An ion-exchange nanomembrane sensor for detection of nucleic acids using a surface charge inversion phenomenon.

We present a novel low-cost biosensor for rapid, sensitive and selective detection of nucleic acids based on an ionic diode feature of an anion exchange nanoporous membrane under DC bias. The ionic diode feature is associated with external surface charge inversion on the positively charged anion exchange nanomembrane upon hybridization of negatively charged nucleic acid molecules to single-stranded oligoprobes functionalized on the membrane surface resulting in the formation of a cation selective monolayer. The resulting bipolar membrane causes a transition from electroconvection-controlled to…

CdSe/ZnS core shell quantum dot-based FRET binary oligonucleotide probes for detection of nucleic acids.

We report the design, synthesis, and characterization of a binary oligonucleotideprobe for selective DNA or RNA detection. The probe is based on fluorescence resonance energy transfer (FRET) from quantum dot (CdSe/ZnS core shell) DNA conjugates to organic dye (cyanine-5) DNA conjugates. Selective hybridization of the donor/acceptor DNA conjugates to target DNA enhances FRET and a change in fluorescence signature was observed.

Interference with spectrophotometric analysis of nucleic acids and proteins by leaching of chemicals from plastic tubes.

Absorbance spectroscopy is routinely used to monitor the concentrations of nucleic acids and proteins within solutions and to assess changes in their structure caused by interaction with chemicals or other biomolecules. Biological samples used for such analyses are manipulated and stored in small microcentrifuge tubes (microtubes) composed of polypropylene plus several plastic additives. Here we demonstrate that normal handling of laboratory microtubes causes leaching of light-absorbing chemicals into biological samples that interfere with spectrophotometric measurements. The leached chromopho…

Targeted correction of a thalassemia-associated beta-globin mutation induced by pseudo-complementary peptide nucleic acids.

Beta-thalassemia is a genetic disorder caused by mutations in the beta-globin gene. Triplex-forming oligonucleotides and triplex-forming peptide nucleic acids (PNAs) have been shown to stimulate recombination in mammalian cells via site-specific binding and creation of altered helical structures that provoke DNA repair. However, the use of these molecules for gene targeting requires homopurine tracts to facilitate triple helix formation. Alternatively, to achieve binding to mixed-sequence target sites for the induced gene correction, we have used pseudo-complementary PNAs (pcPNAs). Due to ster…

Comparison of seven bio- and chemiluminescent reagents for in situ detection of antigens and nucleic acids.

Bio- and chemiluminescence have proved sensitive enough to compete with chromogenic and radioisotopic tracers for in situ detection. However, they must also provide a discriminant morphological analysis of the specific signal. We have tested seven bio- or chemiluminescent reagents for tissue antigen and nucleic acid detection by immunocytochemistry (ICC) or in situ hybridization (ISH). They were based on luminescent detection of peroxidase, alkaline phosphatase, beta-galactosidase or xanthine oxidase. We also explored whether high molecular weight polymers could increase the spatial definition

A bacterial fatty acid enzyme hijacks nucleic acid-sensing Toll-like receptor signaling.

Innate immune recognition shapes infection outcomes by linking microbial detection to host defense. Although pattern recognition receptors are classified by the ligands they detect (lipids, peptidoglycans, or nucleic acids) cross-talk between these pathways is increasingly recognized. Staphylococcus aureus, a major cause of skin and soft tissue infections, can persist intracellularly, evading immunity and antibiotics. Here, we describe a lipid-based immune evasion strategy in which the S. aureus enzyme oleate hydratase (OhyA) converts host fatty acids into hydroxylated lipids that antagonize T…

Cationic poly(lactic-co-glycolic acid) iron oxide microspheres for nucleic acid detection.

Herein, we envisage the possibility of preparing stable cationic poly(lactic-co-glycolic acid) (PLGA) microspheres encapsulating the iron oxide nanoparticles (IONPs; 8-12 nm). The IONPs are incorporated into PLGA in organic phase followed by microsphere formation and chitosan coating in aqueous medium via nano-emulsion technique. The average size of the microspheres, as determined by dynamic light scattering are about 310 nm, while the zeta potential for the composite remains near 35 mV at pH 4.0. These microspheres are electrophoretically deposited onto indium tin oxide (ITO)-coated glass sub

Programmable spherical nucleic acids integrated with MOF-confined copper nanoclusters facilitate electrochemiluminescence detection of prostate-specific biomarkers.

Alpha-methylacyl-CoA racemase (AMACR) is a promising prostate cancer biomarker due to its high specificity in distinguishing prostate cancer from benign prostatic diseases. However, its low abundance in biological environments presents a significant detection challenge. To address this, we developed an innovative all-in-one s̲pherical n̲ucleic a̲cid (SNA) platform for highly sensitive and selective electrochemiluminescence (ECL) detection of AMACR. The SNAs incorporate two types of ice-cream probes (IC probes), each consisting of interlocked hairpins and circular templates. Specifically, the a

Highly Active and Stable Au@Pt Nanozymes-Based Colorimetric Nucleic Acids Detection for Varicella-Zoster Virus Diagnosis.

PURPOSE: Currently, DNA is primarily detected using real-time quantitative polymerase chain reaction (qPCR) kits. However, their widespread clinical application is hindered by their high cost and the need for sophisticated instrumentation. Consequently, there is an urgent need to develop simpler DNA screening methods that are both cost-effective and compatible with resource-limited settings, thereby eliminating the need for complex equipment. METHODS: Taking advantage of the DNA-controlled property as well as the ultrahigh peroxidase-like catalytic activity of the Au@Pt nanozymes, we develope

Coding Therapeutic Nucleic Acids from Recombinant Proteins to Next-Generation Vaccines: Current Uses, Limitations, and Future Horizons.

This study aims to highlight the potential use of cTNAs in therapeutic applications. The COVID-19 pandemic has led to significant use of coding therapeutic nucleic acids (cTNAs) in terms of DNA and mRNA in the development of vaccines. The use of cTNAs resulted in a paradigm shift in the therapeutic field. However, the injection of DNA or mRNA into the human body transforms cells into biological factories to produce the necessary proteins. Despite the success of cTNAs in the production of corona vaccines, they have several limitations such as instability, inability to cross biomembranes, immuno…

Isothermal amplification based on specific signal extraction and output for fluorescence and colorimetric detection of nucleic acids.

Due to the complexity of compositions and low abundance of target in clinical sample, nucleic acids detection often suffers from false positives caused by nonspecific amplification. In in vitro diagnosis (IVD), PCR usually employ TaqMan probe to report specific signals and block false positive signals. However, nucleic acid isothermal amplifications, such as loop-mediated isothermal amplification (LAMP), lack of mature specific signal output mechanism, which prevents them from being used for IVD and point-of-care testing (POCT). In this work, we constructed a specific signal extract-to-output …

Drying and temperature induced conformational changes of nucleic acids and stallion sperm chromatin in trehalose preservation formulations.

Even though dried sperm is not viable, it can be used for fertilization as long as its chromatin remains intact. In this study, we investigated drying- and temperature-induced conformational changes of nucleic acids and stallion sperm chromatin. Sperm was diluted in preservation formulations with and without sugar/albumin and subjected to convective drying at elevated temperatures on glass substrates. Accumulation of reactive oxygen species was studied during storage at different temperatures, and the sperm chromatin structure assay was used to assess DNA damage. Fourier transform infrared spe…

Tetrahedral Framework Nucleic Acids Reestablish Immune Tolerance and Restore Saliva Secretion in a Sjögren's Syndrome Mouse Model.

As one of the most frequent autoimmune diseases, Sjogren's syndrome (SS) is characterized by overactive lymphocytic infiltration in the exocrine glands, with ensuing dry mouth and dry eyes. Unfortunately, so far, there are no appropriate therapies without causing overall immunosuppression. Tetrahedral framework nucleic acids (tFNAs) were regarded as promising nanoscale materials whose immunomodulatory capabilities have already been verified. Herein, we reveal, for the first time, that tFNAs were utilized to treat SS in female nonobese diabetic (NOD) mice, the animal model used for SS. We prove…

Ultra-sensitive and rapid detection of nucleic acids and microorganisms in body fluids using single-molecule tethering.

Detection of microbial nucleic acids in body fluids has become the preferred method for rapid diagnosis of many infectious diseases. However, culture-based diagnostics that are time-consuming remain the gold standard approach in certain cases, such as sepsis. New culture-free methods are urgently needed. Here, we describe Single MOLecule Tethering or SMOLT, an amplification-free and purification-free molecular assay that can detect microorganisms in body fluids with high sensitivity without the need of culturing. The signal of SMOLT is generated by the displacement of micron-size beads tethere…

In vivo magnetofection: a novel approach for targeted topical delivery of nucleic acids for rectoanal motility disorders.

In these studies, we developed a novel approach of in vivo magnetofection for localized delivery of nucleic acids such as micro-RNA-139-5p (miR-139-5p; which is known to target Rho kinase2) to the circular smooth muscle layer of the internal anal sphincter (IAS). The IAS tone is known to play a major role in the rectoanal continence via activation of RhoA-associated kinase (RhoA/ROCK2). These studies established an optimized protocol for efficient gene delivery using an assembly of equal volumes of in vivo PolyMag and miR139-5p or anti-miR-139-5p (100 nM each) injected in the circular smooth m…

Site-directed spin labeling measurements of nanometer distances in nucleic acids using a sequence-independent nitroxide probe.

In site-directed spin labeling (SDSL), local structural and dynamic information is obtained via electron paramagnetic resonance (EPR) spectroscopy of a stable nitroxide radical attached site-specifically to a macromolecule. Analysis of electron spin dipolar interactions between pairs of nitroxides yields the inter-nitroxide distance, which provides quantitative structural information. The development of pulse EPR methods has enabled such distance measurements up to 70 A in bio-molecules, thus opening up the possibility of SDSL global structural mapping. This study evaluates SDSL distance measu

Fetal cells and cell free fetal nucleic acids in maternal blood: new tools to study abnormal placentation?

The analysis of fetal cells, and more recently cell free fetal nucleic acids, in maternal blood has to date largely been concerned with the development of risk free methods for prenatal diagnosis. Although elevated feto-maternal cell trafficking has long been associated with preeclampsia, it has only recently been shown that this perturbation is an early event in these pregnancies, occurring well in advance of the onset of symptoms. In a separate development, analogous observations have been made concerning the levels of circulatory fetal nucleic acids in maternal plasma. Subsequent studies ha…
