Changes in electroporation thresholds of lipid membranes by surfactants and peptides.

This article reviews recent work from our laboratory that explores how chemical additives may alter the threshold of electroporation of synthetic lipid bilayers. The addition of the nonionic block copolymeric surfactant, poloxamer 188 (P188), at a concentration of 1 mM increased the electroporation thresholds of planar lipid bilayer membranes made of azolectin. For a 10-microsecond rectangular pulse, P188-treated membranes were found to have a statistically higher threshold voltage, longer latency time to rupture, and lower postpulse conductance. Addition of the nonionic surfactant, octaethyle…

Oxidation Promotes Distinct Huntingtin Aggregates in the Presence and Absence of Membranes.

Expansion of a polyglutamine (polyQ) domain within the first exon of the huntingtin (htt) protein is the underlying cause of Huntington's di … at the expense of fibril elongation. The interaction of unoxidized and oxidized htt with supported lipid bilayers was directly observed using in situ atomic force microscopy. Without oxidation, granular htt aggregates developed on the bilayer surface. However, in the presence of H2O2, distinct plateau-like regions initially developed on the bilayer surface that …

Cholesterol Modifies Huntingtin Binding to, Disruption of, and Aggregation on Lipid Membranes.

Huntington's disease (HD) is an inherited neurodegenerative disease caused by abnormally long CAG-repeats in the huntingtin gene that encode an expanded polyglutamine (polyQ) domain near the N-terminus of the huntingtin (htt) protein. Expanded polyQ domains are directly correlated to disease-related htt aggregation. Htt is found highly associated with a variety of cellular and subcellular membranes that are predominantly comprised of lipids. Since cholesterol homeostasis is altered in HD, we investigated how varying cholesterol content modifies the interactions between htt and lipid membranes.

Rational design of new product candidates: the next generation of highly deformable bilayer vesicles for noninvasive, targeted therapy.

Amphipat bilayer vesicles are a subgroup of "fat-and-water" mixtures useful as drug carriers. Scrutinising amphipat aggregation in terms of the popular molecular descriptors (esp. the Israelachvili's form-factor or HLB number) is "too static" to foretell reliably and quantitatively bilayer vesicle formation. A better predictor introduced in this work is the effective area per lipid chain (cross-section of a "tail", A(c)), which also correlates, quasi-exponentially, with the ease of bilayer vesicle formation and bilayer deformability. The latter is highest near an uppermost, bilayer-compatible

Physicochemical Characterization of Anionic Lipid Membranes under Low and Physiological Ionic Strength: Effects of Moxifloxacin Assessed by Calorimetry, Spin-Label, Steady-State, and Time-Resolved Fluorescence.

In the present study, we comparatively investigate the interaction between the antibiotic moxifloxacin (MFX) and extruded (100 nm) lipid vesicles composed of the anionic lipid 1,2-dipalmitoyl-sn-glycero-3-phospho-(1'-rac-glycerol) (DPPG), under both low ([NaCl] = 3 mmol L-1) and physiological ionic strength conditions ([NaCl] = 150 mmol L-1). By employing differential scanning calorimetry (DSC) and electron spin resonance (ESR) spectroscopy with spin-labeled lipids positioned at two distinct depths within the bilayer, we evaluated structural alterations induced by increasing concentrations of

Gating modifier toxins isolated from spider venom: Modulation of voltage-gated sodium channels and the role of lipid membranes.

Gating modifier toxins (GMTs) are venom-derived peptides isolated from spiders and other venomous creatures and modulate activity of disease … d against a panel of NaV channel subtypes. We show that GMTs adsorb to the outer leaflet of anionic lipid bilayers through electrostatic interactions. We did not observe a direct correlation between GMT amphipathicity and affinity for lipid bilayers. Furthermore, GMT-lipid bilayer interactions did not correlate with potency or selectivity for NaVs. We therefore …

Sulpiride, Amisulpride, Thioridazine, and Olanzapine: Interaction with Model Membranes. Thermodynamic and Structural Aspects.

Neuroleptic drugs are widely applied in effective treatment of schizophrenia and related disorders. The lipophilic character of neuroleptics means that they tend to accumulate in the lipid membranes, impacting their functioning and processing. In this paper, the effect of four drugs, namely, thioridazine, olanzapine, sulpiride, and amisulpride, on neutral and negatively charged lipid bilayers was examined. The interaction of neuroleptics with lipids and the subsequent changes in the membrane physical properties was assessed using several complementary biophysical approaches (isothermal titrati…

Molecular communication system using Zn2+ selective supramolecular nanochannel to induce photoregulated catalysis.

We developed synthetic ionophores that self-assemble into supramolecular nanochannels for the transport of Zn2+ across lipid bilayers. The translocated Zn2+ forms a photoactive PPIX-Zn2+ complex that generates 1O2 to oxidize tryptophan, establishing a Zn2+-mediated cascade reaction network within confined spaces, which is crucial for developing photoregulated molecular communication systems.

Effects of antimicrobial peptides on membrane dynamics: A comparison of fluorescence and NMR experiments.

Antimicrobial peptides (AMPs) represent a promising class of compounds to fight resistant infections. They are commonly thought to kill bacteria by perturbing the permeability of their cell membranes. However, bacterial killing requires a high coverage of the cell surface by bound peptides, at least in the case of cationic and amphipathic AMPs. Therefore, it is conceivable that peptide accumulation on the bacterial membranes might interfere with vital cellular functions also by perturbing bilayer dynamics, a hypothesis that has been termed "sand in the gearbox". Here we performed a systematic

Cholesterol autoxidation in phospholipid membrane bilayers.

Lipid peroxidation in unilamellar liposomes of known cholesterol-phospholipid composition was monitored under conditions of autoxidation or as induced by a superoxide radical generating system, gamma-irradiation or cumene hydroperoxide. Formation of cholesterol oxidation products was indexed to the level of lipid peroxidation. The major cholesterol oxidation products identified were 7-keto-cholesterol, isomeric cholesterol 5,6-epoxides, isomeric 7-hydroperoxides and isomeric 3,7-cholestane diols. Other commonly encountered products included 3,5-cholestadiene-7-one and cholestane-3 beta, 5 alph

Effect of late endosomal DOBMP lipid and traditional model lipids of electrophysiology on the anthrax toxin channel activity.

Anthrax toxin action requires triggering of natural endocytic transport mechanisms whereby the binding component of the toxin forms channels (PA63) within endosomal limiting and intraluminal vesicle membranes to deliver the toxin's enzymatic components into the cytosol. Membrane lipid composition varies at different stages of anthrax toxin internalization, with intraluminal vesicle membranes containing ~70% of anionic bis(monoacylglycero)phosphate lipid. Using model bilayer measurements, we show that membrane lipids can have a strong effect on the anthrax toxin channel properties, including th

The mitochondrial ryanodine receptor in rat heart: a pharmaco-kinetic profile.

A protein discovered within inner mitochondrial membranes (IMM), designated as the mitochondrial ryanodine receptor (mRyR), has been recognized recently as a modulator of Ca(2+) fluxes in mitochondria. The present study provides fundamental pharmacological and electrophysiological properties of this mRyR. Rat cardiac IMM fused to lipid bilayers revealed the presence of a mitochondrial channel with gating characteristics similar to those of classical sarcoplasmic reticulum RyR (SR-RyR), but a variety of other mitochondrial channels obstructed clean recordings. Mitochondrial vesicles were thus s…

Molecular actions of pentobarbital isomers on sodium channels from human brain cortex.

New planar lipid bilayer technology enabled the pharmacologic study of single sodium channels from human brain, overcoming the limitations of tissue availability and the rapid loss of protein function in conventional experimental preparations. Synaptosomal vesicles prepared from human brain cortical tissue were fused with planar lipid bilayers. In the presence of batrachotoxin, sodium channels were incorporated into lipid bilayers and their single-channel properties studied. Pentobarbital was found to depress two major functions of the sodium channel, leading to a voltage-independent reduction…

Rational design of microfluidic liposome formulations: Integrating flow control with multi-analytical characterization.

Liposomes are vesicular structures consisting of one or more lipid bilayers that enclose an aqueous inner core, allowing the encapsulation of both hydrophilic compounds in the inner compartment and lipophilic and amphiphilic molecules within the lipid bilayer. In recent years, microfluidic-based manufacturing techniques have emerged as a highly attractive approach for liposome production, as they allow precise control over fluid dynamics at the micrometer scale, thereby enabling reproducible and scalable fabrication of nanocarriers with narrow size distributions. Since even minor variations in…
