KIT (CD117): a review on expression in normal and neoplastic tissues, and mutations and their clinicopathologic correlation.

CD117 (KIT) is a type III receptor tyrosine kinase operating in cell signal transduction in several cell types. Normally KIT is activated (phosphorylated) by binding of its ligand, the stem cell factor. This leads to a phosphorylation cascade ultimately activating various transcription factors in different cell types. Such activation regulates apoptosis, cell differentiation, proliferation, chemotaxis, and cell adhesion. KIT-dependent cell types include mast cells, some hematopoietic stem cells, germ cells, melanocytes, and Cajal cells of the gastrointestinal tract, and neoplasms of these cell

Interleukin-8 and human cancer biology.

The aggressive nature of metastatic human cancer has been shown to be related to numerous abnormalities in growth factors and their receptors. These perturbations confer a tremendous growth advantage to the malignant cells. Interleukin-8 (IL-8), originally discovered as a chemotactic factor for leukocytes, has recently been shown to contribute to human cancer progression through its potential functions as a mitogenic, angiogenic, and motogenic factor. While it is constitutively detected in human cancer tissues and established cell lines, IL-8 expression is regulated by various tumor microenvir

[Chromosome aberrations and genes in human and experimental leukemias].

Although determination of chromosomal abnormalities may be of limited usefulness for the diagnosis of leukemia, the recent advances in the molecular mechanism associated with chromosome aberration has been rapid. Chromosome translocation in Burkitt lymphoma and chronic myeloid leukemia was the most striking evidence for the oncogene activation. Other specific chromosome abnormalities for FAB-classified leukemias are also known. Translocated type of chromosome abnormalities between immunoglobulin or T-cell receptor genes and oncogenes may also affect the T and B-cell leukemogenesis. However, th

Spatial profiling of human pancreatic ductal adenocarcinoma reveals molecular alterations associated with venous invasion.

In pancreatic ductal adenocarcinoma (PDAC), venous invasion (VI) is a critical step in metastasis and is associated with poor survival. However, little is known about the molecular features of VI. To investigate, we performed spatial transcriptomic analysis of 95 human PDAC tissue samples from eight treatment-naïve patients. Our analysis revealed that, compared with PDAC in stroma, PDAC with VI demonstrated up-regulation of genes associated with epithelial differentiation, classical subtype, and benign exocrine function. Conversely, PDAC with VI demonstrated down-regulation of genes associated

Drug resistant glioblastoma stem cells exhibit enriched stemness signatures and share extracellular matrix overexpression.

BACKGROUND: Understanding the complex molecular mechanisms driving drug resistance in glioblastoma (GBM) is crucial to develop effective therapeutic strategies. While prior studies have identified resistance mechanisms tied to specific drugs or pathways, a multi-modal molecular analysis of resistance across a range of drug classes is lacking. METHODS: We identified highly drug-resistant (n = 5) and drug-sensitive (n = 4) cultures from a cohort of 32 patient-derived glioblastoma stem cell (GSC) cultures screened against a broad panel of ~ 500 anti-cancer drugs. To elucidate the key drivers of d

Cancer-prone oxyradical overload disease.

Oxyradical overload disease develops in conditions involving chronic inflammation and may be of inherited etiology, e.g. haemochromatosis and Wilson disease, be acquired, e.g. infection with hepatitis B or C virus or Helicobactor pylori, or be chemically induced, e.g. acid reflux in Barrett oesophagus. Susceptibility to cancer is frequently a pathological consequence of extensive oxyradical damage that leads to a cycle of cell death and regeneration and causes mutations in cancer-related genes. In this brief review, we focus on the possible interactive effects of nitric oxide and the p53 tumou

Spatial ecology of breast cancer reveals co-evolution of proliferative and dormant niches.

BACKGROUND: Cancer progression involves not only uncontrolled proliferation but also the strategic entry of tumour cells into reversible (quiescent) or irreversible (senescent) states of cell cycle arrest (G0). These states can give rise to rare persister-like cancer cells that survive hostile tumour microenvironment conditions, facilitating drug resistance, metastasis and disease relapse. Despite their importance, identifying and understanding the mechanisms regulating these cell populations remains challenging. METHODS: We leveraged single-cell and spatially profiled primary breast tumours

STE-DC2I Uncovers Driver Genes in Colorectal Cancer Subtypes Using Symbolic Trajectory-Embedded Dark Causal Inference.

Colorectal cancer (CRC) exhibits substantial molecular heterogeneity, necessitating the inference of subtype-specific driver genes and their interactions for drug-target discovery and precision oncology. Prior studies often fail to capture subtle, latent nonlinear regulatory mechanisms (dark causal relationships) driving tumor progression in specific subtypes. Here, we develop an explainable intelligence computational framework, Symbolic Trajectory-Embedded Dark Causal Interaction Inference (STE-DC2I), which combines symbolic trajectory embedding with historical prediction mechanisms to model

Unraveling the Therapeutic Landscape of miRNAs in Kaposi Sarcoma.

Kaposi sarcoma (KS) is a vascular malignancy caused by human herpesvirus 8 (HHV-8), also known as Kaposi sarcoma-associated herpesvirus (KSHV). Clinically, KS presents with characteristic lesions on the skin or mucosal surfaces and may extend to internal organs, leading to serious complications such as gastrointestinal bleeding and lymphedema. The prevalence of KSHV varies markedly by geography, with the highest rates reported in sub-Saharan Africa, the Mediterranean region, and high-risk populations such as individuals living with HIV/AIDS. KS pathogenesis is primarily driven by KSHV-induced

Stage-Specific Mucin Reprogramming Across the Gastric Cancer Cascade: Unveiling Molecular Mechanisms and Novel Therapeutic Vulnerabilities.

Mucins are a class of highly glycosylated macromolecules that constitute a cornerstone of the gastrointestinal mucosal barrier and play multiple essential roles in maintaining tissue homeostasis. During the progression from normal gastric mucosa through precancerous lesions to gastric cancer and metastasis, the expression profiles, glycosylation patterns, and spatial distribution of mucins undergo systematic and programmatic alterations, a process referred to as "mucin reprogramming." Recent studies have revealed that this reprogramming is by no means a passive bystander phenomenon accompanyin

NAT10-mediated ac4C-modified genes predict tumor prognosis and immune response in colon adenocarcinoma.

N-4 cytidine acetylation (ac4C), facilitated by N-acetyltransferase 10 (NAT10), is essential in regulating mRNA stability, rRNA biogenesis, cell proliferation, and epithelial-mesenchymal transition. However, the prognostic implications of NAT10-mediated ac4C modifications in colon adenocarcinoma (COAD) remains unclear. Our study demonstrated that NAT10 facilitates tumor growth and modulates an immune response in COAD. Moreover, we identified 2156 ac4C-related genes from the literature and obtained COAD patient information from TCGA. We constructed a prognostic risk model based on NAT10-mediate

MiRNA-211 Dysregulation contributes to the pathogenesis of digestive tract cancers: Mechanistic and clinical implications.

Gastrointestinal tract cancers are a significant cause of mortality, with cancers of the colon, stomach, liver, and pancreas being the most common types of cancer. Early detection is crucial for improving survival rates. Epigenetic modifications, such as microRNA (miRNA) gene silencing, play a crucial role in the development and progression of gastrointestinal cancer. Aberrant miRNA expression may contribute to cancer growth, proliferation, apoptosis, and drug resistance. This study focuses on miRNA-211, a dysregulated miRNA in gastrointestinal cancer that may act as a tumor suppressor or prom

Pseudouridine Synthase 7 in Cancer: Functions, Mechanisms, and Therapeutic Potential.

Pseudouridylation, the most abundant RNA modification, plays a critical role in modulating RNA structure, stability, and function. Among the family of pseudouridine synthases, Pseudouridine Synthase 7 (PUS7) has recently gained attention for its emerging roles in human health and disease. Originally characterized for its function in modifying tRNA and small non-coding RNAs, PUS7 is now recognized as a dynamic regulator of mRNA pseudouridylation, influencing gene expression at the post-transcriptional level. Aberrant expressions or activity of PUS7 have been linked to a variety of pathological

Nucleotide Substitution Biases in Related Cancer Driver Genes.

Nucleotide substitutions are common in cancer cells, and they occur in both protein coding regions and non-coding regions (5' and 3' UTRs and introns). Although substitutions in non-coding regions have the potential to alter gene expression, it is the alteration of coding regions that affects protein function and has the most drastic effect on cellular transformation. Mutations in certain genes (e.g., TP53, KRAS) are common to nearly all cancers, but most cancers are characterized by specific gene mutation signatures. In this report, we investigated nucleotide substitution signatures in coding

Joint analysis of sQTL and Hi-C reveals spatial proximity between sQTLs and target genes in cancer tissues.

Gene expression and regulation with or without alternative splicing are crucial for tissues and cells to function correctly. They have been studied from three almost independent perspectives at the genome level: 1- Recognition of splicing quantitative trait loci (sQTLs), 2- Expression quantitative trait loci (eQTLs) recognition, and 3- Recognition of longer-range physical chromatin interactions between genome segments which model 3D dynamics of cells and tissues. Even though the associations between eQTLs and longer-range chromatin interactions have been previously studied, a similar relations
