Interacting short-term regulatory mechanisms enable the conversion of light energy to chemical energy in photosynthesis.

Photosynthesis is a complex sequence of physical, electrochemical, biochemical, and physiological processes that convert light energy and carbon dioxide into sugars. These sugars then provide the energy and carbon backbone for all metabolic pathways involved in plant growth and development. However, if light energy is not managed effectively within the thylakoid membrane, it can destroy the photosynthetic apparatus in an oxygenic environment generated by photosynthesis itself. Effective photoprotection requires a variety of partially overlapping regulatory mechanisms that control energy and el

A sonosensitive diphenylalanine-based broad-spectrum antimicrobial peptide.

The antimicrobial effect of antimicrobial peptides is typically slow; they can be rapidly biodegraded and often have non-selective toxicity and elaborate sequences. Here we report a short peptide that is activated by ultrasound, that shows high broad-spectrum antibacterial efficiency (>99%) against clinically isolated methicillin-resistant bacteria (specifically, Staphylococcus aureus, Escherichia coli, Staphylococcus epidermidis, Enterobacter cancerogenus and Pseudomonas aeruginosa) with 15 min of ultrasound irradiation, and that has negligible toxicity and low self-antibacterial activity. We

Double Electrochemiluminescence Quenching-Based "Signal-on/off" Immunosensor for CD44 Detection Using Glucose Oxidase-Functionalized CuFe2O4 and Ti3C2Tx@Ni(OH)2-Ru/TPrA Nanoarrays.

The rapid and accurate quantification of CD44 in serum is crucial for the early detection and prevention of malignant tumors. In this study, we developed a "signal-on/off" electrochemiluminescence (ECL) immunosensor for the ultrasensitive detection of CD44, leveraging the dual quenching effects of CuFe2O4-NH2-Pd-GOD on Ti3C2Tx@Ni(OH)2-Ru/TPrA nanoarrays. A novel electroactive and catalytic multilayered nanoarray, Ti3C2Tx@Ni(OH)2, was first synthesized and employed as an efficient signal amplifier to accelerate the ECL response by promoting the reaction between Ru(bpy)32+ and tri-n-propylamine

Mitochondria in corneal physiology and pathology: A mechanistic perspective.

The cornea is the eye's outermost protective and refractive barrier whose physiological homeostasis depends on mitochondrial competence and dynamic stability. Mitochondrial dysfunctions are characterized by impaired electron transport chain efficiency, excessive reactive oxygen species generation, calcium dysregulation, and disrupted dynamics. Clinically, mitochondrial dysfunctions underlie the pathogenesis of diverse corneal disorders, including Fuchs endothelial corneal dystrophy, dry eye disease, diabetic keratopathy, keratoconus and infectious keratitis. Although prior work has linked mito

Synaptic wiring motifs in posterior parietal cortex support decision-making.

The posterior parietal cortex exhibits choice-selective activity during perceptual decision-making tasks1-10. However, it is not known how this selective activity arises from the underlying synaptic connectivity. Here we combined virtual-reality behaviour, two-photon calcium imaging, high-throughput electron microscopy and circuit modelling to analyse how synaptic connectivity between neurons in the posterior parietal cortex relates to their selective activity. We found that excitatory pyramidal neurons preferentially target inhibitory interneurons with the same selectivity. In turn, inhibitor

Triphenylene-Based 2D cMOFs: Unraveling the H2S Sensing Mechanism and Applications for a Real-Time Wireless Chemiresistive Sensor.

Two-dimensional conductive metal-organic frameworks (2D cMOFs) stand at the forefront of chemiresistive sensing innovations due to their high surface areas, distinctive morphologies, and substantial electronic conductivity. Particularly, 2D cMOFs crafted using 2,3,6,7,10,11-hexahydroxytriphenylene (HHTP) and 2,3,6,7,10,11-hexaiminotriphenylene (HITP) organic ligands have garnered a large amount of attention due to their designable active sites and proper conductive characteristics. Nevertheless, a deeper exploration into their sensing mechanisms is imperative for a comprehensive understanding

Cooperative amplification of Prussian blue as a signal indicator and functionalized metal-organic framework-based electrochemical biosensor for an ultrasensitive HE4 assay.

Human epididymis protein 4 (HE4), a diagnostic biomarker of ovarian cancer, is crucial for monitoring the early stage of the disease. Hence, it is highly important to develop simple, inexpensive, and user-friendly biosensors for sensitive and quantitative HE4 assays. Herein, a new sandwich-type electrochemical immunosensor based on Prussian blue (PB) as a signal indicator and functionalized metal-organic framework nanocompositesas efficient signal amplifiers was fabricated for quantitative analysis of HE4. In principle, ketjen black (KB) and AuNPs modified on TiMOF (TiMOF-KB@AuNPs) could accel

Quaternary structure of patient-homogenate amplified α-synuclein fibrils modulates seeding of endogenous α-synuclein.

Parkinson's disease (PD) and Multiple System Atrophy (MSA) are progressive and unremitting neurological diseases that are neuropathologically characterized by α-synuclein inclusions. Increasing evidence supports the aggregation of α-synuclein in specific brain areas early in the disease course, followed by the spreading of α-synuclein pathology to multiple brain regions. However, little is known about how the structure of α-synuclein fibrils influence its ability to seed endogenous α-synuclein in recipient cells. Here, we aggregated α-synuclein by seeding with homogenates of PD- and MSA-confir

Highly sensitive photoelectrochemical neuron specific enolase analysis based on cerium and silver Co-Doped Sb2WO6.

A signal-enhanced photoelectrochemical immunoassay technique for detecting neuron specific enolase (NSE) was proposed. As a photoactive matrix, (Ce,Ag):Sb2WO6 was firstly investigated via doping Ce and Ag into Sb2WO6. It could be found that the presence of Ce and Ag not only had enormous variation on the morphology of Sb2WO6, but also showed excellent PEC behavior. In order to further improve the visible light utilization rate of (Ce,Ag):Sb2WO6, In2S3 was modified onto the surface of (Ce,Ag):Sb2WO6 to enhance visible light absorption. In addition, the CdS/PDA was served as a secondary antibody

Ultrastructural evidence for an unusual mode of ciliogenesis in mouse multiciliated epithelia.

Multiciliogenesis is a cascading process for generating hundreds of motile cilia in single cells. In vertebrates, this process has been investigated in the ependyma of brain ventricles and the ciliated epithelia of the airway and oviduct. Although the early steps to amplify centrioles have been characterized in molecular detail, subsequent steps to establish multicilia have been relatively overlooked. Here, we focused on unusual cilia-related structures previously observed in wild-type mouse ependyma using transmission electron microscopy and analyzed their ultrastructural features and the fre

Boosting the Photodynamic Degradation of Islet Amyloid Polypeptide Aggregates Via a "Bait-Hook-Devastate" Strategy.

Photosensitizers that can generate reactive oxygen species (ROS) upon irradiation have emerged as promising agents for photodynamic degradation of toxic amyloid aggregates that are linked to many amyloidogenic diseases. However, due to the ultrastable β-sheet structure in amyloid aggregates and inefficient utilization of the generated ROS, it usually requires high stoichiometric concentration of the photosensitizer and/or intensive light irradiation to fully dissociate aggregates. In this work, we have developed a "bait-hook-devastate" strategy to boost the efficiency of the photodynamic degra

A regenerative microchannel neural interface for recording from and stimulating peripheral axons in vivo.

Neural interfaces are implanted devices that couple the nervous system to electronic circuitry. They are intended for long term use to control assistive technologies such as muscle stimulators or prosthetics that compensate for loss of function due to injury. Here we present a novel design of interface for peripheral nerves. Recording from axons is complicated by the small size of extracellular potentials and the concentration of current flow at nodes of Ranvier. Confining axons to microchannels of ~100 µm diameter produces amplified potentials that are independent of node position. After impl

Evaluation of implantable actuators by means of a middle ear simulation model.

The extension of indication of implantable hearing aids to cases of conductive hearing loss pushed the development of these devices. There is now a great variety of devices available with different actuator concepts and different attachment points to the middle ear or inner ear fluid. But there is little comparative data available about the devices to provide an insight into advantages and disadvantages of different types of actuators and attachment points at the ossicular chain. This paper investigates two principle (idealized) types of actuators in respect of attachments points at the ossicu

Development of redox-labeled electrochemical immunoassay for polycyclic aromatic hydrocarbons with controlled surface modification and catalytic voltammetric detection.

A redox-labeled direct competitive electrochemical immunoassay for polycyclic aromatic hydrocarbons (PAHs) was developed. A ruthenium tris(bipyridine)-pyrenebutyric acid conjugate was synthesized as the redox-labeled tracer. Its recognition by an anti-PAH monoclonal antibody was confirmed by surface plasmon resonance. In the immunoassay, the antibody was immobilized on (3-glycidoxypropyl)-trimethoxysilane (GPTMS)-modified indium tin oxide (ITO) electrodes. The assay was quantified by measuring the electro-catalytic current of the redox label in an oxalate-containing electrolyte which served as

Characterization of a new isolate of Ehrlichia platys (Order Rickettsiales) using electron microscopy and polymerase chain reaction.

A mixed-breed pup approximately 3 months old obtained in north central Oklahoma by the Laboratory Animal Resources Unit of Oklahoma State University presented with platelet inclusions. The dog developed severe thrombocytopenia (< 10,000 microliters-1) following the appearance of inclusions. Blood films were monitored daily and when about 75% of platelets had inclusions, samples were collected in EDTA and processed for electron microscopic (EM) studies and polymerase chain reaction (PCR). EM studies on glutaraldehyde-fixed buffy coat revealed rickettsia-like inclusions in numerous platelets. Se

CRISPR/Cas13a-Based Nucleic Acid-Functionalized Discrete Floating-Gate Field-Effect Transistor for Amplification-Free Rapid Nipah Virus Detection.

Nipah virus (NiV) is a highly lethal zoonotic pathogen, and existing rapid diagnostic tools are scarce and reliant on nucleic acid amplification. In this study, we designed and fabricated a thin-film field-effect transistor with an IGZO channel and, for the first time, combined a discrete AuNP floating gate with the CRISPR/Cas13a system to achieve amplification-free detection of NiV. A pair of partially complementary nucleic acid reporter probes was used to attach AuNPs onto the channel, forming a nucleic acid-functionalized discrete floating gate. Under positive gate voltage, the AuNPs electr

FAST-MaP: Chemical Mapping of RNA Structures Using Primer-less Sequencing.

RNA structure governs the function of non-coding RNAs and influences mRNA stability and translation, making testing of models of experimental RNA structure relevant to fields spanning structural biology, virology, and molecular therapeutics. Here we present FAST-MaP (Fast and Accessible Sequencing Technology for Mutational Profiling), a protocol that enables per-nucleotide RNA structure characterization using standard molecular biology equipment and requiring no sequencing infrastructure or bioinformatics expertise. RNA is chemically modified with orthogonal probes (2A3 and DMS), reverse-trans

Mitochondria-Targeted Photodynamic Agents Drive Robust Pyroptosis by Caspase-3/GSDME Activation and Cardiolipin Externalization.

Photodynamic therapy (PDT)-induced pyroptosis has attracted considerable interest as a strategy for cancer therapy, yet rational design of photosensitizers (PSs) that efficiently elicit pyroptosis remains underdeveloped. Herein, we report mitochondria-targeted supramolecular PSs that simultaneously activate the caspase-3/gasdermin E (GSDME) signal pathway and promote cardiolipin (CL) externalization to drive robust PDT-induced pyroptosis. A series of BODIPY-based supramolecular PSs, containing quaternary-ammonium moieties and N-containing aromatic heterocyclic substituents, were rationally des

Electronic profiling of neuronal extracellular vesicles enables early prediction of Parkinson's disease.

Misfolded alpha-Synuclein (α-Syn) accumulation is the defining molecular pathology of Parkinson's disease (PD), yet blood-based assays remain challenged by the high background of peripheral α-Syn originating mainly from red blood cells. Neuronal extracellular vesicles (EVs) circulating in blood offer a biologically enriched source of brain-derived α-Syn proteoforms, but their ultralow abundance has hindered reliable detection. Here, we introduce an organic electrochemical transistor-based multiparametric diagnostic assay that simultaneously detects total, aggregated, and serine-129-phosphoryla

Membrane remodeling by the collective action of caveolin-1.

Caveolin-1 proteins scaffold 50-100 nm large invaginations in the plasma membrane to mediate critical cellular processes. As revealed recently by cryo-electron microscopy, several caveolin-1 protomers can fold into a disk-like structure that embeds in the cytoplasmic leaflet. This 8S complex represents a basal component to drive membrane curvature via higher-order interactions. The biophysical mechanisms behind the membrane remodeling, however, are elusive. To address this shortcoming, we develop a bottom-up coarse-grained model to overcome the substantial computational limitations for this la
