Blockade of dopamine-induced chemosensory inhibition by domperidone.

The sensory discharges from the carotid body chemoreceptors of the cat are transiently inhibited by dopamine (DA) injections. This chemosensory inhibition was effectively blocked by domperidone, a selective antagonist of D2 dopaminoceptors. The basal frequency of spontaneous chemosensory impulses was immediately and sustainly increased after domperidone, suggesting the withdrawal of a tonic inhibition of chemosensory discharges by endogenous DA released from glomus cells. The peripheral dopaminergic modulation of chemoreflexes may be separately blocked by domperidone, a drug unable to cross th…

Effects of dopamine and bromocriptine on colonic motility in dog.

The effects of intravenous infusions of dopamine (0.1 to 1 mg kg-1h-1) and bromocriptine (10 to 40 micrograms kg-1h-1) on colonic motility were investigated in fasted dogs fitted with permanent strain gauges on the ascending, transverse and descending colon. Infused at rates of 0.5 and 1 mg kg-1h-1 during 1 h, dopamine immediately stimulated the motility of the descending colon; after a delay of 40 to 60 min this effect was balanced by an inhibition of the motility of the ascending and transverse colon. Bromocriptine infused intravenously at doses of 10 to 40 micrograms kg-1h-1 stimulated the …

Insulin suppressive effects of aminotetralin analogs and of dopamine.

The study concerned the effects of 2-amino-6,7-(OH)2-1,2,3, 4-tetrahydronaphthalene (A-6,7-DTN), dipropyl-A-6,7-DTN and dopamine on plasma levels of immunoreactive insulin (IRI) in cattle. During infusions of A-6,7-DTN, dipropyl-A-6,7-DTN and dopamine, the circulating levels of IRI decreased. In response to A-6,7-DTN, the levels of glucose increased whereas non-esterified fatty acids did not change. The decrease of IRI during additional infusions of A-6,7-DTN was not modified by prolonged administration of A-6,7-DTN. During infusions of A-6,7-DTN there was no increase in response to the admini…

Study on the striatal dopamine availability in Parkinson's disease.

Striatal dopamine availability in connection with the plasma levodopa levels was studied in 12 parkinsonian patients treated with optimal doses of levodopa. Three patients showed the wearing-off phenomenon and the 9 others showed no daily fluctuations in disability. All of the 3 patients with the wearing-off phenomenon showed an up-and-down pattern of dopamine availability index. Out of the 9 patients presenting no fluctuations in disability, 6 showed an ascending pattern of the dopamine availability index and 3 an up-and-down pattern. Of the former 6 patients, 2 received short-term (less than…

On the prolactin-inhibiting effect of neurotensin. The role of dopamine.

Neurotensin (NT) when injected in a dose of 5 micrograms into the third cerebral ventricle of conscious, unrestrained male rats, outfitted with chronic third ventricle and jugular cannulae, decreased the resting prolactin (PRL) levels. NT (5 micrograms) also markedly inhibited the PRL-releasing effect of activation of central serotonin receptors by 5-hydroxytryptophan (5HTP, 15 mg/kg) in animals pretreated with fluoxetine (4 mg/kg). however, interruption of dopamine (DA) neurotransmission by either alpha-methyl-p-tyrosine (alpha MT, 250 mg/kg) or spiroperidol (0.3 mg/kg) led to a blockade of t…

Dopamine does not affect parathyroid function in man.

There is conflicting evidence for a role of dopamine (DA) in modulating parathyroid gland function. DA stimulates parathormone (PTH) secretion in cattle in vivo and in vitro, but is without effect on dispersed human parathyroid cells from adenomatous and hyperplastic tissue. Therefore, we studied the PTH response to a 30-min infusion of DA (4 micrograms/kg . min) in five normal male volunteers. There was no change in the levels of PTH, as measured by RIA, although this infusion rate was effective in lowering PRL levels. During the infusion period, total calcium levels remained unchanged. We co…

The hemodynamic relevance of free and conjugated dopamine in pheochromocytoma.

Two patients with adrenomedullary hypersecretion (confirmed pheochromocytoma and adrenomedullary hyperplasia) presented 15 spontaneous crises associated with hypertension or hypotension with or without tachycardia. Correlation coefficients calculated between extreme values of pulse rates and of systolic and diastolic blood pressures on the one hand and plasma free and conjugated norepinephrine, epinephrine, and dopamine (DA) sampled at the height of the crises on the other, showed no relationships between free or conjugated norepinephrine or epinephrine and blood pressure or pulse rate. Howeve…

[Dopamine as a regulator of natriuresis. Experimental bases and their implications].

Renal effects of dopamine (DA) infused i.v. in a subpressor dose (0.1 microgram. kg.-1 min-1) were investigated in healthy human subjects during steady hypotonic polyuria. In each experiment four clearance (cl.) periods of 15 min were performed; DA was administered during the second and the third cl. periods. The glomerular filtration rate and renal effective plasma flow were estimated as endogenous creatinine and PAH clearances. Moreover the following variables were evaluated: a) sodium total and sodium isosmotic reabsorption as a % of sodium filtered load (s.f.l.); b) sodium anisosmotic reab…

Motoneurons which may utilize dopamine as their neurotransmitter.

1. Three neurons in the abdominal ganglion ofAplysia elicit gill movements similar to those observed when dopamine is added to a gill perfusate. One, which we designate L9, produces contractions of all medial and lateral external pinnule muscles, the circular and longitudinal muscles of the afferent vessel, and the circular muscles of the efferent vessel. The others produce identical contractions in anterior portions (L9-a) or posterior (L9-p) portions of the gill. These contractions are similar to, but less pronounced than, those produced by the motoneuron, L7. 2. Clear population excitatory …

Dopamine-induced paroxysmal supraventricular tachycardia in an infant.

Dopamine is an effective inotropic agent in the management of circulatory collapse. Ventricular dysrhythmias are reported to occur during dopamine infusion. The following report describes an infant with circulatory failure who experienced an episode of paroxysmal supraventricular tachycardia while receiving dopamine. Instability or immaturity of the cardiac conduction system during early infancy may contribute to the induction of an arrhythmia of supraventricular origin.

Elevation of brain neuroleptic/dopamine receptors in schizophrenia.

To test the dopamine hypothesis of schizophrenia the authors measured specific 3H-neuroleptic/dopamine binding sites in three dopamin-rich regions of 59 postmortem normal human brains and 50 postmortem brains from schizophrenic patients using 3H-haloperidol and 3H-spiperone. The binding of 2 nM 3H-haloperidol and of 1 nM 3H-spiperone was significantly elevated in the brains from schizophrenic patients. The brain regions from patients who had no history of being treated with neuroleptic drugs also exhibited significantly higher binding of the 3H-neuroleptics. These results are compatible with t…

Effect of dopamine on hypotension induced by spinal anesthesia.

Chronically instrumented, near-term pregnant sheep were subjected to autonomic blockade with spinal anesthesia. Systemic arterial pressure, heart rate, and uterine blood flow decreased and uterine vascular resistance increased during the spinal blockade. Infusion of dopamine during the spinal hypotension corrected the disturbed circulatory parameters. These data provide evidence that: (1) the peripheral circulation of the near-term pregnant sheep is as sensitive as that of pregnant women to autonomic blockade, (2) the sensitivity is probably related to factors other than the changes in venous …

Changes in cardiac rhythm in children treated with dopamine.

We reviewed the charts of 31 children receiving dopamine to document possible changes in cardiac rhythm associated with dopamine infusion. Six of these developed a dysrhythmia while on dopamine. All six were receiving dosages greater than 10 microgram/kg/min; in five of the six patients the infusion rate varied from 10-20 microgram/kg/min. Of the remaining 25 patients, 14 had preexisting dysrhythmias of nonventricular origin. Two of these 14 patients converted to sinus rhythm while on dopamine. Eleven patients who were in preexisting sinus rhythm had no rhythm changes on dopamine. Based on the…

Effect of dopamine on the esophageal smooth muscle in vivo.

Intravenous administration of dopamine caused a dose-dependent reduction in the lower esophageal sphincter pressure and contractions in the lowermost part of the body of the esophagus in the opossum. A dose of 5 mug per kg produced maximal response. The onset of contractions in the body occurred around 20 sec after the onset of the sphincter response. The effect of dopamine was different from isoproterenol, which caused no contractions in the body and caused reduction in sphincter pressure, and from phenylephrine, which also caused no contractions in the body but caused contraction of the sphi…

Dopamine receptor blockade and the neuroleptics, a crystallographic study.

The X-ray structures of 12 drugs of the tricyclic class having varying pharmacological profiles have been examined in detail in an attempt to rationalize the known structure-activity relations of neuroleptic drugs with respect to their ability to block dopamine receptors in the brain. Further evidence is presented in support of the theory that the neuroleptics are able to block dopamine receptors because of a conformational complementarity between certain portions of these drugs and dopamine.

Responses of saphenous and mesenteric veins to administration of dopamine.

Others have observed that dopamine (3,4-dihydroxyphenylethylamine) constricts resistance vessels in skin, but dilates these vessels in the mesentery. We studied the effects of dopamine on cutaneous and mesenteric veins of dogs to see if this agent also produced qualitatively different effects on the tone of capacitance vessels (veins) in these vascular beds. The lateral saphenous or the left colic vein was perfused at constant flow with blood from a femoral artery. Pressures at the tip of the perfusion cannula and at the tip of a catheter 15 cm downstream were recorded continuously. Increases …
