergot alkaloids: Metabolic pathways, biological functions, and advances in synthetic reprogramming.

Ergot alkaloids (EAs) are a class of secondary metabolites produced by fungi. These compounds are predominantly synthesized by Ascomycota, with variations in types and biosynthetic pathways among different fungal species. The EA synthesis has minimal impact on the normal growth and development of most EA-producing fungi, but serves as a virulence factor that influences the biocontrol functions of entomopathogenic fungi and symbiotic fungi in plants. In the medical field, EAs have been widely used for treating neurological disorders such as Parkinson's disease. However, the biosynthetic pathway…

Microemulsion-based drug delivery system identifies pepper alkaloids as anti-obesity compounds.

Obesity is a significant contributor to various metabolic diseases such as heart disease and diabetes. Due to the adverse effects of synthetic anti-obesity drugs, natural products from functional food plants, which mimic the effects of synthetic chemicals, present promising alternatives. However, many natural plant-derived compounds are poorly soluble in water, resulting in low bioavailability within the gastrointestinal tract, a key limitation for the effectiveness of many hydrophobic substances. In this study we developed a microemulsion-based drug delivery system in Drosophila, which effect

Targeting erythrocyte-mediated hypoxia to alleviate lung injury induced by pyrrolizidine alkaloids.

Pyrrolizidine alkaloids (PAs) are widely distributed natural toxins and have been extensively studied for their hepatotoxicity. However, PA-induced pulmonary toxicity remains less studied regarding the initiating mechanism and treatment approaches. Our previous study demonstrated the formation of pyrrole-hemoglobin adducts after PA exposure in vivo, which is suspected to affect the oxygen-carrying capacity of erythrocytes [red blood cells (RBCs)] consequently. The present study aimed to investigate the effects of PAs on the oxygen-carrying capacity of RBCs and the potential of targeting RBC-me…
