Streptococcus agalactiae carriage among pregnant women living in Rio de Janeiro, Brazil, over a period of eight years.

Group B Streptococcus (GBS) carriage by pregnant women is the primary risk factor for early-onset GBS neonatal sepsis. Intrapartum antibiotic prophylaxis (IAP) can prevent this transmission route, and two main approaches are recommended to base the selection of pregnant women to be submitted to IAP: the risk-based and the culture-based strategies. In Brazil, compliance to such recommendations is poor, and not much is known about GBS carriage. In the present study, 3,647 pregnant women living in Rio de Janeiro State, Brazil, were screened for GBS anogenital colonization, over a period of 8 year

[Surveillance of Sensitivity to Penicillin in streptococcus Agalactiae recovered from pregnant women between 35-37 weeks of gestation].

Streptococcus agalactiae (GBS) cause severe infections in newborns under three months. Meningitis, pneumonia and sepsis are the main infectious diseases in these children. These infections are among the most serious that an individual can suffer in his first twelve hours of life. The child acquires the infection by vertical transmission of the colonized mother. To prevent neonatal disease, penicillin is recommended as the drug of choice for intrapartum prophylaxis (PIP) in pregnant women colonized. However, strains with decreased susceptibility to penicillin have been detected so it is importa…

Highly Sensitive and Efficient Screening of Streptococcus Agalactiae Through Improved Real-Time PCR Protocols and Optimized Analytical Parameters.

BACKGROUND: The detection of Streptococcus agalactiae colonization during pregnancy is important in order to prevent neonatal infections and reduce mortality rates. METHODS: A highly sensitive (up to 1,000 UFC/mL), fully automated, and cost effective cylB specific real-time PCR protocol was developed for the detection of S. agalactiae in anal and vaginal swabs of pregnant women. RESULTS: From the 62 test subjects, 24 were considered positive by PCR (38%) and 14 by culture (24%; p = 0.0015). CONCLUSIONS: The observed discrepancies and the reduced turnaround time justifies molecular screening…

Diversity of prophage DNA regions of Streptococcus agalactiae clonal lineages from adults and neonates with invasive infectious disease.

The phylogenetic position and prophage DNA content of the genomes of 142 S. agalactiae (group-B streptococcus, GBS) isolates responsible for bacteremia and meningitis in adults and neonates were studied and compared. The distribution of the invasive isolates between the various serotypes, sequence types (STs) and clonal complexes (CCs) differed significantly between adult and neonatal isolates. Use of the neighbor-net algorithm with the PHI test revealed evidence for recombination in the population studied (PHI, P = 2.01 × 10(-6)), and the recombination-mutation ratio (R/M) was 6:7. Neverthele

Oral immunization of Nile tilapia with Chlorella-bioencapsulated multi-epitope fusion antigen effectively protects against Streptococcus agalactiae infection.

Streptococcus agalactiae is an important bacterial pathogen of farmed tilapia; however, practical oral vaccines with stable protective efficacy remain limited because of poor antigen stability and low intestinal delivery efficiency. In this study, three immunodominant epitopes derived from the S. agalactiae surface immunogenic protein Sip, C5a peptidase ScpB, and leucine-rich repeat protein LrrG, as well as a multi-epitope fusion antigen (MEFA) constructed from these epitopes, were individually expressed in Chlorella sorokiniana, thereby establishing a microalgae-based oral vaccine system. Abs…

First identification of pathogenic Edwardsiella anguillarum, Nocardia asteroides, and Streptococcus agalactiae in tilapia (Oreochromis niloticus) farmed in Mexico.

Three different bacteria strains of the Nocardia, Edwardsiella, and Streptococcus genera were isolated from tilapia (Oreochromis niloticus) cultivated in Mexican farms with reports of granulomatous disease and mortality. The isolates were obtained from the tilapia spleen and characterized by biochemical tests, real-time PCR, and 16S rRNA sequencing. Comparisons with the GenBank Database indicated a 99.63 % and 99.87 % sequence identity to the 16S rRNA of the isolated samples with Edwardsiella anguillarum and Nocardia asteroides, respectively, and 100 % to Streptococcus agalactiae. The pathogen…

Selenium Exerts Antimicrobial Activity against the Perinatal Pathogen Streptococcus agalactiae and Perturbs Bacterial Interactions with Human Gestational Membranes.

Streptococcus agalactiae also known as Group B Streptococcus (GBS) is a Gram-positive, encapsulated, pathogenic bacterium. GBS causes severe perinatal infections that lead to chorioamnionitis, funisitis, premature rupture of membranes, preterm birth, maternal sepsis, neonatal sepsis, stillbirth, and maternal demise. Epidemiological data indicate that the nutrient selenium provides protection against infection and adverse disease outcomes and is a critical nutrient for development of a healthy pregnancy. We hypothesized that selenium could have antimicrobial activity against GBS. To test this, …

Immunostimulatory effects of Sarcodia suiae water extracts on Nile tilapia Oreochromis niloticus and its resistance against Streptococcus agalactiae.

In this study, water extracts of the red seaweed Sarcodia suiae were obtained using solid-liquid extraction (SLE) or pressurized liquid extr … of IL-1β, IL-8, TNFα, TP2, TP4, TLR2 and TLR5 in the spleen and immunoprotective efficiency against Streptococcus agalactiae infection. Taken together, these results show that S. suiae can differentially stimulate the immune response of tilapia in vitro and in vivo and could potentially be used as an immunomodulator in tilapia culture.

In silico prediction of conserved vaccine targets in Streptococcus agalactiae strains isolated from fish, cattle, and human samples.

Streptococcus agalactiae (Lancefield group B; group B streptococci) is a major pathogen that causes meningoencephalitis in fish, mastitis in cows, and neonatal sepsis and meningitis in humans. The available prophylactic measures for conserving human and animal health are not totally effective and have limitations. Effective vaccines against the different serotypes or genotypes of pathogenic strains from the various hosts would be useful. We used an in silico strategy to identify conserved vaccine candidates in 15 genomes of group B streptococci strains isolated from human, bovine, and fish sam…
