We uncovered a previously unrecognized role of asparagine synthetase 1 (ASN1) in regulating rice root elongation. The plant root system is crucial for water and nutrient acquisition. Amino acids play pivotal roles in regulating root development. In this study, we uncovered a role of asparagine synthetase 1 (ASN1) in regulating rice root development. Histological analysis of asn1 mutants showed a shortened root length resulting from reduced cell proliferation rates in the root apical meristem. Metabolite profiling indicated that jasmonic acid (JA) metabolism exhibited the most significant alter

Stilbene trans-resveratrol is a key specialized metabolite in grapevine (Vitis vinifera), known for its notable antioxidant, anti-inflammatory, and anticancer properties. Its biosynthesis is primarily induced under stress conditions and can be enhanced in grapevine cell cultures using elicitors such as cyclodextrins and methyl jasmonate. However, the involvement of reactive species in this process remains poorly understood. In this study, we investigated the roles of hydrogen peroxide, nitric oxide, peroxynitrite, and hydrogen sulfide in trans-resveratrol production in grapevine suspension-cul

Plants emit volatile compounds that orchestrate complex ecological interactions, with methylated catabolites of interaction-induced phytohormones being common examples. Salicylic acid (SA) mediates plant antipathogen responses, while its methylated derivative, MeSA, broadly mediates plant-insect interactions without specificity. Here, we identified dimethyl salicylate (DMSA), an unappreciated dimethylated SA catabolite, emitted by rice when attacked by the major destructive pest, the brown planthopper. DMSA biosynthesis requires an O-methyltransferase cascade, BSMT1 (benzoic acid/salicylic aci

Strawberry (Fragaria × ananassa) is highly susceptible to gray mold caused by Botrytis cinerea. WRKY transcription factors and jasmonic acid (JA) are central to defense against necrotrophs, yet the regulatory interaction in strawberry remains unclear. Here, we characterized FaWRKY21, a terpinen-4-ol-responsive, Group IIc WRKY gene encoding a nuclear-localized protein. Transient overexpression in strawberry fruit enhanced defense against B. cinerea and increased endogenous JA concentrations. This enhanced defense was confirmed in transgenic Arabidopsis plants overexpressing FaWRKY21, where AtPD

Potato powdery scab, caused by the soilborne pathogen Spongospora subterranea f. sp. subterranea (Sss), poses a significant threat to potato production, reducing potato value and impacting fresh market quality. Effective management strategies for this disease are currently lacking, and Sss is widespread in many potato-growing regions, highlighting the urgent need for effective control measures. Although the use of disease-resistant cultivars holds potential as a sustainable solution, the genetic mechanisms underlying resistance to Sss remain unclear. In this study, we investigated the role of

Rice (Oryza sativa L.), the staple food for half of the world's population, suffers heavy damage by insect herbivores, especially the emerging fall armyworm (FAW), Spodoptera frugiperda. HIGHER YIELD RICE (HYR), a master regulator of multiple biological pathways with an established gene regulatory network, has been found to improve rice yield to ∼29% and tolerance to environmental stress. However, its impact on defense has not been explored. We hypothesized that, FAW would target HYR plants because of its vigorous growth and lead to trade-offs for defense. Pecan leaf-variegated plant, which was infected with a novel badnavirus named pecan mosaic virus (PMV) detected by small RNA deep sequencing, is a vital model plant for studying the molecular mechanism of retaining green or chlorosis of virus-infected leaves. In this report, PMV infection in pecan leaves induced PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI). PMV infection suppressed the expressions of key genes of fatty acid, oleic acid (C18:1), and very-long-chain fatty acids (VLCFA) biosynthesis, indicating that fatty acids-derived signaling was one of the important def

Adults of many mosquito species feed on plants to obtain metabolic energy and to enhance reproduction. Mosquitoes primarily rely on olfaction to locate plants and are known to respond to a range of plant volatiles. We studied the olfactory response of the yellow fever mosquito Aedes aegypti to methyl jasmonate (MeJA) and cis-jasmone (CiJA), volatile compounds originating from the octadecanoid signaling pathway that plays a key role in plant defense against herbivores. Specifically, we investigated how Ae. aegypti of different ages responded to elevated levels of CiJA in two attractive odor con

Although the anticancer properties of extra virgin olive oil (EVOO) extracts have been recognized, the role of single compounds in non-melanoma skin cancer is still unknown. The in vitro chemopreventive and anticancer action of EVOO extracts and oil-derived compounds in non-melanoma skin cancer models were evaluated on cutaneous squamous cell carcinoma cells and on immortalized human keratinocytes stimulated with epidermal growth factor. Preparation of EVOO extracts and isolation of single compounds was carried out by chromatographic methods. Antitumor activity was assessed by cell-based assay

BACKGROUND: Bois noir is an important disease of grapevine (Vitis vinifera L.), caused by phytoplasmas. An interesting, yet elusive aspect of the bois noir disease is "recovery", i.e., the spontaneous and unpredictable remission of symptoms and damage. Because conventional pest management is ineffective against bois noir, deciphering the molecular bases of recovery is beneficial. The present study aimed to understand whether salicylate- and jasmonate-defence pathways might have a role in the recovery from the bois noir disease of grapevine.
RESULTS: Leaves from healthy, bois noir-diseased and

BACKGROUND: Brachypodium distachyon is a promising model plants for grasses. Infections of Brachypodium by various pathogens that severely impair crop production have been reported, and the species accordingly provides an alternative platform for investigating molecular mechanisms of pathogen virulence and plant disease resistance. To date, we have a broad picture of plant immunity only in Arabidopsis and rice; therefore, Brachypodium may constitute a counterpart that displays the commonality and uniqueness of defence systems among plant species. Phytohormones play key roles in plant biotic st

Heavy metal (HM)-accumulating herbaceous and woody plants are employed for phytoremediation. To develop improved strategies for enhancing phytoremediation efficiency, knowledge of the microstructural, physiological and molecular responses underlying HM-accumulation is required. Here we review the progress in understanding the structural, physiological and molecular mechanisms underlying HM uptake, transport, sequestration and detoxification, as well as the regulation of these processes by signal transduction in response to HM exposure. The significance of genetic engineering for enhancing phyt

Cross-talk between hormones is required for plant response to developmental cues and environmental stresses. This cross-talk is achieved through several regulators located in convergence point of distinct hormonal signaling. In plant defense responses, salicylic acid and jasmonic acid affect each other in antagonistic manner. In a recent study we showed that AtMYB44 transcription factor positively regulates SA-mediated defense expression and enhanced resistance to Pst DC3000. On the other hand, AtMYB44 negatively regulates expression of JA-mediated defense gene expression and downregulated res

Concentrations of carbon dioxide (CO(2)) are increasing in the atmosphere, affecting soybean (Glycine max L.) phytohormone signaling and herbivore resistance. Whether the impact of elevated CO(2) on phytohormones and induced defenses is a generalized response within this species is an open question. We examined jasmonic acid (JA) and salicylic acid (SA) under ambient and elevated CO(2) concentrations with and without Japanese beetle (Popillia japonica Newman) damage and artificial damage across six soybean cultivars (HS93-4118, Pana, IA 3010, Loda, LN97-15076, and Dwight). The effect of previous infestation (preconditioning) by the whitefly Bemisia tabaci (Gennadius) biotype B on the population fitness of subsequent infestations that fed on three isogenic tomato genotypes (wild-type [Wt], a jasmonic acid [JA] defense-enhanced genotype [35S], and a JA-deficient genotype [spr2]) was examined. We tested the hypotheses that whiteflies fed on preconditioned tomatoes (Solanum lycopersicum L.) would have reduced fitness and that the effect would be mediated via the JA-dependent systemic plant defense pathway. Preconditioning by the whitefly resulted in decreased levels

Fusarium graminearum is the principal causative agent of Fusarium head blight (FHB), a devastating disease of wheat and barley. This fungus can also colonize Arabidopsis thaliana. Disease resistance was enhanced in transgenic wheat and Arabidopsis plants that constitutively overexpress the NONEXPRESSOR OF PATHOGENESIS-RELATED GENES 1 (NPR1) gene, which regulates salicylic acid (SA) signaling and modulates the activation of jasmonic acid (JA)-dependent defenses. Here, we provide several lines of evidence that reveal an important role for SA and JA signaling in Arabidopsis defense against F. gra
