Copper binding to mouse liver S-adenosylhomocysteine hydrolase and the effects of copper on its levels.

The dissociation constant and stoichiometry of copper binding to mouse liver S-adenosylhomocysteine hydrolase (SAHH) was determined as part of characterizing the possible roles of SAHH in copper metabolism. Copper (64Cu(II)) binding was measured by an ultrafiltration method in the presence of EDTA as a competing ligand. The KD was 3.9 +/- 0.7 x 10(-16) M, and the stoichiometry was one g atom of copper per 48-kDa subunit. Western blots indicated that the liver contains approximately 12 times more SAHH than the kidney, which in turn contains approximately 5 times more SAHH than the brain. The hi

A single mutation of the fumarylacetoacetate hydrolase gene in French Canadians with hereditary tyrosinemia type I.

BACKGROUND: Hereditary tyrosinemia type I is an autosomal recessive inborn error of metabolism caused by a deficiency of the enzyme fumarylacetoacetate hydrolase. The disorder clusters in the Saguenay-Lac-St.-Jean area of Quebec. In this region, 1 of 1846 newborns is affected and 1 of every 22 persons is thought to be a carrier. Recently, we identified a splice mutation and two nonsense mutations in the fumarylacetoacetate hydrolase gene in two patients from Quebec with tyrosinemia type I. METHODS: We used allele-specific-oligonucleotide hybridization to examine the frequency of these three c

Loss of fumarylacetoacetate hydrolase is responsible for the neonatal hepatic dysfunction phenotype of lethal albino mice.

Mice homozygous for the c14CoS albino deletion die as neonates as a result of liver dysfunction. Previous mapping studies have associated this defect with a 310-kb fragment encoding the hepatocyte-specific developmental regulation locus (alf/hsdr-1). The gene encoding fumarylacetoacetate hydrolase (Fah), a metabolic enzyme that catalyzes the last step of tyrosine catabolism, also maps to the same deletion interval. To test whether the neonatal defects found in the albino deletion mutants are attributable to loss of Fah, and not to another gene mapping to the deletion, we have generated Fah mut

L-Ascorbic acid and lysosomal acid hydrolase activities of guinea pig liver and brain.

The effects of L-ascorbic acid deficiency on guinea pig hepatic and brain lysosomal hydrolases were examined. In general, hepatic beta-N-acetylhexosaminidase, beta-D-glucoronidase, alpha-D-galactosidase, alpha-D-mannosidase, and acid phosphatase were elevated in scorbutic animals. This appears to be independent of the starved state. Brain beta-D-glucoronidase and acid phosphatase followed a similar pattern to that observed with the liver enzymes, but brain beta-N-acetylhexosaminidase was not affected by L-ascorbic acid decreased the activity of hepatic beta-N-acetylhexosaminiadase was unaffect…

Multiplexed Gel-based ABPP Strategy for Target-agnostic Serine Hydrolase Inhibitor Screening.

Serine hydrolases (SHs) represent a functionally defined enzyme class with significant roles in human physiology, yet many of the approximately 240 human SH's lack selective chemical inhibitors required for functional validation. To address this, we developed a medium throughput screening platform based on multiplexed gel-based activity-based protein profiling (ABPP). Using mouse brain proteome as a source, we performed a target-agnostic screen of 1,664 covalent compounds, leading to the identification of three micromolar-potent fatty acid amide hydrolase (FAAH) inhibitors featuring urea warhe…

A novel positive selection system for plant transformation based on microbial biuret hydrolase and biuret.

Conventional selectable marker genes (SMGs) in plant transformation, such as those conferring antibiotic resistance, raise biosafety concerns due to their permanent genomic integration. Alternative systems, such as the phosphite (Phi)/phosphite dehydrogenase (PtxD) system, have been developed to address these concerns. This system works by enabling transgenic plants to convert non-metabolizable, toxic Phi into usable orthophosphate (Pi), thereby providing both a detoxification mechanism and a positive growth advantage for selection. Building on this principle of detoxification-based positive s

N-terminal phosphatase of soluble epoxide hydrolase forms anandamide from anandamide phosphate.

Soluble epoxide hydrolase exhibits two distinct activities: C-terminal epoxide hydrolase (C-EH) and N-terminal phosphatase (N-phos). Epoxy fatty acids are the primary substrates of C-EH and exhibit anti-inflammatory properties. N-phos hydrolyses lipid phosphate monoesters, such as isoprenoid pyrophosphates, lysophosphatidic acid (LPA), and sphingosine-1-phosphate (S1P). We found that inhibiting both sEH activities, rather than just C-EH, suppressed vascular endothelial cell (VEC) activation. A negatively charged polar lipid (NCPL) fraction from mouse liver extracts also suppressed VEC activati

Carbamoyl fluorides as serine hydrolase inhibitors: a case study on FphI from Staphylococcus aureus.

Selective covalent inhibitors have recently gained popularity as potent therapeutic drugs or molecular tools to investigate protein function. Serine hydrolases are a particular class of enzymes involved in diverse diseases. They possess a nucleophilic catalytic serine residue which can be targeted by covalent warheads. Carbamoyl fluoride is a relatively underexplored and underutilized organofluorine warhead. Here, we have designed and screened a focused library of carbamoyl fluoride fragments that successfully inhibited a recently discovered biofilm-associated serine hydrolase, FphI, from the …

Mechanistic insights into ochratoxin A biodetoxification by Pediococcus acidilactici NX8 involving a multi-hydrolase toolbox.

This study aimed to develop efficient and environmentally friendly detoxification strategies for ochratoxin A (OTA). Pediococcus acidilactici NX8, isolated from traditional fermented dairy products, completely degraded 1 μg/mL OTA within 24 h. By integrating degradation pathway analysis and multi-omics approaches, the molecular mechanism underlying OTA degradation was elucidated, and three novel OTA-hydrolyzing enzymes (XlyAB, AmiE, and Pbp4b) were identified. The recombinant variants completely degraded 2 μg/mL OTA within 5 min and remained stable at pH 4-7 and 4-50 °C. Molecular simulations

Prodrug activation in malaria parasites mediated by an imported erythrocyte esterase, acylpeptide hydrolase (APEH).

The continued emergence of antimalarial drug resistance highlights the need to develop new antimalarial therapies. Unfortunately, new drug development is often hampered by undesirable drug-like properties of lead compounds. Prodrug approaches temporarily mask undesirable compound features, improving bioavailability and target penetration. We have found that lipophilic diester prodrugs of phosphonic acid antibiotics, such as fosmidomycin (Fsm), exhibit significantly higher antimalarial potency than their parent compounds [R.L. Edwards et al., Sci. Rep. 7, 8400 (2017)]. However, the activating e

Global distribution of α/β hydrolase family macrolide esterases in Gram-positive bacteria.

Macrolide antibiotics are vital for controlling infections in humans, animals, and agriculture, yet their effectiveness is increasingly compromised by antimicrobial resistance. Macrolide esterases (MLEs) are key mediators of macrolide resistance but have only been detected in Gram-negative bacteria, with no evidence in Gram-positive species. Here, we mined over 500 000 Gram-positive genomes and identified 8707 candidate proteins. Six representative MLEs were functionally validated, conferring resistance to 16-membered macrolides and increasing minimum inhibitory concentrations (MICs) up to 16-

Characterization of a GDS(L)-like hydrolase from Pleurotus sapidus with an unusual SGNH motif.

The GDS(L)-like lipase from the Basidiomycota Pleurotus sapidus (PSA_Lip) was heterologously expressed using Trichoderma reesei with an activity of 350 U L-1. The isoelectric point of 5.0 was determined by isoelectric focusing. The novel PSA_Lip showed only 23.8-25.1%, 25.5%, 26.6% and 28.4% identity to the previously characterized GDSL-like enzymes phospholipase, plant lipase, acetylcholinesterase and acetylxylan esterase, from the carbohydrate esterase family 16, respectively. Therefore, the enzyme was purified from the culture supernatant and the catalytic properties and the substrate speci

High expression of oleoyl-ACP hydrolase underpins life-threatening respiratory viral diseases.

Respiratory infections cause significant morbidity and mortality, yet it is unclear why some individuals succumb to severe disease. In patients hospitalized with avian A(H7N9) influenza, we investigated early drivers underpinning fatal disease. Transcriptomics strongly linked oleoyl-acyl-carrier-protein (ACP) hydrolase (OLAH), an enzyme mediating fatty acid production, with fatal A(H7N9) early after hospital admission, persisting until death. Recovered patients had low OLAH expression throughout hospitalization. High OLAH levels were also detected in patients hospitalized with life-threatening

Catalytic mechanism, computational design, and crystal structure of a highly specific and efficient benzoylecgonine hydrolase.

Enzyme therapy for cocaine detoxification should break down both cocaine and its primary toxic metabolite, benzoylecgonine (BZE), which is also the main form of cocaine contaminant in the environment. An ideal BZE-metabolizing enzyme (BZEase) is expected to be highly efficient and selective in BZE hydrolysis. Here, BZEase4 was engineered from bacterial cocaine esterase (CocE) by our reactant state-based enzyme design theories (RED), which has a 34,977-fold improved substrate discrimination between BZE and the neurotransmitter acetylcholine (ACh), compared with wild-type CocE. Under the physiol

Structural basis for substrate recognition by a S-adenosylhomocysteine hydrolase Lpg2021 from Legionella pneumophila.

S-Adenosyl-l-homocysteine hydrolase (SAHH) is a crucial enzyme that governs S-adenosyl methionine (SAM)-dependent methylation reactions within cells and regulates the intracellular concentration of SAH. Legionella pneumophila, the causative pathogen of Legionnaires' disease, encodes Lpg2021, which is the first identified dimeric SAHH in bacteria and is a promising target for drug development. Here, we report the structure of Lpg2021 in its ligand-free state and in complexes with adenine (ADE), adenosine (ADO), and 3-Deazaneplanocin A (DZNep). Genetic Knockout of Fatty Acid Amide Hydrolase Ameliorates Cisplatin-Induced Nephropathy in Mice.

Cisplatin is a potent first-line therapy for many solid malignancies, such as breast, ovarian, lung, testicular, and head and neck cancer. However, acute kidney injury (AKI) is a major dose-limiting toxicity in cisplatin therapy, which often hampers the continuation of cisplatin treatment. The endocannabinoid system, consisting of anandamide (AEA) and 2-arachidonoylglycerol and cannabinoid receptors, participates in different kidney diseases. Inhibition of fatty acid amide hydrolase (FAAH), the primary enzyme for the degradation of AEA and AEA-related N-acylethanolamines, elicits anti-inflamma

Synergic action of an inserted carbohydrate-binding module in a glycoside hydrolase family 5 endoglucanase.

Most known cellulase-associated carbohydrate-binding modules (CBMs) are attached to the N- or C-terminus of the enzyme or are expressed separately and assembled into multi-enzyme complexes (for example to form cellulosomes), rather than being an insertion into the catalytic domain. Here, by solving the crystal structure, it is shown that MtGlu5 from Meiothermus taiwanensis WR-220, a GH5-family endo-β-1,4-glucanase (EC 3.2.1.4), has a bipartite architecture consisting of a Cel5A-like catalytic domain with a (β/α)8 TIM-barrel fold and an inserted CBM29-like noncatalytic domain with a β-jelly-rol

Development of a Highly Efficient Long-Acting Cocaine Hydrolase Entity to Accelerate Cocaine Metabolism.

It is particularly challenging to develop a truly effective pharmacotherapy for cocaine use disorder (CUD) treatment. Accelerating cocaine metabolism via hydrolysis at cocaine benzoyl ester using an efficient cocaine hydrolase (CocH) is known as a promising pharmacotherapeutic approach to CUD treatment. Preclinical and clinical studies on our first CocH (CocH1), in its human serum albumin-fused form known as TV-1380, have demonstrated the promise of a general concept of CocH-based pharmacotherapy for CUD treatment. However, the biological half-life of TV-1380 (t1/2 = 8 h in rats, associated wi
