Malaria is caused by mosquito-borne Plasmodium spp. parasites that must infect and survive within mosquito salivary glands (SGs) prior to host transmission. Recent advances in transcriptomics and the complete genome sequencing of mosquito vectors have increased our knowledge of the SG genes and proteins involved in pathogen infection and transmission. Membrane solute carriers are key proteins involved in drug transport and are useful in the development of new interventions for transmission blocking. Herein, we applied transcriptomics analysis to compare SGs mRNA levels in Anopheles stephensi f…

Plasmodium vivax is the most geographically widespread malaria parasite. Unique features of transmission biology complicate P. vivax control. Interventions targeting transmission are required for malaria eradication. In the absence of an in vitro culture, transmission studies rely on live isolates from non-human primates or endemic regions. Here, we demonstrate P. vivax gametocytes from both India and Brazil are stable during cryopreservation. Importantly, cryopreserved gametocytes from Brazil were capable of infecting three anopheline mosquito species in feedings done in the United States. Th

Chitin synthase (CHS) represents an attractive target site for combating insect pests as insect growth and development are strictly dependent on precisely tuned chitin biosynthesis and this pathway is absent in humans and other vertebrates. Current knowledge on CHS in insects, especially their structures, functions, and regulations is still very limited. We report the identification and characterization of two chitin synthase genes, AgCHS1 and AgCHS2, in African malaria mosquito, Anopheles gambiae. AgCHS1 and AgCHS2 were predicted to encode proteins of 1,578 and 1,586 amino acid residues, resp…

The endosymbiotic bacterium Wolbachia is being investigated as a potential control agent in several important vector insect species. Recent studies have shown that Wolbachia can protect the insect host against a wide variety of pathogens, resulting in reduced transmission of parasites and viruses. It has been proposed that compromised vector competence of Wolbachia-infected insects is due to up-regulation of the host innate immune system or metabolic competition. Anopheles mosquitoes, which transmit human malaria parasites, have never been found to harbor Wolbachia in nature. While transient s…

Mosquitoes transmit serious human diseases, causing millions of deaths every year. Natural products of plant origin with insecticidal properties have been used in recent years for control of a variety of pest insects and vectors. The present study was based on assessments of the larvicidal activity to determine the efficacies of hexane, chloroform, ethyl acetate, acetone and methanol extracts of ten medicinal plants tested against fourth instar larvae of malaria vector, Anopheles stephensi Liston and lymphatic filariasis vector, Culex quinquefasciatus Say (Diptera: Culicidae). The larvicidal a…

BACKGROUND: Control of the Anopheline mosquito vectors of malaria by use of insecticides has been shown to impact on both morbidity and mortality due to this disease. Evidence of insecticide resistance in different settings necessitates surveillance studies to allow prompt detection of resistance should it arise and thus enable its management. Possible resistance by Anopheles arabiensis mosquitoes from Mwea rice irrigation scheme in Central Kenya to insecticides in the four classes of insecticides approved by WHO for indoor residual spraying was investigated.

Mosquito natriuretic peptide (MNP), an uncharacterised peptide from the yellow fever mosquito, Aedes aegypti, acts via cyclic AMP to stimulate secretion of Na+-rich urine by opening a Na+ conductance in the basolateral membrane of Malpighian tubule principal cells. Corticotropin releasing factor (CRF)-related peptides and calcitonin (CT)-like diuretic peptides use cyclic AMP as a second messenger and were therefore considered likely candidates for MNP. BLAST searches of the genome of the malaria mosquito Anopheles gambiae, gave sequences for the CRF-related peptide Anoga-DH44 and the CT-like p…

The karyotype of the African malaria mosquito Anopheles gambiae contains two pairs of autosomes and a pair of sex chromosomes. The Y chromosome, constituting approximately 10% of the genome, remains virtually unexplored, despite the recent completion of the A. gambiae genome project. Here we report the identification and characterization of Y chromosome sequences of total length approaching 150 kb. We developed 11 Y-specific PCR markers that consistently yielded male-specific products in specimens from both laboratory colony and natural populations. The markers are characterized by low sequenc…

The Anopheles pseudopunctipennis nitric oxide synthase gene (ApNOS) was identified and its partial sequence showed high homology with NOS from A. stephensi, A. gambiae (putative sequence), and Drosophila melanogaster. ApNOS was mainly expressed in male and female adult mosquitoes and was induced by a blood meal. Nitric oxide (NO) was produced by in vitro-cultured mosquito midguts inoculated by enema with Plasmodium berghei ookinetes, Saccharomyces cerevisiae, Gram-positive bacteria (Micrococcus luteus), but not with Gram-negative bacteria (Klebsiella pneumoniae, Escherichia coli or Serratia ma…

Anopheline mosquito larvae and adults were sampled at Ban Pu Teuy, Tri-Yok District, Kanchanaburi Province, western Thailand, from January 2000 to December 2001. Over the period of 2 yr, Anopheles minimus sensu lato was the most commonly collected species, followed by Anopheles swadiwongporni and Anopheles dirus sensu lato; all three species are important vectors of malaria in Thailand. Attempted blood feeding by An. minimus occurred throughout the night, with two distinct feeding peaks: strong activity immediately after sunset (1800-2100 hours), followed by a second, less pronounced, rise bef…

Intracellular melanization, a defense or an immune response in the thoracic muscle cells, was investigated in a refractory strain of Anopheles quadrimaculatus infected with larvae of Brugia malayi. In mosquitoes fed on B. malayi-infected jirds, intracellular melanization against first-stage larvae (L1) was better expressed when fewer than 40 microfilariae reached the thoracic muscle cells than when more than 40 microfilariae reached the thoracic muscle cells. This result suggests that when large numbers of microfilariae invade the thoracic muscle cells, the immune response of the mosquito may …

Cellular G3BP proteins are essential for alphavirus infection in both vertebrate and mosquito hosts, but the underlying mechanism of their proviral activity is poorly understood in any host. Whether the mosquito G3BP ortholog, Rasputin (Rin), interacts with host immunity to influence alphavirus infection has not been investigated, and anopheline mosquito interactions with arboviruses have been little studied. Here, we find that Rin silencing in Anopheles mosquitoes results in decreased ONNV infection levels, indicating a proviral activity for Anopheles Rin. We find that Rin function is require…

The modification of mosquito populations at scale through CRISPR-Cas9-mediated homing gene drives is a promising route for malaria vector control. Integral gene drives (IGDs) are designed to utilize the regulatory sequences of endogenous genes to reduce the size of the modification required for nuclease and effector expression. In this study, we describe the creation and characterization of the nanosd integral gene drive, which targets and is inserted into the nanos gene of the malaria vector Anopheles gambiae, and show that it achieves high rates of gene drive (98.4% in females, 99.5% in male…

Anopheles stephensi is an important malaria vector in India which has been repeatedly incriminated for transmitting urban malaria in different parts of the country. The fight for its control in the metropolitan cities of India dates back to as early as 1911. Using an integrated approach of bio-environmental control along with community education and strict enforcement of legislatures, India has shown exemplary success in the control of malaria in different urban cities. From this perspective, we discuss the different success stories and the way forward for An. stephensi control in a global man…

A recent study conducted in West Khasi Hills (WKH) and West Jaintia Hills (WJH) districts of Meghalaya has documented a low occurrence of the primary malaria vectors, Anopheles baimaii and An. minimus, alongside a relatively higher abundance of other anophelines, particularly members of the Maculatus Group. Plasmodium infection in the An. maculatus sensu lato (s.l.) was last reported in 1941, with no subsequent implication of An. maculatus s.l. in malaria transmission from Meghalaya or Northeast (NE) India for over eight decades. Given this apparent gap between species abundance and its recogn…

Anopheles stephensi is an invasive malaria vector that has been rapidly spreading within Africa since it was first identified in Djibouti in 2012. As of 2025, it has been identified in eight other countries: Ethiopia, Sudan, Somaliland, Eritrea, Nigeria, Kenya, Ghana and Niger. We collected mosquitoes from an urban slaughterhouse in Kisumu City in western Kenya in mid-2022 for bloodmeal analyses. 19.6% (11/56) of the amplicon sequence variants (ASVs) assigned using the basic local alignment search tool (BLAST) returned matches for mosquito species, one of which was An. stephensi, from one pool…

BACKGROUND: The progress made against malaria has resulted in a nationwide reduction of the disease burden in Senegal. The observed overall low transmission levels are, however, marked by an important spatial heterogeneity with hotspots subsisting in several parts of the country. This requires the determination of the local and regional factors of the observed disparities for tailored interventions to accelerate malaria elimination everywhere. This study aimed to demonstrate the role of larval breeding sites on malaria epidemiological trends in Djilakh, which is one of the malaria hotspots of

Global changes are associated with the emergence of several invasive species, although genetic determinants of their adaptive success remain poorly understood. To address this problem, we investigated the role genome structural variants (SVs) play in adaptations of Anopheles stephensi, a primary vector of urban malaria in South Asia and an invasive malaria vector in South Asian islands and Africa. Using whole genome sequencing data, we identified 2,988 duplications and 16,038 deletions of SVs in 115 mosquitoes from invasive island populations and four locations from mainland India, the species…
