Gastrointestinal organs and organoids-on-a-chip: advances and translation into the clinics.

Microfluidic organs and organoids-on-a-chip models of human gastrointestinal systems have been established to recreate adequate microenvironments to study physiology and pathophysiology. In the effort to find more emulating systems and less costly models for drugs screening or fundamental studies, gastrointestinal system organoids-on-a-chip have arisen as promising pre-clinicalin vitromodel. This progress has been built on the latest developments of several technologies such as bioprinting, microfluidics, and organoid research. In this review, we will focus on healthy and disease models of: hu…

Neuroepithelial organoid patterning is mediated by a neighborhood watch mechanism.

During epithelial tissue patterning, morphogens operate across multiple length scales to instruct cell identities. However, how cell fate changes are coordinated over these scales to establish spatial organization remains poorly understood. Here, we use human neural tube organoids as models of epithelial patterning and develop an in silico approach to define conditions permissive to patterning. By systematically varying morphogen position, diffusivity, and fate-inducing concentration levels, we show that cells follow a "neighborhood watch" (NW) mechanism that is deterministically dictated by i…

Establishment of Adenomyosis Organoids as a Preclinical Model to Study Infertility.

Adenomyosis is related to infertility and miscarriages, but so far there are no robust in vitro models that reproduce its pathological features to study the molecular mechanisms involved in this disease. Endometrial organoids are in vitro 3D models that recapitulate the native microenvironment and reproduce tissue characteristics that would allow the study of adenomyosis pathogenesis and related infertility disorders. In our study, human endometrial biopsies from adenomyosis (n = 6) and healthy women (n = 6) were recruited. Organoids were established and hormonally differentiated to recapitula…

An efficient and user-friendly method for cytohistological analysis of organoids.

Organoid culture is a recently developed in vitro three-dimensional (3D) cell culture technology. It has wide applications in tissue enginee … er without damaging or loss of samples. This method could be used to process even a small number of organoids with high efficiency. In addition, with minor modifications, the method is readily applied for OCT embedded organoid blocks. The slides generated were ready for H&E staining, immunohistochemistry staining and immunofluorescent staining. The method d …

Cerebral organoids as tools to identify the developmental roots of autism.

Some autism spectrum disorders (ASD) likely arise as a result of abnormalities during early embryonic development of the brain. Studying human embryonic brain development directly is challenging, mainly due to ethical and practical constraints. However, the recent development of cerebral organoids provides a powerful tool for studying both normal human embryonic brain development and, potentially, the origins of neurodevelopmental disorders including ASD. Substantial evidence now indicates that cerebral organoids can mimic normal embryonic brain development and neural cells found in organoids …

An Organoid-Based Preclinical Model of Human Gastric Cancer.

BACKGROUND & AIMS: Our goal was to develop an initial study for the proof of concept whereby gastric cancer organoids are used as an approach to predict the tumor response in individual patients. METHODS: Organoids were derived from resected gastric cancer tumors (huTGOs) or normal stomach tissue collected from sleeve gastrectomies (huFGOs). Organoid cultures were treated with standard-of-care chemotherapeutic drugs corresponding to patient treatment: epirubicin, oxaliplatin, and 5-fluorouracil. Organoid response to chemotherapeutic treatment was correlated with the tumor response in each pat…

[Organoid nevus (nevus sebaceus) dominated by the apocrine glands].

We report a case of congenital organoid nevus with an excessive number of apocrine glands. A one-year-old boy presented with a slowly enlarging keratotic element in the left temporal region, and biopsy showed hyperkeratosis, acanthosis, follicular plugging and multiple transverse sections of mature apocrine glands filling the reticular dermis. The lesion was benign and treatment was therefore conservative. In rare cases, various types of adnexal neoplasms may develop, but malignant tumours occur very seldom.

Guiding Principles: Reporting Elements for Gastrointestinal Organoid Research.

Gastrointestinal (GI) organoids are now widely used for disease modeling, drug discovery, regenerative-, and precision medicine. As publications using GI organoids have increased exponentially, methodological reporting across GI organoid studies remains inconsistent, making results difficult to interpret, compare, and reproduce. To address this, we present community-informed guidance for reporting key experimental details in GI organoid research. Our recommendations were developed by integrating established biomedical reporting frameworks, organoid-focused resources from international organiza…

[Current progress in brain organoid research for human biology].

Understanding human brain development, function, disease, and drug responses remains a central challenge in neuroscience and medicine. However, detailed analyses of living human brain tissue are technically and ethically difficult, and conventional studies have therefore relied largely on animal models and two-dimensional culture systems. Nevertheless, substantial differences exist between humans and animals in developmental processes, cellular composition, neural circuit, and pharmacological responses. Brain organoids are three-dimensional neural tissues generated from human embryonic stem ce…

A Protocol for the In Vitro Culturing of Vascularized Pancreatic Islet Organoids.

This study presents a protocol for co-culturing pancreatic islet cell clusters derived from pancreatic tissue with human umbilical vein endothelial cells (HUVECs) on Matrigel using a specialized culture medium to form vascularized pancreatic islet organoids. We established a novel culture system for vascularized pancreatic islet organoids and compared the survival and insulin secretion capabilities of pancreatic islet cells in the presence and absence of glucose stimulation. Our results indicate that matrix adhesive materials can effectively facilitate the self-assembly of the vascularized end…

Laboratory-Engineered Glioblastoma Organoid Culture and Drug Screening.

Glioblastoma (GBM) is described as a group of highly malignant primary brain tumors and stands as one of the most lethal malignancies. The genetic and cellular characteristics of GBM have been a focal point of ongoing research, revealing that it is a group of heterogeneous diseases with variations in RNA expression, DNA methylation, or cellular composition. Despite the wealth of molecular data available, the lack of transferable pre-clinic models has limited the application of this information to disease classification rather than treatment stratification. Transferring the patients' genetic in
