Epstein-Barr virus-specific RNA. I. Analysis of viral RNA in cellular extracts and in the polyribosomal fraction of permissive and nonpermissive lymphoblastoid cell lines.

We analyzed the viral RNA in permissive and nonpermissive Epstein-Barr virus (EBV) Pinfected lymphoblastoid cell lines by observing the kinetics of hybridization of labeled EBV HR-1 DNA with unalabeled RNA extracted from whole cells or from the polyribosomal fraction. The data indicate the following. (i) RNA, homologous to only 3% of the EBV HR-1 DNA, is present in the polyribosomal fraction of the nonpermissive Namalwa and Kurgans cells, suggesting that the function of only a small fraction of the EBV genome is required for the expression of the EBV-related intranuclear antigen and to maintai

Effects of Protective Ventilation with Lung Expansion versus Permissive Atelectasis on Pulmonary Inflammation and Mechanical Power of Ventilation in an Experimental Model of Acute Lung Injury.

BACKGROUND: The energy transferred from the mechanical ventilator to the respiratory system over time, so-called mechanical power, may cause lung injury even with protective ventilation. This study hypothesized that protective ventilation aiming at moderate lung expansion results in less mechanical power and lung injury than aiming at permissive atelectasis or maximum lung expansion. METHODS: Twenty-four anesthetized pigs with acute lung injury induced by saline lung lavage were randomly assigned to ventilation according to either the Acute Respiratory Distress Syndrome Clinical Network's low

Protective parents and permissive children: what qualitative interviews with parents and children can tell us about the feasibility of juvenile idiopathic arthritis trials.

BACKGROUND: Patient recruitment can be very challenging in paediatric studies, especially in relatively uncommon conditions, such as juvenile idiopathic arthritis (JIA). However, involving children and young people (CYP) in the design of such trials could promise a more rapid trajectory towards making evidence-based treatments available. Studies involving CYP are advocated in the literature but we are not aware of any early stage feasibility studies that have qualitatively accessed the perspectives of parents and CYP with a long term condition to inform design and conduct of a trial. In the co

Immunization with neural derived peptides plus scar removal induces a permissive microenvironment, and improves locomotor recovery after chronic spinal cord injury.

BACKGROUND: Immunization with neural derived peptides (INDP) as well as scar removal-separately-have shown to induce morphological and functional improvement after spinal cord injury (SCI). In the present study, we compared the effect of INDP alone versus INDP with scar removal on motor recovery, regeneration-associated and cytokine gene expression, and axonal regeneration after chronic SCI. Scar removal was conducted through a single incision with a double-bladed scalpel along the stump, and scar renewal was halted by adding α,α'-dipyridyl. RESULTS: During the chronic injury stage, two exper

Is it permissible for a child with neurodevelopmental disabilities to be a living donor transplant candidate, but not a deceased donor candidate? Should donor source influence transplant center deliberations?

Limited data from surveys and highly publicized cases suggest that pediatric transplant providers and the public may use different standards when evaluating a potential transplant candidate with severe neurodevelopmental disabilities depending on whether a parent or other living donor has been identified. The purpose of this article is to explore the arguments in favor and against differing standards of candidacy for living and deceased donor transplants in pediatric solid organ transplantation using the example of children with severe neurodevelopmental disabilities. I conclude that donor sou

Prostaglandin E1 improved the function of transplanted fatty liver in a rat reduced-size-liver transplantation model under conditions of permissible cold preservation.

The aim of this study was to determine whether the minimum necessary volume of a moderate fatty liver graft was similar to the normal liver volume and to elucidate means for improving the function of the transplanted fatty liver if it were inferior in volume to a normal liver under conditions of permissible cold preservation. Nine-week-old male Wistar rats were used. Normal rat chow was fed to the normal liver group, and fat-enriched rat chow was fed to the fatty liver group for 4 weeks to induce a moderately fatty liver. Liver transplantation with various volumes of reduced-size grafts, inclu

Nitric oxide improves transpulmonary vascular mechanics but does not change intrinsic right ventricular contractility in an acute respiratory distress syndrome model with permissive hypercapnia.

OBJECTIVE: To test the hypothesis that in a swine model of acute respiratory distress syndrome (ARDS) with permissive hypercapnia, inhaled nitric oxide would improve transpulmonary vascular mechanics and right ventricular workload while not changing intrinsic right ventricular contractility. DESIGN: Prospective, randomized, controlled laboratory trial. SETTING: University research laboratory. SUBJECTS: Eleven swine (30 to 46 kg). INTERVENTIONS: The swine were anesthetized, intubated, and paralyzed. After median sternotomy, pressure transducers were placed in the right ventricle, pulmonary

Youth Involvement in Family Drinking and Parental Permission to Sip/Taste Alcohol: A Test of an Early Risk Pathway to Perceived Parental Approval of Underage Alcohol Use.

BACKGROUND: Several alcohol-specific parenting behaviors are associated with adolescent alcohol use. An important next step in the literature is integrating these behaviors into unified risk pathways, which have the potential to inform preventive interventions and public health messaging. Youth involvement in family drinking (i.e., asking children to fetch, pour, and open drinks) reflects parental-approved contact with alcohol, which may present the opportunity for sipping/tasting alcohol with parental permission. In turn, sipping/tasting alcohol with parental permission may increase adolescen

Nitric oxide generates a coordinated histone modification signature consistent with chromatin compaction, linked to methyl-modifying enzyme dysregulation and tumor-permissive gene silencing.

Our laboratory has established that the free radical signaling molecule nitric oxide (NO) is an endogenous regulator of epigenetic methylation through direct inhibition of the Fe(II)/2-oxoglutarate-dependent dioxygenase (2-ODD) family, including histone demethylases (KDMs)[1, 2], mRNA demethylases (FTO, ALKBH5) [3], and DNA demethylases (TETs, ALKBH2) [4]. We previously showed that NO alters 38 histone post-translational modifications (PTMs) and genome-wide transcription in breast cancer cells [3]. Here, we comprehensively quantify 76 histone modifications, including 30 combinatorial marks on

Does permissive hypoxaemia during extracorporeal membrane oxygenation cause long-term neurological impairment?: A study in patients with H1N1-induced severe respiratory failure.

BACKGROUND: The Extracorporeal Life Support Organisation accepts permissive hypoxaemia in adult patients during extracorporeal membrane oxygenation (ECMO). The neurological long-term outcome of this approach has not yet been studied. OBJECTIVES: We investigated the prevalence of brain lesions and cognitive dysfunction in survivors from the Influenza A/H1N1 2009 pandemic treated with permissive hypoxaemia during ECMO for severe acute respiratory distress syndrome (ARDS). Our hypothesis was that this method is reasonable if tissue hypoxia is avoided. DESIGN: Long-term follow-up study after ECM

Health impact of supplying safe drinking water containing fluoride below permissible level on flourosis patients in a fluoride-endemic rural area of West Bengal.

BACKGROUND: The problem of high fluoride concentration in groundwater resources has become one of the most important toxicological and geo-environmental issues in India. Excessive fluoride in drinking water causes dental and skeletal fluorosis, which is encountered in endemic proportions in several parts of the world. World Health Organization (WHO) guideline value and the permissible limit of fluoride as per Bureau of Indian Standard (BIS) is 1.5 mg/L. About 20 states of India, including 43 blocks of seven districts of West Bengal, were identified as endemic for fluorosis and about 66 million

Carrier detection and rapid newborn diagnostic test for the common Y393N maple syrup urine disease allele by PCR-RFLP: culturally permissible testing in the Mennonite community.

The turnaround time for diagnosis of maple syrup urine disease (MSUD) by classic serum amino acid analyses often requires 3-4 days. This is due to the need for branched-chain amino acids (BCAA) to accumulate in the serum of the newborn before testing. The accumulation of BCAAs in infants with MSUD during this time increases the risk of the infant becoming clinically symptomatic. We have developed a noninvasive DNA-based mismatch PCR-RFLP assay for the Y393N BCKDHA allele (E1alpha gene of the branched chain alpha-keto acid dehydrogenase complex), the primary cause of MSUD in Old Order Mennonite

M-1 and M-2 muscarinic receptor-mediated inhibition of dopamine-sensitive adenylate cyclase in rat neostriatum: a permissive role for D-2 dopamine receptors.

The interactions between dopamine and muscarinic receptor subtypes coupled to adenylate cyclase in superfused rat neostriatal slices were investigated using the efflux of cyclic AMP, in the presence of the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine, as a highly sensitive parameter of cyclic AMP production. Cyclic AMP efflux induced by simultaneous activation of (stimulatory) D-1 and (inhibitory) D-2 dopamine receptors by dopamine was reduced profoundly by the muscarinic receptor agonist oxotremorine and by inhibition of acetylcholinesterase with physostigmine, but not by the M-1 m

The Linkage Between Ecotoxins Within Maximum Permissible Concentrations, Oxidative Stress and Antibodies Against Cyclic Citrullinated Peptides in Patients and Persons at Preclinic Stages of Rheumatoid Arthritis.

Environmental factors are suspected of triggering rheumatoid arthritis (RA). One such factor is oxidative stress (OS), which is a step in ecotoxin detoxification and a damaging factor. The linkage of ecotoxin-triggered OS with clinical and laboratory RA indices in patients and individuals at the pre-RA stage was studied in patients with early (e) (n = 35) and advanced (a) stages of RA (n = 25) and individuals at pre-RA stages (FDR-First-Degree Relative(s), pre-RA, n = 72) in comparison with 52 healthy individuals without autoimmune and immunoinflammatory diseases in their family history (Contr

Role of permissive hypotension, hypertonic resuscitation and the global increased permeability syndrome in patients with severe hemorrhage: adjuncts to damage control resuscitation to prevent intra-abdominal hypertension.

Secondary intra-abdominal hypertension (IAH) and abdominal compartment syndrome (ACS) are closely related to fluid resuscitation. IAH causes major deterioration of the cardiac function by affecting preload, contractility and afterload. The aim of this review is to discuss the different interactions between IAH, ACS and resuscitation, and to explore a new hypothesis with regard to damage control resuscitation, permissive hypotension and global increased permeability syndrome. Review of the relevant literature via PubMed search. The recognition of the association between the development of ACS a

Naip5 affects host susceptibility to the intracellular pathogen Legionella pneumophila.

BACKGROUND: Legionella pneumophila is a gram-negative bacterial pathogen that is the cause of Legionnaires' Disease. Legionella produces disease because it can replicate inside a specialized compartment of host macrophages. Macrophages isolated from various inbred mice exhibit large differences in permissiveness for intracellular replication of Legionella. A locus affecting this host-resistance phenotype, Lgn1, has been mapped to chromosome 13, but the responsible gene has not been identified. RESULTS: Here, we report that Naip5 (also known as Birc1e) influences susceptibility to Legionella. …

A Case of Paraplegia Treated with Cerebrospinal Fluid Drainage and Permissive Hypertension after Graft Replacement of the Ascending Aorta and the Total Aortic Arch for Acute Aortic Dissection Stanford Type A.

Paraplegia after an operation for acute aortic dissection Stanford type A (AADA) is fairly uncommon, and there is no consensus about optimal treatment. We present a case in which cerebrospinal fluid drainage (CSFD) and permissive hypertension were used for treatment of paraplegia. When the patient showed complete bilateral paraplegia after operation for AADA, we immediately began CSFD and maintained mean arterial blood pressure at over 90 mmHg. His neurological deficit gradually recovered, and he was eventually able to walk without support. The combination of CSFD and permissive hypertension c
